Biophysical studies of membrane transport proteins from Nramp/MntH family and their function

Three synthetic peptides corresponding to transmembrane segments TMS1, TMS3 and TMS6 of secondary-active transporter MntH from Escherichia coli were used as a suitable alternative model enabling to study TMS structure, TMS interaction with membranes, TMS mutual interaction and also function of MntH. The secondary structure of the peptides was estimated in different environments using circular dichroism spectroscopy. These peptides interacted with and adopted helical conformation in lipid membranes. Electrophysiological experiments demonstrated that individual TMS were able under certain conditions to form ion channels in model biological membranes. Electrophysiological properties of these weakly cation-selective ion channels were strongly dependent on surrounding pH. Manganese ion, as a physiological substrate of MntH, enhanced the conductivity of TMS1 and TMS6 channels, influenced the transition between closed and open states and affected the conformation of all studied peptides. For TMS3 Mn2+ was crucial for formation of ion channels. It was shown that a single functionally important TMS can retain some of the functional properties of the full-length protein. My bachelor´s thesis is a digest of word problems solved through equations. Word problems are on level of 8th a 9th classes of elementary school. The work is divided into two main parts, which are divided into another parts. Main parts contain theory, elaborate examples and examples with solutions. Some examples include pictures, which help to understand situations.

Proteins - New educational materials for education in biochemistry at secondary level

Diploma thesis is focused on creation of educational materials supporting the education of biochemistry, specifically amino acids and proteins, at secondary level. At first the analysis of Czech chemistry textbooks concerning the two topics - amino acids and proteins was performed. The major problems found were related to the insufficient graphical representation of biomolecules, unsatisfactory motivational components and insufficient integration of the topic with biology or everyday life experience. The supporting educational materials, presented in this work, supplement the widely used chemistry textbooks. The materials includes a graphic oriented presentation, interactive animations demonstrating the process of denaturation and precipitating of proteins at macroscopic and molecular level, poster presenting the structural formulas of standard amino acids, 3D models of selected proteins, additional texts supporting the current topics concerning amino acids and proteins and the laboratory protocols for students. The presented support materials were evaluated at the secondary school conditions. Contractual freedom versus protection of employees

This diploma thesis deals with the problematics of the contractual freedom and the protection of employees in labour law relations and the concept of flexicurity which seeks to find balance between these two opposing notions. Its main objective is to evaluate the applicable Czech legislation regulating formation and termination of an employment relationship from the view of flexicurity, including by proposing possible changes of legal instruments within the framework of reflections de lege ferenda. The first part characterizes the contemporary Czech labour law legislation and the concept of flexicurity, followed by its reflection into the Czech Labour Code. This part concludes with an introduction of successful flexicurity models. The following part analyses both development and understanding of flexicurity at European level with the focus on impacts of EU strategy for the Member States. The key parts of this thesis are parts number three and four which deal with the legislation regulating formation and termination of the employment relationship in the Czech Republic. Resolving banking crises

The Diploma Thesis deals with the banking crisis and aims to account for different approaches to the understanding of the nature of banking crisis. The Thesis further aims to provide answers why the banking crisis are caused and why so far there are no measures that would effectivelly prevent such crisis. At first, the Thesis states the definition of banking with the emphasis on banks as the institutional backbone of the current functioning of the financial system. The Thesis pays specific attention to the characteristics of two basic legal banking instruments, particulary loans and deposits. While acknowledging the loan expansion, the thesis argues the role of regulatory authorities and the limits of their competences in regards to the prevention of crisis. Followingly, the Thesis offers a view on a banking crisis phenomenon with appropriate historical overview outlining the banking crisis in Czechoslovakia in years 1918 to 1939 and after 1989, the loan expension influence on the business cycles and approaches to banking crisis solving in respect to stabilization of the banking system and restructuring of banks. Corporate Income Tax in Accounting

The aim of this thesis is to describe in detail the issue of accounting treatment for income tax of legal persons in accordance with current accounting and tax legislation. For a comprehensive understanding of the theory display the corporate accounting is appropriate to first deal with the two systems separately and only then proceed to define the operating results, respectively. tax base as one of the bridges connecting the two systems. Transformation operating results for the income tax base is outside the corporate accounting. Information provided by accounting systems are the most important basis for the determination of the tax at the entity. An integral part of this thesis is a detailed description of the accounting operations, which may be an entity in relation to tax corporate income to meet during the accounting, respectively. period. For clarity, the final part includes practical examples of application of the tax accounting theory.

Administrative development of Pecka townlet during Carthusian era (1627-1782) based on town books

This work aims at studying and depicting a small town Pecka municipality within the time of 17th and 18th centuries, both based on the local town books. Introductory part gets the reader to know Carthusians, Pecka suzerain, as well as the Pecka domain itself and its administration. As Pecka town books have been the main archival source, its classification (accompanied by the characteristics of the town office) comes next. The core of the thesis unveils the activities of the municipality and to understand all of them, showing historical background, rules of the parish administrative and the guild system has been added as a must.

Management of Helicobacter pylori infection and dyspepsia in primary care

This thesis is devoted to dyspepsia and Helicobacter pylori infection, both significant issues for primary care. Various epidemiological and clinical studies from primary care are described. Current scientific opinions on dyspepsia and Helicobacter pylori infection important for optimal primary care management are presented. The critical interest of the author is in the severity of complaints and assessment of the individual risk of organic disease in patients presenting dyspepsia in primary care. In another study the author contributes to understanding and mutual cooperation between gastroenterologists and primary care physicians. The Lean concept arises from Toyota Production Syste (TPS). Then, the concept of leanness includes Value, Waste and Continuous Improvement. An organization with high level of leanness creates higher value to customers with lower level of waste within a continuous improvement environment. The examination of these diploma theses determines a new approach of lean measurement. Although, the use of ethnography is new in Lean management, it opens new possibilities to understand the behavior of the people about the organizational leanness.
