The importance and methods of measuring hematocrit and hemoglobin in pre-hospital emergency care

This bachelor thesis is called: "The importance and methods of measuring hematocrit and hemoglobin in pre-hospital emergency care". There has been recent progress in technologies which allows to measure various laboratory values by small portable systems. These values can be Hb and Ht, measuring of these values is not standard in pre-hospital medical care in Czech Republic. It is the reason why this thesis deals with possibility of its implementation. The theoretical part deals with composition and physiology of hemoglobin and associated diseases such as anemia and polyglobulia. It also contains the issue of diagnostics, guidelines of physical examination procedures as well as description of what should be focus on when one is gathering case history. There are also provided facts and information about portable measuring systems, their measuring methods and overview of systems which are available in Czech Republic. There was conducted a qualitative research and was presented in practical part of this bachelor thesis. Collection of data was based on technique of standardized anonymous questionnaire which included only structured answers. The paramedics of Emergency medical services of South Bohemia region were the respondents. The research took place in March 2014 and 54 respondents anticipated in it. There were stated two aims in this bachelor thesis. The heart of first aim was to find out whether the paramedics are able to use values of hematocrit and hemoglobin to diagnose a patient. The second aim was to map paramedics' attitude to measuring values of hematocrit and hemoglobin within emergency medical service. There were also stated hypothesis which expressed the expectations that paramedic are able to use values of hematocrit and hemoglobin and they are interested in and enthusiastic about measuring hematocrit and hemoglobin within pre-hospital emergency care. From data, which were collected and analyzed in the practical part, it is obvious, that paramedics do have satisfactory amount of skills and knowledge which are absolutely necessary for diagnosing a patient by using values of hemoglobin and hematocrit. Contrary to the fact that paramedics have certain knowledge about benefits which would measuring values of hemoglobin and hematocrit brought they do not believe that portable measuring systems would be efficient in pre-hospital emergency care and they are not interested in using them.
Biometric System Performance Testing - Vein Technology

This thesis aims at performance testing of a biometric device based on a vein recognition technology. Basic principles of this technology are briefly described. PalmSecure device was tested to illustrate the principle of the vein pattern scanning. Preformance tests and tests to fool the device were carried out. Finally, based on all acquired information a prototype of a palm vein pattern scanner was designed.
Pulse Oxymeter in LabVIEW

One of the most important vital functions of the human body is providing sufficient oxygen to all its parts. Oxygen in the blood binds to hemoglobin and it is transported to tissues. Oxymetry is non-invasive method for measuring the amount of oxygen bound to hemoglobin. Oxygenated hemoglobin and deoxygenated have different absorption characteristics for light of different wavelengths. Therefore, pulse oxymetry transmits tissue with red and infrared light. Signal passing through the tissue is affected by the absorption properties of hemoglobin and heart pulse. These signals are measured and the ratio of their size allows us to calculate blood oxygen saturation.
Oxygen blood saturation measurement

This thesis works with measuring possibilities of blood oxygen saturation, analysis methods used and their calibration. It also deals with design of the block diagram of pulse oximeter for measuring blood oxygen saturation.
Hemoglobin-mediated oxidation of marine liposomes

Cílem této práce bylo studium mechanismu oxidace lipidů katalyzované hovězím methemoglobinem a zhodnocení účinků různých experimentálních podmínek a antioxidantů (EDTA, askorbová kyselina, kávová kyselina, a-tokoferol, d-tokoferol, astaxanthin a L-askorbyl-6-palmitát) na methemoglobinem zprostředkovanou oxidaci lipidů v modelovém systému liposomů připravených z fosfolipidů. K monitorování oxidace lipidů při pH 5,5 a teplotě 30 °C bylo použito spotřeby kyslíku. Pro zhodnocení antioxidační aktivity v modelovém systému liposomů se ukázaly být důležitými faktory typ prooxidantu a koncentrace prooxidantu a antioxidantu. Dalšími důležitými faktory jsou struktura molekuly antioxidantu, jeho hydrofilita/lipofilita a umístění v systému. Všechny testované antioxidanty ve všech koncentracích (kromě koncentrace 0.1 % astaxanthinu and 0.1 % askorbyl palmitátu) inhibovaly oxidaci vyvolanou methemoglobinem. Účinnost antioxidantu stoupala s jeho zvyšující se koncentrací. Koncentrace 0.1 % astaxanthinu neměla žádný vliv na oxidaci liposomů. Koncentrace 0.1 % askorbyl palmitátu měla prooxidační efekt, který lze vysvětlit prooxidačním působením radikálu askorbylu, který může urychlit štěpení hydroperoxidů. Volné železo uvolněné z methemoglobinu se podílelo jen velmi málo na oxidaci liposomů, zatímco část prooxidační aktivity methemoglobinu byla přisouzena tvorbě singletového kyslíku (methemoglobin jako fotosenzitizátor). Antioxidační aktivita astaxanthinu, askorbyl palmitátu a tokoferolu byla z části přisouzena schopnosti zhášet singletový kyslík. Ovšem hlavním prooxidačním mechanismem methemoglobinu se ukázal být rozklad lipidových hydroperoxidů, tvorba volných radikálů a hypervalentních forem hemoglobinu. EDTA utlumila oxidaci liposomů díky chelataci přechodných kovů obsažených v liposomech a chelataci volného železa přítomného v methemoglobinovém roztoku. Velmi důležitým antioxidačním mechanismem (který vykazují askorbyl palmitát, askorbová a kávová kyselina) se ukázala být redukce hypervalentních forem hemoglobinu. Askorbová kyselina, kávová kyselina, tokoferoly a astaxanthin inhibovaly methemoglobinem zprostředkovanou oxidaci lipidů odstraňováním volných radikálů. Při použití peroxidu vodíku nebyl pozorován žádný vliv na oxidaci liposomů vyvolanou methemoglobinem. Působení vysoké teploty (tepelná denaturace) mírně utlumilo oxidaci. Významná inhibice oxidace byla pozorována u liposomů obsahujících TPP (triphenylphosphin), což značí, že je methemoglobinem vyvolaná oxidace liposomů závislá na přítomnosti již vzniklých lipidových peroxidů. Výsledky této práce přispívají k hlubšímu pochopení prooxidačních a antioxidačních mechanismů a faktorů, které ovlivňují oxidaci liposomálních roztoků, buněčných membrán a emulzí typu olej ve vodě stabilizovaných fosfolipidy.
Evaluation of antioxidant effect using different analytical methods

