Determination of pepsinogens using IgY and IgG

A decreased concentration of pepsinogen A in serum was found to be a marker of gastric cancer, similarly as a low ratio of pepsinogen A to pepsinogen C. In the present study we have compared properties of immunoglobulin fraction isolated from the egg yolks after immunization of laying hens with pepsinogen isolated from porcine gastric mucosa with those of present in rabbit antiserum obtained after the animal immunization with the same antigen. The characteristics of chicken antibodies against porcine pepsinogen and the comparison with rabbit antibodies raised against the same antigen was carried out using the following methods: ELISA, affinity chromatography on immobilzed antigen and bovine serum albumin, SDS and native electrophoresis and MALDI-TOF MS/MS. While the rabbit specific antibodies interacted with the used antigen and only slightly with bovine serum albumin and there was a diference between pre-immune IgG and specific IgG, in the case of chicken antibodies IgY it did not work. No diference was observed between ELISA tests performed with pre-immune serum and the serum after immunization with porcine pepsinogen and a high interaction of IgY with bovine serum albumin in pre-immune serum and specific IgY after the immunization were detected. Cycloaddition of 3-(deoxyribos-1-yl)propynoate with alkynes

The object of this work is development of method for a syntesis of Dewar benzenes bearing the deoxyriboside group. The synthesis was based on ethynylation of halogenose resulting in the formation of a mixture of anomeric ethynyldeoxiriboses, which was followed by silylation and separation of individiual anomers. After removal of the silyl group the isomerically pure ethynyldeoxyriboses were transformed into corresponding propynoates that were the key substance for the formation od Dewar benzenes. The reaction of the propynoates with the tetramethylcyclobutadiene-aluminium trichloride complex provided the desired Dewar benzenes bearing the deoxyribose group. Finally, rearrangement of the formed Dewar benzenes to 1-aryldeoxyriboses was studied under thermal and photochemical conditions. Keywords: cycloaddition, arenes, Dewar benzene
Analysis of proteins and their changes in biological tissues

(EN) Proteins are the important biologically active substances for the organism, their qualitative and quantitative composition determines thein function in the organism. Protein analysis in biological material is an important part of biological and medical research. This dissertation is focused on the optimalization of sample preparation - the biological materials - namely the pre-analytical phase of sample processing and their preparation for their own analases and also introducing new procedures in theie praparation. The conditions of individual analysis approaches were always optimized by means of one- dimensional and two-dimensional gel electrophoresis and mass spectrometry. The obtained results were statistically evaluated and other possible approaches to futher research were outlined. The main biological material studied was human saliva with regard to oral health. Comparative study of human saliva was performed (human saliva obtained from caries-free and caries-susceptible people). The saliva composition of individual fractions was investigated with regard to oral health. The study was carried out to compare differences in the abundances of proteins in the saliva of caries-free and caries-susceptible persons, based on label-free mass spectrometry quantification, and to perform a gender...

Gamification in the quantitative online research

The thesis concentrates on the application of gamificaton principle in the quantitative online research. The main goal of the thesis is to evaluate the effects of gamification on answers provided by respondents and their overal perception of the questionnaire. The hypotheses, based on the research of theoretical and methodological literature and metaanalysis of already conducted case studies, refers to the most discussed topics concerning gamification: survey evaluation, evaluation of respondent's behavior and the impact on the survey data. The results are validated using the split- ballot method on a carried out quantitative research. As a research tool serve a standard questionnaire and a questionnaire with small gamification adjustments. The results favor the version of questionnaire with gamification in the area of overall perception of the questionnaire and increased time spent on the research by the respondents. However, the results don't show any measurable impact on the respondent's behavior or survey data.

Law and needs of adoptive parents and problemtic areas in the process of adoption

Bachelor thesis deals with the adoption system issue in Czech Republic. It focuses primarily on the rights and needs of adopters in the adoption process and problematic parts of the adoption system. The thesis is divided into three parts. The theoretical part defines the basic terms and legal regulations of adoption and chronologically describes the process of adoption from the decision making for adoption through the submission of application form, expert assessment of applicants, completion of preparation and psychological testing to the period of waiting for the child and the period after adoption. In the methodological part is defined the conducted qualitative research, in which were interviewed various participants of the adoption process: social workers, NGO workers and adoptive parents. Results of the research are then interpreted in the last, analytical part. There were defined many problematical parts and they permeate all phases of adoption process. As significant problems can be considered lack of clarity in the motivation of applicants while entering into the adoption process, lack of credibility of social workers caused by the negative media image, the trend of increasing age of the applicants, inconsistent methodic and quality level of mandatory preparation, lack of finance and...
