Molecular mechanisms in homocystinuria: spatial arrangement of human cystathionine β-synthase

Protein misfolding is considered to be the major pathogenic mechanism in homocystinuria due to cystathionine beta-synthase (CBS) deficiency. The aim of this work was to study molecular mechanisms underlying protein misfolding of CBS mutants. Firstly, we studied spatial arrangement of normal human CBS protein. Using data from differential covalent labeling of solvent-exposed aminoacid residues, we identified interdomain contact area between the catalytic core and the regulatory domain in human CBS, and we subsequently generated the structural model of the full-length CBS. In the next step, we studied evolutionary divergence of CBS protein structures. We performed phylogenetic analysis that revealed unique spatial arrangement of CBS enzyme in nematodes; the domain architecture of CBS in Caenorhabditis elegans was studied experimentally in more detail. Finally, we determined conformational properties of a representative set of human CBS mutants that exhibited in various extent affected formation of tetramers and decreased catalytic activity. Using thermolysin-based proteolytic techniques for analysis of nine mutants expressed in E.coli, we found that an unfolded structure is a common intermediate occurring in CBS misfolding. Main goal of this bachelor thesis is designing a new procedure for 3D surface mapping and using it on sample data, evaluating results and comparing it with existing approaches. Theoretical part contains definition of UAV concept, its classification depending on field of use, brief history of UAVs, description of surface mapping and types of 3D reconstruction. Practical part deals with causes of false reconstruction, recommendations which should eliminate or restrict creation of problematic situations, results of reconstructions from editted images and design of proper procedure during surface mapping.

Using the contact profilometer to measure the dimensional changes of metal samples after laser treatment

The paper presents several specific examples of the use of contact profilometer Taylor Hobson. It is used here for mapping of the metal surface samples after laser hardening and remelting. There is a change in the volume of the sample after laser material processing. The contact profilometer seems to be the best choice for measuring of modification of volume and dimensions.
2D and 3D mapping of solid surfaces using Form Talysurf Series 2 device

Report deals briefly advantages and disadvantages of contact surface mapping. Despite of some disadvantages this way of exploring has its irreplaceable position in technical practice and research respectively.
