This thesis investigates the influence of habitat, dispersal abilities and evolution on distribution of mycoheterotrophic plants represented by six orchid species. Several methods including seed trapping, in situ seed germination, fungal barcoding, gene flow study and stable isotopes analyses were used to examine this topic. The final synthesis of these approaches shows lower limitation by dispersal abilities and stronger limitation by habitat.
Genetic diversity and structure of populations of Campanula glomerata on regional scale in White Carpathians

Habitat fragmentation can adversely affect species populations due to reduced genetic diversity and increased population differentiation due to habitat isolation, reduced population size, and disruption of gene flow. Many species suffer from habitat fragmentation and isolation, and Campanula glomerata is a good example of a species that is noticeably declining in many places due to changes in landscape management. The question is how the model species responds to these changes and whether is the change are reflected in genetic diversity and population structure. The aim of this work is to determine the degree of genetic diversity and to reveal the genetic structure of populations of the model species C. glomerata on a small regional scale in the White Carpathians. This system will allow us to compare populations from two different regions that have been affected to varying degrees by agricultural intensification and fragmentation in the last century. It is a southwestern and northeastern part of the region, which differs from each other in the history of management in the landscape, but also in the continuity of meadows, species composition and environmental conditions. Comparison of three trials used for assassement of ability to disperse by wind and its importance for colonazition of abandoned fields in Asteraceae species

Several methods are used for studying seed dispersal (seed traps, tracking individual seeds, tracking seeds coloured by fluorescent colours, etc.) However, only a few studies compared results obtained by several methods. In first part of this master thesis, I compared the three above mentioned methods used for studying seed dispersal using species from Asteraceae family. From previous research within the study area (Úštěcko), it is known that dry grassland species are able to colonize abandoned fields. Using seed dispersal data obtained within this theses, I tried to assess the role of seed dispersal on distribution of dry grassland species on abandoned fields both on local and regional scale. The results showed that dispersal curves obtained by the three methods differ significantly. This results may be due to different wind conditions during the experiments. Seed trap data show results from long-term seed dispersal influenced by highly variable wind conditions. In contrary, seed release experiments showed results based on single dispersal event under limited wind conditions. Germination course and mycorrhizal association of two \kur{Neottia} species

Germination course and mycorrhizal partners during germination and adulthood were studied in Neottia ovata and N. cordata. Neottia ovata was studied in two different biotops: meadows and forests. Research was conducted in in situ conditions in the Šumava and the Smrčiny mountains.

The first part of this thesis is a review on limitations of orchids by their myccorhizal symbiosis. The second part presents on outline of my master thesis, which is focused on ecology and myccorhizal partners of two Listera species.
