Microbiological Project Using the Disc Diffusion Method for Lower Secondary School ABSTRACT This thesis deals with project-based education and his involvement in the functional structure of the czech educational theory and practice. It refers to the project as an innovative teaching process, which is qualitatively able to shield asked RVP requirements and the implementation of which contributes to the development of educational goals and core competencies. This thesis contains both theoretical aspects are developed through the study of literature and other sources in order to summarize the current state of knowledge of the subject, so practical. Theoretical bases emphasize not only on the characteristics of project-based education as such, but also on the appropriate application of educational strategies in the learning process. Specifically mentioned are possibilities of project-based teaching in the teaching of natural history, biology and ecology. The theoretical background followed by the practical part, which shifts the theory of project learning into practice. There is particular designed a project called "Strength of antibiotics", which is then implemented and reflected. This diploma thesis deals with the antimicrobial properties of herbal substances. Characteristics of these substances, including mechanism of action on microorganisms and their industrial use, is presented in the theoretical part. The aim of microbiological analysis was to determine the extent of diluted (10%) and concentrated essential oils isolated from clove (Syzygium aromaticum), cinnamon (Cinnamomum), and thyme (Thymus vulgaris) against bacteria (Escherichia coli, Enterococcus faecalis, Enterococcus faecium) and yeast (Yarrowia lipolytica, Candida tropicalis and Saccharomyces cerevisiae). To determine them, the disc diffusion method was used. The results confirmed an antimicrobial effect of tested essential oils. Concentrated thyme essential oil had the hightest inhibitory activity on all used microbial strains.

This paper is dedicated to gathering information about utilization of Vitis vinifera L. extracts, mainly grape seed extract (GSE), production of GSE in two variants (aqueous or methanol GSE) and description of its characteristics. Extract was rich in polyphenols (13462 mg/l GAE), measured by Folin-Ciocaltel method. Also 3615 mg/l GAE of flavonols was present in GSE. Antioxidant activity measured by DPPH method was high, 7978 mg/l GAE. Disc diffusion method was used for antimicrobial activity analysis. GSE's effect was tested on five common microorganisms. Results of the experiment show that GSE strongly inhibits growth of Bacillus cereus, Candida tropicalis and Enterococcus faecalis. However, it was not effective against Escherichia coli and Pseudomonas aeruginosa.
