Characterization of mice with constitutively active Wnt/beta-catenin signaling pathway in lens

Lens development and differentiation are strictly regulated processes. Various disturbances of these processes can lead to vision-limiting pathologies. The vertebrate lens is composed of epithelial cells and terminally differentiated fiber cells. Differentiation of fiber cells is connected with expression of fiber cell specific proteins (such as crystallins), cell cycle exit, and finally with the degradation of cell nucleus and organelles. Wnt/β-catenin signaling plays important roles during early eye development as well as later during the lens differentiation. To investigate the consequences of constitutive activation of Wnt/β-catenin signaling in lens fiber cells transgenic mouse strain, called CLEF, was created. Constitutive activation of Wnt/β-catenin signaling in fiber cells of CLEF mouse is achieved by transgenic protein CLEF that contains C-terminal activation domain of β-catenin fused to the amino terminus of full-length protein Lef1. The expression of CLEF transgene is under the control of αA-crystallin promoter. As a result of constitutive activation of Wnt/β-catenin signaling in fiber cells, adult CLEF mice develop cataracts and microphthalmia, and the morphology of adult mutant lenses is disrupted. Legal issues of Surrogacy

This master thesis deals with the issue of legislation of surrogacy. Development in the area of reproductive medicine as the first "test tube baby" in 1978 and following medical progress has inevitably brought development expansion of this institute. This institute, although that it brings with itself many ethical, social and legal questions, is not legally regulated in the Czech Republic (except one provision, Art. 804 of the Civil Code). Therefore, this master thesis tries to provide a proposal of legislation which might be adopted in the Czech Republic, using primarily analytical and comparative methods. After the surrogacy is explained and forms and models of it are described, the international framework is clarified. Although many states realize actuality of this topic, there has not been any international legislation adopted that would regulate e.g. surrogate tourism, until this day. However, there is a certain guideline, namely the interest of a child, as the European Court of Human Rights judged. State's legislations range on the scale from criminalization (France) to legalization of its commercial form (Ukraine). Work-life balance and flexible forms of performance of work

The aim of this rigorous thesis was to analyze the legal regulation of flexible forms of performance of work in the Czech Republic, through which it is possible to achieve harmony between personal and working life of employees. Therefore, the institutes of flexible forms of performance of work that employees use to harmonize their personal activities and work, are described in the thesis together with their advantages and disadvantages, their legal regulation including potential insufficiency of their current legal regulation and proposals for modification of such regulation and further, there are some recommendations for their contractual arrangements. There is a general description of flexibility of performance of work in the first chapter of the thesis, including its division, connected directory and imperative character of the labor law and approaches of the European Union and the Czech Republic. In the second chapter, there is explained how the working hours and their scheduling work because that is a key topic for the other chapters of the thesis. Following chapters are dedicated to the institutes of the flexible forms of the performance of work, concretely to the uneven distribution of working hours, reduced working hours, part time, flexible working time pattern, job sharing, working time...
