Role of antioxidant enzymes in cardioprotective regimens of cold acclimation

The effect of cold on the organism depends on the degree of cold and its duration. Cold acclimation induces a thermoregulatory response, shivering thermogenesis and subsequently the development of non-shivering thermogenesis associated with the activation of brown adipose tissue. Exposure to mild cold in the form of hardening possesses beneficial effects on the whole organism and recently the cardioprotective effects of mild cold acclimation has been described. Increased antioxidant capacity has been demonstrated in the mechanism of cardioprotection induced by hypoxia. The aim of this thesis was to find out whether the protective effect of mild cold acclimatization (8 ± 1 řC) is associated with a change in the capacity of the antioxidant system. To achieve this goal, the protein levels of the main antioxidant enzymes (superoxide dismutase, catalase, glutathione peroxidase, thioredoxin, thioredoxin reductase, aconitase, heme oxygenase) were determined during acclimation to mild cold, specifically in 1-3-10 days and subsequently after 5 weeks, and after a 2-week recovery. The results showed changes in most of the antioxidant proteins, however the effect on cold-elicited cardioprotection must be further verified. Association of genetic polymorphism of oxidative stress with diabetes mellitus type 1 and 2

Diabetes mellitus is a chronic autoimmune disease in which the immune system attacks the insulin-secreting ß-cells in the pancreas. It leads to an absolute deficiency of insulin. Chronic hyperglycemia induces increased production of reactive oxygen species, which leads to a decrease of natural antioxidant level in blood, and it contributes to genesis of diabetes complications (e.g. vascular or pulmonic). Moreover, the oxidative stress results in onset of pancreas inflammations and the damage of its ß-cells. Aims: Our aim was to assess whether or not certain genotypes or their combinations occur with higher frequency among groups of patients of type 1 diabetes (T1D) and type 2 diabetes and in a control group of healthy individuals. Methods: The study included groups of 40 T1D patients, 40 T2D patients and 45 healthy individuals. The polymorphisms of genes involved in the oxidative stress response were analyzedby using RFLP, PCR with TaqMan probes and allele specific PCR. The target genes involved superoxide dismutase SOD1 and SOD3 genes; glutathione-S-transferase GSTM1, GSTT1, GSTP1 genes; glutathioneperoxidase gene GPX1 and catalase gene CAT. Production and elimination of superoxide radical in relation to the compatibility of snails and trematodes

Almost all trematodes use snails as the intermediate host in their life cycles. To survive within the host, they have to efficiently avoid defense reactions of its immune system. The most important effector cells, haemocytes, produce reactive oxygen species with the first molecule known as superoxide radical. Various snail species produce different levels of these radicals in relation to the compatibility with the invasive trematode species. The parasite decreases the levels of toxic radicals by using antioxidant enzymes including superoxide dismutase which catalyzes transformation of superoxide radical into hydrogen peroxide. This dismutation reaction is the first step during the oxidative burst and likely influences survival of trematodes within the host. Based on the current knowledge the production and elimination of superoxide radical in relation to the compatibility between snails and trematodes have been described thoroughly for a few models such as for example Biomphalaria glabrata-Schistosoma mansoni. However, this interaction appears to play a key role and, therefore, it deserves more attention in another models as well. Oxidative stress could play an important role in the pathogenesis of inflammation and sepsis, acute and chronic pancreatitis or in the development of cancer. Organisms are protected against RONS from antioxidant system that is composed of antioxidant enzymes and non-enzymatic antioxidants. To the most important antioxidant enzymes belong superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase, glutathione reductase and paraoxonase (PON). The aim of this Doctoral Thesis was to investigate the behaviour of three of these antioxidant enzymes - CuZnSOD, CAT and PON1 in different pathophysiological states. Materials and methods: The activities of CuZnSOD, CAT and PON1 were measured in six different pathophysiological states. Forty patients with metabolic syndrome (MetS), 35 women with depressive disorder (DD), 30 septic patients (SP), 50 patients with pancreatic cancer (PC), 50 patients with chronic pancreatitis (CP) and 13 patients with acute pancreatitis (AP) were included in different studies together with sex- and age-matched healthy controls (CON). 3.2. Abstract Soil contamination by heavy metals represents rather serious environmental problem for both human health and an environment itself. One of the perspective technologies dealing with this threat that only recently has been intensely developed is phytoremediation by means of short rotation coppice plantations. As plants used in this technology (mostly poplars and willows) host two major groups of mycorrhizal fungi substantially influencing plant physiology it is important to study plant-mycobiontheavy metals interactions rather than just plant-heavy metals interactions. The present thesis aimed to contribute to the growing knowledge of the field by search for suitable mycobionts of poplar or willow tolerant to heavy metals, by evaluating an activity of the key antioxidative enzyme in selected mycobionts and by looking at physiological responses of plant hosts to their mycobionts in a soil polluted by heavy metals. The first experiment in vitro focused on screening of morphometric criteria of fungi growing on solid growth media amended with mixture of heavy metals.