Cílem diplomové práce je zhodnotit antioxidační účinky pěti různých sloučenin s potenciálním využitím jako antioxidanty v potravinách (propylgalát, kávová kyselina, ferulová kyselina, p-kumarová kyselina a L(+)-askorbová kyselina), a to prostřednictvím čtyř běžně používaných testů antioxidační kapacity (FC, FRAP, DPPH, ABTS) a v modelovém systému liposomů za použití volného železa (Fe2+, Fe3+) a hovězího hemoglobinu jako prooxidantů se zvláštním zaměřením na různé koncentrace testovaných sloučenin v systému. K nepřetržitému monitorování oxidace lipidů při pH 5,5 a teplotě 30 °C je použita spotřeba kyslíku. Výsledky této práce mají přispět k lepšímu pochopení základních pro- a antioxidačních mechanismů a faktorů ovlivňující oxidaci buněčných membrán, liposomálních roztoků a emulzí typu olej-ve-vodě stabilizovaných fosfolipidy.
Influence of dietary components and redox enzymes on intestinal microbiota proliferation in the tick \kur{Ixodes ricinus}

In this work, we have analysed the temporal dynamics of gut-dwelling bacteria and Borrelia in the gut of the deer tick Ixodes ricinus. Using quantitative PCR, we have shown that levels of the tick intestinal microflora are profoundly decreased at later stages of feeding on whole blood but not on serum. Even though we noted that host complement system manages to interfere with Borrelia viability in vitro, we did not see any effect of host complement on Borrelia acquisition in adult ticks in vivo. However, we revealed that host hemoglobin is essential for Borrelia proliferation in the tick gut. All together, these data imply that, during feeding, levels of gut-dwelling bacteria and Borrelia are determined by the host. While hemoglobin seems to be detrimental for gut-dwelling bacteria, Borrelia require it in order to proliferate. During off-host stage, we showed that levels of gut-dwelling microflora are regulated by an intestinal transmembrane enzyme Dual oxidase. In conclusion, we aimed, and mostly succeeded, to perform pilot experiments describing the biology of a complex process of regulating gut microflora in the vector Ixodes ricinus and extend it by its impact on Borrelia acquisition
Seasonal dynamics of selected blood parametres of selected flesh breeds of sheep bred in foothills conditions

Sheep breeding is nowadays a developing branch of agriculture again. There is a lot of different breed and they react distinctly on the same conditions of the enviroment. Sheep of breeds charollais, suffolk, šumavská ovce and valaška bred in similar conditions were observed in spring and autumn of years 2007 and 2008. In these seasons were taking blood samples (from {$\pm$}7{--}24) ewes and lambs and were analyzed in hematology laboratory. There were determined haemoglobin level, haematocrit indicator, erytrocytes and leucocytes levels, glucose, cholesterol, triglycerides levels, urea and plasmatic proteins, activity of ALP and GMT enzymes, phosphor, calcium, magnesium, zinc and copper levels. The main objective of this project was determine seasonal changes in observed parametres. Next objectives were determine different changes in blood parametres in different breeds and different aimes of breeds. There were recognized that all the observed breeds don`t react the same way on similar conditions. There were not provably determined seasonal changes in observed parametres, but average Hb level was higher in autumn than in spring. Urea level was conversely higher in spring season than in autumn. The demostrable fact is, that the similar conditions induce different answers not only in different breeds but also in different aimes of breeds.
