Content of adaptogens in vegetable Schizandra chinensis

This thesis deals with the content of adaptogens in vegetable Schizandra chinensis. The theoretical part deals with the plant Schisandra chinensis, its occurrence, characteristics, uses and cultivation. Location of plants Schisandra chinensis are classified according to Köppen climate classification and compared with the climate in the Czech Republic, due to possible prediction of growing plants in the country. Further are described adaptogens (bioactive substances) contained in this plant and to methods for their extraction and analysis. The experimental part of the thesis deals with the identification of schisandrin in the plant Schisandra chinensis grown in our conditions. The assessment was performed by ultra-performance liquid chromatography (UPLC), high-performance liquid chromatography (HPLC) and direct injection into tha mass spectrometry (MS). Schisandrin was successfully identified in all the samples.
Issues of perfluorinated compounds in the environmental compartments

Perfluoroalkylated substencies are priority organics polutants. They are evolved to the environment from many sources. We classify them as new xenobiotics which are present in the environment. There will be described their chemical, physical and environmental properties in theoretical part and there will be described the analytical chemistry of these substancies in varios types of environmental samples. There will be developed metod for their identification and quantification. For this assignment will be use especially chromathografic methods. There will be developed standard operation procedure. The procedure will be verify on real environmental samples.
Application of acid proteases from nepenthes in hydrogen/deuterium exchange.

Application of acid proteases from Nepenthes in hydrogen/deuterium exchange Petra Darebná (Department of Biochemistry, Faculty of Science, Charles University in Prague, Czech Republic) Nepenthes are mostly found in Borneo and Sumatra. They are one of a few carnivorous plants which produce its own proteolytic enzymes (nepenthesin I and nepenthesin II), which provide an alternative source of nitrogen and other nutrients in case that these plants grow in a soil which lacks such nutrients. These aspartate proteases are capable of proteolysis at a very low pH of a digestive gastrovascular cavity fluid after catching insects and in a cooperation with other proteins participating in digestion process. Processing of a digestive fluid, isolated from a digestive gastrovascular cavities of carnivorous plants of the Nepenthes genus and its possible application as a tool in a protein study using hydrogen/deuterium exchange were done in this thesis. Isolates of the digestive fluids were purified from coarse-grained impurities by centrifugation, activated by acidification and concentrated by ultrafiltration. The amount of proteins and their protein profile were monitored and an activity of acidic proteases was determined by enzymatic assay. Proteomic characterization of human NK cell membrane microdomains

Proteomic characterization of human NK cell membrane microdomains. (in Czech) Bc. Alan Kádek (Department of Biochemistry, Faculty of Science, Charles University in Prague, Czech Republic) Natural killer (NK) cells are one of the important components of innate immune system. Their main function is to fight against tumors, virally infected or otherwise malformed cells. Plasma membranes of NK cells contain regions with specific lipid composition compared to the surrounding membrane (called membrane microdomains or rafts). Because of their lipid composition, microdomains preferentially accommodate some immunologically relevant proteins and play a role during cellular polarization and signalization. Characteristic feature of membrane microdomains is their partial resistance to solubilization by mild non-ionogenic detergents. In this thesis, microdomains were isolated in a detergent-resistant membrane fraction (DRM) from human NK-92MI cell line and from NK cells immunomagnetically enriched from peripheral blood of non-leukemic donors.
