The remarkable structure of the cell wall of mycobacteria has a great influence on the treatment of tuberculosis itself, together with the ever-increasing incidence of multidrug- resistant and extensively resistant strains, especially in Mycobacterium tuberculosis. Infections caused by mycobacteria are widespread worldwide and a large part of the population is infected with them each year. The most common forms are pulmonary, but practically any organ of the human body can be affected. Although these diseases are treatable with combinations of antimycobacterial drugs, the number of cases with drug resistant forms of mycobacterial diseases is currently on the rise. Therefore, emphasis is placed on the development and testing of new drugs that would be able to treat these resistant forms of diseases in the future. A microdilution broth method with already proven procedure on Mycobacterium aurum and Mycobacterium smegmatis strains was used to determine the minimum inhibitory concentration of compounds. Altogether 28 compounds of natural origin from the Department of Pharmaceutical Botany of the Faculty of Pharmacy of the Charles University were tested. Tested was the sensitivity to potentially antifungal substances with 8 micromycete strains, namely Candida albicans, Candida krusei, Candida parapsolosis, Candida tropicalis, Aspergillus fumigatus, Aspergillus flavus, Absidia corymbifera, Trichophyton interdigitale. Methods Molecules were tested with the broth microdilution method. Results 28 newly synthesized compounds were put to the test to prove the antimycotic effect. 11 out of them proved to be hopeful for further testing. Antifungal activity against all testing micromycetes was found with 5 compounds, 6 compounds proved antifungal activity against at least one micromycete. Klára Doležalová Supervisor of diploma thesis: PharmDr. Ondřej Janďourek, Ph.D. Title of diploma thesis: Pathogenesis of infectious disease caused by bacterium Mycobacterium tuberculosis The theoretical part of this diploma thesis is concerned about tuberculosis where the main part of the thesis is focused on pathogenesis of this disease. Tuberculosis is recognized as highly infectious disease leading to the total exhaustion of the organism. The main causative agent of the above mentioned disease is Mycobacterium tuberculosis, which is transmitted to respiratory tract via inhalation of droplets from infected person and leads to the granuloma formation. For tuberculosis is typical a creation of granulomas, mainly with caseous necrosis. Highly important is accurate diagnosis and appropriately chosen treatment. The treatment consists of a long-term administration of antituberculotic drugs in combinations. Marcela Vejsová, Ph.D. Name of diploma thesis: Evaluation of activity of potentional antibacterial substances throught of the use microdilution broth metod Background Aim of this diploma work was research of activity of potentional antimicrobiotic substances. Research of antibacterial substances is one of the most important factor in pharmaceutic industry. The main reason is being the never stopping growth of bacterial resistence. Methods The microdilution broth metod was used to test the substances. This metod was used because of low difficulty and low cost. All the steps including the final analising was done by hand. Results The substances were divided in groups depending their chemical struction. The most effective was the salicylanilide derivates group. The other groups inhibitated growth of bacteria very little or were non-functional. In case of every bacterial stems were analised all substances which were effective. Conclusion Depending on the results the most sensitive and the most resistant bacterial stem were choosen. The most sensitive reaction on tested substances had bacterial stem Staphylococcus aureus. Lenka Králíková Supervisor: PharmDr. Ondřej Janďourek, Ph.D. Title of diploma: Atypical mycobacteria as causative agents of human diseases The aim of this diploma thesis is to summarize the history of mycobacteria, their classification, epidemiology of nontuberculous infections, clinical significance, diagnosis, resistance, and treatment options. Within the epidemiology, the way of managing tuberculosis and non-tuberculous mycobacterioses is compared. Mycobacterioses are rare diseases, which incidence is gradually increasing with the increasing number of immunocompromised patients. Based on the spot of infection, we divide them into lymphadenitis, skin lesions, lung disease and disseminated infections. The most frequently isolated pathogens are Mycobacterium avium complex, Mycobacterium ulcerans, Mycobacterium abscessus, Mycobacterium marinum and Mycobacterium kansasii. Tomáš Hampl Supervisor: PharmDr. Ondřej Janďourek, Ph.D. Title of diploma thesis: Mycobacterium tuberculosis as causative agent of grave illness The aim of this diploma thesis is to deal with tuberculosis, describe the pathogen, taxonomic classification, describe the origin, course and clinical manifestations of the disease, diagnosis, including laboratory diagnostics, epidemiology of tuberculosis, treatment and prevention. Tuberculosis (TBC) is one of the deadliest infectious diseases in the world caused by bacterium M. tuberculosis in humans. TBC is sometimes referred as a disease of the past, but the recent rise in cases of the disease, and especially the spread of forms of drug-resistant TBC, makes the disease very dangerous. Currently, a major problem is the alarming rise in resistant and multi-resistant tuberculosis, which is not counteracted by commonly used first-line drugs.
