Characterization of soils by the DGT technique

The DGT measurement (Diffusive Gradients in Thin films technique) reflects metal ion concentrations in the soil solution, the rate of re-supply from soil to solution and the transport rate of metal ions through the soil. DGT is likely to provide a good surrogate of the bioavailable pool of metals in soils.

Aerosol behaviour in the indoor environment and relation to outdoor aerosols, including study of the influence of some indoor sources were studied using measurement of aerosol size distributions using SMPS, APS and BLPI. The size distribution of several elements and main ions were also studied.

Differential diagnosis of selected rare benign tumors of the lung and pleura

Selected rare benign tumors of the lung and pleura were studied with special reference to immunohistochemical findings and their differential diagnosis. The major objective is to better characterize unusual rare pulmonary tumors, which are unknown to most pathologists not specialized in pulmonary pathology. Moreover the clinicians, in particular, radiologists, pneumologists, surgeons, and oncologists are not familiar with these rare tumors as well. Therefore, these rare entities may cause a considerable problems in differential diagnostics and therapeutic management of the patients. The study of rare benign pulmonary and pleural tumors includes the following entities: papillary adenoma, alveolar adenoma, mucous gland adenoma of the bronchus, granular cell tumor, lymphangioleiomyomatosis of the lungs, desmoid fibromatosis of the pleura, sclerosing hemangioma associated with carcinoid tumorlet tumor, sclerosing hemangioma with with regional lymph node metastases. A special emphasis is given to previously unrecognized rare entities we have characterized in the world literature, such as multiple recurrent glomangioma of the lung, epithelial plasma cell granuloma-like tumor of the lungs, glandular papilloma of bronchus arising in sclerosing hemangioma and chondroid hamartoma of the lung associated with...

Characterization of plasma during organosilicone thin film deposition using haxamethyldisiloxane monomer

The aim of this thesis is plasma diagnostic during deposition of thin films based on organosilicones. Hexamethyldisiloxane was used as a precursor for plasma deposition in the presence of oxygen, and the diagnosis was performed by using optical emission spectroscopy. The theoretical part summarizes the basic characteristics of plasma and processes occurred at plasma polymerization and deposition. It also deals with thin films and their use in coating materials. A relatively large part is devoted to the specification of the physical-chemical nature of the used analytical method – optical emission spectroscopy. Finally, the principles of rotational, vibrational and electron temperature calculations are described. The contemporary deposition process was carried out in continuous and pulsed mode of radiofrequently excited capacitively coupled discharge. The effects of monomer flow rate, plasma power and duty cycle on the deposition process were studied. In the individual spectra, atomic hydrogen lines of Balmer’s series as well as an atomic oxygen line were identified. Molecular bands of CO have been identified as Angstrom’s system and 3. positive system. In the case of the identified fragments, the intensity dependencies on the monomer concentration in mixture, plasma power and duty cycle were detected. Electron temperature of plasma was also calculated using the relative intensities of atomic hydrogen lines of Balmer’s series. Vibrational and rotational temperature could not have been determined because no suitable fragments for their determination were found in the spectrum. Based on findings mentioned above, partial composition of plasma and some of its properties were estimated. The subject of the further research will be determination of the exact content and structure of the thin films and investigation of other plasma characteristics. Plasma deposition is influenced by many factors, and the study of their optimal combination for the most efficient deposition process is a goal for the future research.

Comparative evaluation of protein composition in human breast cancer cells using mass spectrometry

Bottom-up proteomic approach was used for detailed characterization of proteins from\ntwo human tumour cell lines representing major clinically different types of breast\ncancer. The aim was to show the differences between them on proteomic level. Here\nwe present almost 100 unequivocally identified proteins out of which 60 were mutually\ndifferently expressed for MDA-MB-231 and MCF-7. Some well-known breast cancer\nmarkers like annexins A1, A2 and vimentin were found in the MDA-MB-231 cell line.\nOn the other hand, MCF-7 cells were found to be positive for cytokeratins and keratins\nand thus we were able to distinguish both cell lines sufficiently.

Structure characterization of glucanes by 13C NMR spectroscopy

The principles of high resolution solid state 13C NMR spectroscopy are explained in this contribution. Some possibilities of this method in the structural characterization of glucans and their derivatives are demonstrated on examples of cellulose, chitin, chitosan and chitin/glucan complexes.

Biodegradation of environmental pollutants - Structural characterization of a new type of haloalkane LinB32 from Sphingobium japonicum UT26

The main aim of this thesis was focused on protein crystallography and attempt to crystallize model protein Thaumatin and a new prepared haloalkane dehalogenase LinB32 of Sphingobium japonicum UT26. Crystallization trials were performed using the sitting and hanging drop vapour diffusion methods, crystallization under oil and crystallization in the capillaries. After measuring of diffraction data, the structure of LinB32 was modeled by the use of visualizing molecular modeling programs WinCoot and VMD.
