Mechanisms of microbiota regulation during the estrous cycle of the house mouse.

There is a very few papers to provide an overview of the characteristics of the estrous cycle, the relationship of the estrous cycle to physiological manifestations such as the pH of the vaginal environment, as well as the dynamics of the vaginal microbiota in wild mice. The aim of this thesis is to contribute to the understanding of the dynamic relationship between external influences and the physiology of the female reproductive system, to develop a reliable methodology for measuring the pH of the vaginal microenvironment in mice as well as to quantify the overall abundance of some bacterial taxons by comparing sequencing and qPCR methods. The results suggest that the physical presence of the male in the cage has the most significant effect on the prolongation of the estrus phase, in contrast to non-significant olfactory stimulation of the urine. Fluctuation in the pH of the vaginal environment have also been shown to be cyclic, and the qPCR method shows that the composition of the vaginal microbiota, during the estrus phase, differs significantly from other phases of the estrous cycle, as we confirmed by 16S rRNA sequencing. Reprodukce slona afrického (Loxodonta africana) v Zoo Zlín - Lešná

ŠIMKOVÁ, D. Reproduction of an African elephant (Loxodonta africana) in a Zoo Zlín – Lešná. Mendel University in Brno, 2019. Diploma thesis. The thesis analyzes the issue of reproduction of critically endangered species of African elephant. The work is focused on monitoring of animal weight and blood test results during the reproductive cycle. The first part describes the general characteristics of the reproduction cycle of the African elephant, the issue of pregnancy and childbirth, and the female reproductive system. In the second part, there are given data from three particular elephants from the Zoo in Zlín. The data are processed into graphs and these are then commented on and compared with the conclusions of already conducted studies and scientific papers. At the end, all the knowledge and forecast of the African elephant reproduction in the Zoo - Lešná in the future are evaluated. All the results show that females are fully capable of reproduction and there is no need to make any changes in the breeding of these animals.

In this study we employed a standard open-field test as an approximation of the first phase of dispersal, using females from wild-derived inbred strains representing two subspecies of the house mouse, M. m. musculus and M. m. domesticus. We tested the prediction that exploration of neutral area varies during different phases of the oestrus cycle and is different between the two subspecies. We also compared female's behaviour with male exploration tested in the same conditions.
Bacterial populations in mucosal tissues of the house mouse

Microbiota becomes one of the most important subjects in biological research and numerous studies revealed that microbiota plays a broad spectrum of essential roles in different organisms. This master thesis focuses on the bacterial part of microbiota contained in mucosal tissues of wild house mice (Mus musculus musculus). Male and female samples were collected by nasal and oral cavity lavages, vaginal mucosa lavages and from urine and stool. We aimed to detect quantitative, qualitative and sex-specific differences in bacterial populations between mucosal tissues with particular focus on bacterial cycling in vaginal mucosa during the estrous cycles. Bacterial abundances were estimated by qPCR whilst bacterial diversity was detected by targeted metagenomic sequencing of the hypervariable region of the 16S rRNA gene. Significant differences were detected in bacterial abundances and alpha diversity between particular mucosal tissues. Stool samples contained the highest number of bacteria, while samples from the nasal mucosa and urine contained low amount of bacteria. The highest alpha diversity was discovered in stool samples, the least alpha diversity was found in the urine.
