Contamination of the environment by crude oil originated fuels, its detection, remediation and prevention

This bachelor thesis focuses on crude oil-based fuels and water and soil contaminations caused by these substances. Crude oil and the process of producing fuels out of crude oil as well as the fuels themselves (car petrol and diesel oil) are characterized in the work. The thesis also describes the methods used for their detection in the environment, the decontamination processes and the ways of petroleum products leaks prevention.
Study of the occurrence of colistin in soil

Colistin is a polypeptide antibiotic used as a "last resort" effective against Gram-negative bacteria. However, increase in its consumption in veterinary medicine in the last 30 years has led to the development of bacterial resistance even to colistin. The application of slurry to soil containing unmetabolized colistin and resistant bacteria poses a risk, because the resistance of bacteria to colistin can be further developed and spread to other components of the environment. Therefore, this work was focused on the creation and optimization of extraction and analytical methods for soil samples containing colistin. Colistin was detected by liquid chromatography in connection with mass spectrometry (HPLC/MS and UPLC/MS/MS). Many extraction solutions have been tested in connection with ultrasonic extraction and purification on various SPE columns, yet colistin has not been successfully extracted from the soil. Colistin was successfully detected only in the slurry extract. The reason why colistin was not detected in soil extracts could be the sorption of colistin on soil organic matter or the formation of complexes of colistin with humic substances.

This thesis focuses on pesticides in the environment, more specifically in cereals. Pesticides were monitored in various samples of cereals (barley, rye, oats, and wheat) within the thesis. The monitored substances were isolated in selected samples and concentrated using the QUECHERS method. Gas and liquid chromatography were used to determine pesticides. The theoretical part consists of the common types of pesticides, divided by the ease with which these substances into food and their impact on humans and environment. It also mentions current health protection legislation and environmental legislation.

Vývoj elektrochemického detektoru pro monitorování těžkých kovů

The aim of this dissertation thesis "Development of electrochemical detector for heavy metal monitoring" was to create sensible sensors for the determination of heavy metals in the aquatic ecosystem and which would be usable for real applications, determination of the content of various metals in waste industrial waters. For the simultaneous determination of Zn(II), Cd(II), Pb(II) and Cu(II), a carbon paste electrode modified with chromium oxide was developed, based on this modification were produced the screen printed electrodes for the simultaneous determination of said metals with good limit of detection. For the usability of printed sensors in situ, a new electrochemical cell design was designed. This proposed cell also facilitates dosing of samples and buffer necessary for electrochemical analysis. Further, for determination of the total chromium content in aqueous solutions, was introduced a new method using a silver film on a glassy carbon electrode. Finally, various methods for waste water treatment from heavy metals have been tested.
