Identification DNA of Plant and Animal Species in Food by Polymerase Chain Reaction

We were developing detection methods for three food allergens of plant origin. We used real-time PCR for soy detection in food oriented on gene lec, that is coding lektine specific for soy. On this target sequence were oriented PCR system with primers Le2F and Le2R and TaqMan probe Le2P. Detection limit (2,75 pg), practical detection limit (0,02 %), inclusivity and exclusivity were determined. Whole system were quantified. Real-time PCR for pistachio detection were based on primers and probe for gene COR. Detection limit (3,5 pg), practical detection limit (0,002 %), inclusivity and exclusivity were determined. For almond detection we were not succeed system, that fulfil all qualitative parametres.
Characterization of selected vegetable oils for application in food

This bachelor thesis deals with the characterization of selected types of nut oils made from almonds, walnuts, and hazelnuts. These oils were cold pressed, and they are available on the regular market. The theoretical part deals with the characterization of individual types of nuts and nut oils. The experimental part deals with the comparison of oils in terms of profile and content of volatile substances and fatty acids. The volatile compounds were identified by gas chromatography with mass spectrometry in conjunction with solid phase microextraction. Ten substances, mainly alcohols, were identified in the nut samples. Most substances were found in hazelnut. A total of 44 volatile substances were found in oil samples, mostly in walnut oil. The main substances were aldehydes and alcohols. Fatty acids in free and bound form were converted to methyl esters by acid esterification with methanolic boron trifluoride solution as catalyst. Fatty acids were identified by gas chromatography with flame ionization detection. Unsaturated fatty acids, mainly oleic (almond, hazelnut) and linoleic (walnut) acids, were present in large quantities in the samples.

Tato bakalářská práce je soustředěná na zkoumaní přírodních komerčně dostupných extraktů, které se používají do přírodní kosmetiky. Literární rešerše byla zaměřena na téma obsahu těkavých látek v přírodních extraktech používaných v kosmetice, jejich charakterizace a možnosti stanovení, problematika výskytu alergenů v přírodní kosmetice. Na základě tohoto přehledu byla navržena optimální metoda pro analýzu vzorků přírodních extraktů s přihlédnutím k dostupné analytické instrumentaci. Následující výtažky byly studovány se zaměřením na kvalitativní stanovení: mandle, aloe vera, hřebíček, Etiopská káva, zázvor, cibule.
