Preconcentration of ion compounds from sample by epitachophoresis

Epitachophoresis is newly developed method, which allows to separate and concentrate analytes from large volumes of complex samples. Principle is based on isotachophoresis and utilize discontinuous electrolyte system. Different design of device is used, its round shaped and due to that acceleration of analytes zone migration occurs during separation towards centre of device and at the same time widening of zones happens. Focusation, concentration and separation of analytes between leading and terminating electrolyte progress from outer line of device towards its centre. It is necessary to stabilize this boundary to prevent mixing of leading and terminating electrolyte. Main target of this diploma thesis is optimization of epitachophoretic conditions, mainly electrolyte boundary stabilization options. Tested stabilizing media were agarose gels, foamed polymer disks and PDMS columns. All leading and terminating electrolyte boundary stabilizing media undergo several optimizations so the best option can be selected for concreate applications. DNA was used as a model analyte and the media with the highest recovery were selected. The best results offered NEEO and PFEG agarose gels and Dyneema foamed polymer with recovery over 70%. Another interesting alternative are PDMS columns, with their open structure that allow concentration of large volume analytes such as microorganisms.
Testing of food authenticity using DNA analysis

Authenticity verification of food products is an important step before their release to the market. This testing ensures that the product contains what the label states and also confirms the medical harmlessness. One of methods possible to use for authenticity verification is polymerase chain reaction (PCR). The experimental part was focused on the DNA isolation using magnetic particles on PCR – ready quality from food product – ketchup. DNA isolated from seeds and stalk of tomato were used as a controls. Amplificalibity of DNA isolated with magnetic particles using magnetic separator and needle was compared. The method requires further optimization.

1 Blockchain and copyright Abstract The aim and objective of this Thesis is to introduce the phenomenon of blockchain technology and some of its applications in the field of copyright. The thesis focuses primarily on the areas of databases, NFT and Smart Contracts, and their application in the existing, particularly Czech law. In its first chapter the author defines the blockchain technology, describing the basic terminology, such as node, block, hash, transaction, peer-to-peer network, proof of work. The chapter then explains how blockchain works, describing its key characteristics. The author dwells on three types of blockchain and finally discusses the issue of ethics and privacy protection. The next chapter on copyright summarizes basics of this area of law which has been experiencing fast development of new technologies. A sub-chapter on author's property rights, whereby special attention is paid to the issue of digital reproductions, constitutes an important part the the Thesis. Central chapters deal with Smart Contracts, blockchain, collective rights management and NFT (Non-fungible Tokens).
