Utilization of thermal analysis in study on protein denaturation

This bachelor thesis deals with the use of techniques of thermal analysis in the study of thermal denaturation of proteins. As a model protein was chosen lysozyme. In the experimental part were used two methods of thermal analysis – differential scanning calorimetry (DSC) and temperature modulated differential scanning calorimetry (TMDSC). One of the objectives of the study was to determine the utility of DSC and TMDSC in the study of lysozyme denaturation and to futher characterize the results we can obtain from the individual methods. Another goal was the optimization of TMDSC. During optimalization of TMDSC heat-iso, heat-only and heat-cool modes were compared in 60 seconds and 100 second periods. By selected methods was observed denaturation of lysozyme. Denaturation of lysozyme is predominantly created by reversible component of the heat flow and the denaturation temperature is in the range of 71,16 °C to 75,21 °C depending on the set parameters.
Preparation of some application formula of antimicrobial substances

The main aim of this study is to determine antimicrobial activity of herbal extracts and also encapsulate these extracts and lysozyme to liposomes. The extracts were prepared from above ground part of wormwood (Artemisia absinthium), aerial part of breckland thyme (Thymus serpyllum), the root of purple coneflower (Echinacea purpurea), liquorice (Glycyrrhiza glabra), corolla of pot marigold (Calendula officinalis) and berries of sea buckthorn (Hippophae rhamnoides). In all extracts antioxidant activity and the amount of total polyphenols and flavonoids was determined. All of these extracts and also lysozyme were used for encapsulation to liposomes. Prepared liposomes were then tested for their stability. Further, encapsulated as well as free extracts were tested for their antimicrobial activity on Escherichia coli, Serratia marcescens, Micrococcus luteus and Bacillus subtilis. Extract from Echinacea purpurea showed the highest antimicrobial activity.
Molekulárna charakterizácia vybraných obranných faktorov v čeľadi Lumbricidae

Earthworms belonging to oligochaete annelids have been a model for comparative immunology for over 40 years. They possess various defense mechanisms efficiently recognizing and responding to non-self substances. Among these there are molecules with many biological activities including cytolytic, antimicrobial and proteolytic. This work is aimed to compare the immunological features of two closely related earthworm species Eisenia andrei and Eisenia fetida. Due to many morphological and life cycle similarities they have been, until recently, regarded as members of subspecies. Interestingly, their natural habitat varies considerably, and it was of particular interest to investigate how these environmental differences affect the features of innate immunity of both species. Variability of the domestic chicken breeds in selected immunological traits of hen and egg

The avian immune system is a complex system of defence mechanisms that protect bird hosts against threats from ubiquitous pathogens. According to the co-evolutionary models, variability in immune traits of hosts is the key component providing ability to adapt and enhance their defence mechanisms in presence of constant selective pathogen pressure. Domestic chicken (Gallus gallus f. domestica) is used as a model organism in avian biology and also is one of the most important food-producing animals, not only for their meat but also for the egg production. Unfortunately, in research usually only inbred chicken lines are used and modern poultry husbandry is tight with unilateral breeding towards highly productive breeds. Those approaches decrease intra-population polymorphism in chickens. However, especially in case of farm animals, searching and extending the pool of immune variability and enhancing pathogen resistance is crucial for sustaining healthy and biologically secure populations and their products. Morphologically highly distinct traditional chicken breeds, which have evolved for hundreds years under different selective pressures, may represent this desirable immunological variability.
