Impact of PHA accumulation on stress resistance of bacteria against disinfection agents

This bachelor thesis deals with the influence of acumulation of PHA balancing on the resistence of bacteria against desinfecting means. Two bacterial strains, PHA accumulation Cupriavidus necator H16 and its mutant strain insoluble PHA synthase Cupriavidus necator PHB-4 were used as model microorganisms. These bacteria were exposed to three different concentration of three desinfecting means (etanol, savo and ajatin). The viability of bacteria by the action of disinfectants was determined by the plate method and by the flow cytometry. The influence of desinfecting means against bacteria was examined by the broth dilution method which rested in measurement of absorbance over time. Agar diffusion method was the last one technique use. The results show that PHA production positively aids cell defence against desinfectants because bacterial strain Cupriavidus necator H16 is more resistant than its mutant strain Cupriavidus necator PHB-4. Of the free selected disinfectants, ajatin is the most effective.
Isolation, characterization and application of biomedically important polymer P(3HB-co-4HB)

This diploma thesis deals with the isolation and characterization of copolymer P(3HB-co-4HB). The teoretical part was prepared as a literature search which describe polyhydroxyalkanoates in general, their structure, synthesis, degradation and isolation. Furthermore, copolymers containing 4HB units are mentioned in this thesis and there is also no mentioned of the biosynthesis and biodegradation of copolymer P(3HB-co-4HB). The first aim of this diploma thesis was to find out which solvent is the best for copolymer extraction and then characterize obtained copolymer P(3HB-co-4HB). The isolated copolymer was characterized by gas chromatography, SEC-MALS, thermal analysis and SEM. In the second part of the thesis, release of model biologically active substance from the PHA films was studied. Rhodamine 6G dye was selected for the simulation, which was used to stain the copolymer films and the P(3HB) films. It was observed that film prepared from P(3HB-co-4HB) copolymer released entrapped substance much faster than film made from P3HB homopolymer, and, in addition, the copolymer was substantially more susceptible to enzyme degradation.
War on Drugs during Reagan administration

This paper focuses on the topic of the "War on Drugs" during Reagan administration. Its main objective is to compare goals set by the Reagan administration in the "War on Drugs" in the 1980s with their consequences and analyze the means that were used to fulfill these goals, which were set for domestic "War on Drugs" and introduce its "side effects" in the socio-economic sphere as well as to determine, whether goals set by Reagan administration were fullfilled or not. By comparing the goals set by Reagan's administration in 1986 and the analyzed results of Reagan's "War on Drugs" in late 1980s I determined the success of Reagan's "War on Drugs".
