Design of algorithms for a continuous steam quality measurement system

This bachelor thesis deals with proposal of algorithm for continuous system of sterilizated steam. At the beginning we deal with the process of sterilization and its methods. Later on we talk over about the saturated steam and parameters of saturated steam. At the end of the bachelors thesis we discuss about current measurement of sterilization steam quality. Then, the procedure of simultaneous measurement of sterilization steam quality is described here. The last part is the evaluation of measured data and presentation of their results.
Fluorescence detection in pathogen analysis

This bachelor thesis deals with isolation and detection of bacteria. The used method is called molecular imprinting and is connected with fluorescence microscopy. The theoretical part concentrates on comparison of the developed method with methods that have been used till now. The practical part describes preparation and optimization molecularly imprinted polymers. These polymers were prepared on two different carriers – multititration wellplate and magnetic particles. The bacteria used as imprinted template was Enterococcus faecalis. Staphylococcus aureus was used as its competitor.
Testování účinnosti vybraných povolených přípravků proti patogenním houbovým organismům dřevin v podmínkách in vitro

This diploma thesis deals with testing the effectiveness of six selected approved preparations against pathogenic fungal organisms of woody plants in in vitro conditions. Specifically, they are Propizol (propiconazole), Phosphojet (potassium dihydrogen phosphate), Horizon 250 EW (tebuconazole), Ortiva (azoxystrobin), Topas 100 EC (penconazole) and Previcur Energy (fosetyl, propamocarb). Solutions of different concentrations (from 0.0001 % to 100 %) were prepared from these fungicidal substances, which were subsequently tested against the pathogenic fungal organisms Cylindrocarpon cylindroides and Sphaeropsis sapinea. The selected pathogens mainly attack conifers. In the case of Cylindrocarpon cylindroides, it is the genus Abies, and in the case of Sphaeropsis sapinea, the genus Pinus. All operations were carried out in the phytopathological and mycological laboratory at the Institute of Forest Protection and Hunting. Here, the tested pathogens were regularly checked and measured. After 13 weeks, the effectiveness of individual fungicidal preparations at given concentrations on 2 selected fungal pathogens was evaluated. The results show effective fungicidal preparations as well as preparations that, on the contrary, support the growth of the pathogen. The most effective fungicidal substance in both cases of pathogens was Horizon 250 EW (tebuconazole), which stopped the mycelium growth of both pathogens already at a 1 % concentration of the active substance.

The bachelor's thesis is focused on the occurrence of key pests and diseases in breeding stands of maize in the Veselí nad Moravou locality in the Hodonín district. Observations were made possible on the lands of the VESELAN Agricultural Cooperative from the harvest of the previous crop to the harvest of the corn. Three plots were selected, a different variety was sown on each of the plots, they were the Pyroxenia, Centa and Cegrand varieties. Diseases and pests were found in the stands. Among the diseases, common smut of corn and maize leaf blight appeared on the plots, from pests european corn-borer, western corn rootworm, wireworms and to a small extent aphids. The harmfulness of pathogens was more severe than the harmfulness of pests.
Evolution of intraspecific diversity of invasive pathogen of alder Phytophthora ×alni

Phytophthora alni s. l. (Peronosporomycetes: Stramenopila) is an invasive pathogen causing phytophthora root and collar rot in alders. It has been known from Europe since the '90s and from the Czech Republic since 2001. Three species were found are known within species complex P. alni s. l. Two of these species hybridized in Europe and created third triploid species called Phytophthora ×alni (Phytophthora alni subsp. alni). Dozens of genotypes have been identified with the help of microsatellites in previous studies, nevertheless only one genotype (Pxa-1) was dominating and most of other genotypes were closely related to it. The trend of increasing or decreasing incidence of this genotype differed among these studies. This thesis compares changes in the P. ×alni population in Vltava basin over the last 14 years during which a significant decrease in number of available hosts of this pathogen took place. Multilocus genotypes were determined using a set of 10 microsatellite markers. Among the 94 P. ×alni isolates reliably genotyped, 23 were of different genotypes. None of these genotypes, however, were found on the same localities repeatedly (except the dominant genotype). Incidence of the dominant genotype increased over time (from 63 % to 86 %). Septic condition from the perspective of clinical microbiology

This thesis focused on the issue of bloodstream infections from the poin of view of clinical microbiology. Diagnostics using BACT/ALERT Virtuo automatic blood culture system. In the theoretical part of the thesis, there are mentions of diseases of the blood stream, the causative agents of sepsis, and the method of isolation. In the practical part of the thesis, data obtained in the Pilsen University Hospital are processed, focusing on the most common agents causing sepsis.
Screening of carriage of group B (GBS) Streptococcus in pregnant women

In the theoretical part of my bachelor thesis, I first focus on the topic of genitourinary infections. I also mention infections that occur in men, then I deal mainly with infections of the genitourinary tract of a woman. I also mention sexually transmitted diseases. Then I go straight to the topic of my bachelor thesis, where I deal with Streptococcus agalactiae bacteria and its screening. The practical part is devoted to the adoption of methods that are used to determine the presence of Streptococcus agalactiae. The whole practical part was carried out in the microbiological laboratory SYNLAB s.r.o. in České Budějovice. The methods of common cultivation were used, cultivation that took place after multiplication of the pathogen in Todd Hewitt Broth + Antibiotics. If Streptococcus agalactiae was detected during cultivation, another test had to be carried out to ensure that this is indeed the pathogen. MALDI-TOF MS is the most frequent determinant, but latex agglutination, CAMP test or API 20 STREP biochemical test can also be used. During the research, 100 samples were processed, which were subjected to normal cultivation and then cultivation according to the recommended procedure, where the swab is incubated in the Todd Hewitt Broth + Antibiotics for 24 hours. Of the 100 samples, the positivity was shown to us after the first cultivation in 14 cases, after multiplication in the broth, another 5 were revealed. From the obtained data and data from the last 3 years provided by the laboratory, I compiled statistics, which I further compared with nationwide data. The aim of my bachelor thesis was to get acquainted with the issue of genitourinary infections and the possibilities of their laboratory diagnosis. I performed the examination of a group of patients by bacteriological cultivation of vaginal swabs and further cultivation after multiplication in the Todd Hewitt Broth + Antibiotics. Subsequently, I compared the results of these two methods and compared the results of the laboratory with the frequency of positive findings with the nationwide data.
