Y Soft Campus - the semi-abandoned shooting range area in Brno-Pisarky

I have designed the campus and the headquarters of the company Y-Soft. It is situated in area Brno-Pisárky. The whole site is 104 900 m large. We have done a research of the needs of employees and management. I have integrated he results into my project. The designed campus includes Y-Soft’s office buildings, a sport gymnasium, a fitness center, a park, residential buildings and kindergarten. The headquarters of Y-Soft is designed as a compact building for easier communication between departments. I tried to create large building with the human scale and nice spaces to work and relax. I have chosen a skeleton construction made from monolithic concrete. The reinforced concrete frame will be insulated by mineral wool. The facade will be created from the part of windows with colored frame and part covered by Thermowood.

The WOOX company is focused on the production of sport goods - mainly clothes. It began to grow and filled up the capacity of its first factory. For the new one, it has chosen the brownfield of the former Jeseník’s textile factory. It is located five minutes from the city center. I approached the owner of this company and because he was just working on the reconstruction, we agreed on cooperation. My intention is to provide the conceptual idea and possible solutions. It should be used as a source of inspiration. Based on my analysis of the plot and the surroundings, my thesis creates a production hall, a design office, dwellings and a multifunctional hall with a bistro. The work is based on SMART principles. It is trying to interconnect the interests of Jeseník’s residents and the company WOOX.
Bioanalytical method development for the investigation of the role of pyridoxal isonicotinoyl hydrazone in isonazid intoxication

High-performance liquid chromatography is one of the most frequently used separation methods for an analysis of drugs in biological material. It is suitable both for quantitative and qualitative drug analysis. Isoniazid is used for the treatment and a prevention of tuberculosis. Acute isoniazid intoxication is manifested by tonic-clonic convulsions. Vitamine B6 (pyridoxine) is used in clinical practice as an antidotum. Although this type of toxicity is considered to be associated with the deficiency of vitamine B6 (or its active form, pyridoxal-5-phosphate - PLP), description of mechanisms and processes, which are responsible for this effect, is still missing. The aim of this study was to develop suitable chromatographic conditions for monitoring intoxication by isoniazid and detection of potential hydrazone metabolites. These conditions should allow a separation of seven analytes (isoniazid, acetylisoniazid, pyridoxal, pyridoxol, pyridoxal-5-phosphate, and the o-108 - internal standard). Furthermore, the aim was to study possibilities of isolating all those substances from rabbit plasma. The diploma thesis is targeted to usage of High Performance Liquid Chromatography in the field of Quality control of drug substances. Methylergometrine is a derivative of ergometrine that belongs to Ergot alkaloids. These native origin substances have strong pharmacological effects on different human body systems already in low concentrations. Active components from ergot are acquired by extraction and subsequent separation of individual components from a mixture. These substances are repurified. It is very expensive to produce totally pure substances and thus final substances may contain defined amount of impurities. In case of methylergometrine Related substances are analysed according to the valid European pharmacopoeia. Nine substances are in the specification of possible analysed components, tenth component (-)methylergometrinine has not been described in the pharmacopoeia so far. Title of diploma thesis: Study of chiral aspects of drug metabolism using instrumental analytic methods For the evaluation of stereospecificity of rat and human cytosolic carbonyl reductases and activity of microsomal oxidases involved in nabumetone biotransformation, a new chiral high-performance liquid chromatographic method was developed. The prepared LLE-HPLC-PDA method enabled extraction, separation and ultraviolet detection of prochiral nabumetone and its five phase I metabolites including enantiomers of two chiral biotransformation products of carbonyl reduction. Methyl ester of naproxen served as an internal standard. Diethyl ether was used for liquid-liquid extraction (LLE) of biomatrices. Chiralcel OD-R 250 mm×4.6 mm column with a mobile phase methanol-1M NaClO4/HClO4 aqueous solution pH=3 (75:25, v/v) were employed in isocratic sufficient separation of nine analytes. Influence of mobile phase strength, mobile phase pH and buffer strength on the separation of biologically important polar compounds: adenosine monophosphate (AMP), adenosine diphosphate (ADP), adenosine triphosphate (ATP), cyclic adenosine monophosphate (cAMP), creatine (C) and phosphocreatine (CP) was tested. Analysis of selected polar compounds in HILIC mode

Nowadays High Pressure Liquid Chromatography is the most important and the most frequently used separation technique that enables qualitative and quantitative analysis of the complex mixtures. One of its variants is Hydrophilic Interaction Liquid Chromatography. Lately this technique has experienced an increasing boom particularly, due to its ability to improve separation of very polar and hydrophilic compounds. The adeninribonucleotides (ATP, ADP, AMP, cAMP), creatine and phosphoceratine are among these highly polar compounds that are of great importance not only for human body. Number of analytical methods has been developed and published for the analysis of these compounds; however HPLC methods using the reverse phases have been dominated. The basic overview of these methods is a part of this diploma thesis. The experimental part of this thesis is focused on the development of chromatographic conditions suitable for the simultaneous analysis of ATP, ADP, AMP, cAMP, creatine and phosphocreatine in HILIC mode. Employment of HPLC in the field of Drug analysis I.

Curcumin with it's derivates belongs to pharmacologically acting diarylheptanoids. They are obtained from the herbal rhizomes of genus Curcuma spp. (Zingiberaceae), which are widespread in tropical regions of south-east Asia. For last decades the drug has been in focus of scientists. On the human organism there have been proven many interesting effects. Therapeutically are used it's choleretic, antioxidant and antiinflammatory activities. Nowadays the interest orientates to the prevention and treatment of cancer and to the favourable influence on Alzheimer's disease. After oral administration, however, curcumin achieves the system circulation only in traces. In introduced study we developed HPLC method for the determination of curcuminoids. The most advantageous way is extracting the pigments from the herb or extract with methanol as relatively polar solvent. In the aqueous medium is practically unsoluble. The presence of three acid hydrogens exhibits in the alkaline aqueous solutions. The resultating ionisation is documented by color change from yellow to red. These conditions prejudice the stability of the substance - in part we may prevent the degradation by using the antioxidants and keeping in dark place.
