Analytical methods for verifying the authenticity of the wine

This bachelor’s thesis deals with the authenticity of wine, focusing on the authenticity of geographic origin. The theoretical part of this work summarises the main components of wine and their changes during the production proces. Control techniques are introduced, including the ones used by the goverment’s control bureau (SZPI) and other documented methods. Areas recognised for growing wine are described, including the Morava area in Czechia and its imminent adjacent areas in Austria and Slovakia. Experimental part deals with the trace amount analysis of selected elements and phenolic compounds. The element analysis was conducted using mass spectrometry and optical emission spectrometry, whilst the phenolic compounds were separated using high-pressure liquid chromatography with spectrophotometric detection. The results were analysed using discrimination analysis to separate groups with different areas of origin, with the aid of data from former research.

This thesis deals with the analysis of wines using high-pressure liquid chromatography in order to analyze the content of alcohols, organic acids, and carbohydrates in wine. The most important groups and their representatives are presented in the literary review, as well as the theoretical basis of chromatographic methods, selected types of detectors, and used stationary and mobile phases. In the experimental part, analytical parameters of wines from the retail network, laboratory-prepared must and wines prepared by fermentation were measured using liquid chromatography methods with refractometric and UV detection. The results indicate the suitability of this combination of detectors for the selected wine analyses.
Determination of tryptophan, serotonine and melatonin in plants by using HPLC

This thesis deals with the development and optimization of a method for the determination of tryptophan and its metabolites - serotonin and melatonin - in plant material, in grapevine, during one analysis. It uses a high-pressure liquid chromatography. The theoretical part is about tryptophan, its metabolism and basic properties of its metabolites - serotonin and melatonin. Their occurrence in wine is also discussed. Analytical techniques by which these analytes can be determined are also provided. Then information about modern stationary phases, that are suitable for this species, is included. The experimental part consists of optimization of the method, measurement of calibration dependences and measurement of real samples. It is measured by the method of reverse phase chromatography. As first stationary phase it is used a C18 column with core-shell packing, second is a BEH Phenyl column. The mixture of 10 mM acetate buffer (pH = 4.5) and methanol is used as the mobile phase. For detection UV at wavelength 254 nm is used, then for greater sensitivity mass detectionis is used. The basic conditions for the experiment have been set.
