Preprocessing of 1D gel electrophoresis image

The aim of this work is to propose an algorithm in MATLAB, allowing image processing and evaluation of one-dimensional gel electrophoresis. At first describes the detection of samples based on the horizontal segmentation of electrophoresis gel image, followed by detection of the minimum intensity values of the image, a vertical segmentation and determination of the sample boundaries in the image. The final phase involves testing of the method on publicly available images and evaluation of the results.

Gel electrophoresis is an important method used for separation and analysis of macromolecules with electric potential. This well-established method has a big potential in medicine and science, however, the output images often suffer from distortion. This work describes several known sources of distortion and elaborates on existing methods of the track detection in gel electrophoresis. The core of this thesis introduces a new method to detect individual tracks and image pre-processing based on the standard deviation calculated on the horizontal segmentation of the image. The proposed algorithm is implemented and tested in Matlab. The success rate of the proposed method reaches 96,7 % on 35 test images of gel electrophoresis. The next phase is the band detection in the vertical profile and the binary image drawing. Although the band detection is not fully automated, a detection success rate achieves up to 86,4 % when proper setting is used.
