The baptism as a rite of passage connected with the childbirth

The bachelor's thesis is focused on the investigation of symbolism and ritual action in the context of the Christian liturgy of baptism. I place special emphasis on the element of physicality, the dimensions of space and time, as aspects of the liturgical ceremony and their integration into the dialogical concept of the liturgy. My goal in this work is to point out how dialogue, the element of physicality, the symbolism of time and space are "transcribed" into the baptism ceremony as a transition ritual. At the same time, my intention is to show how important the life situations of the individual are, and how much the Easter mystery of the story of Jesus Christ is the primary purpose of the baptism ceremony.

This thesis deals with nitrogen ions, i. e. nitrite and ammonia nitrogen in samples of natural waters and the stability of the resulting values. The theoretical part includes list of determination methods of nitrogen in aquatic environment. The practical part contains results of nitrogen ions concentration in chosen water samples and in standards. The aim of this thesis is to evaluate the impact of conservation reagents used to determine nitrogen ions on the final value and changes in these resulting values in a time depending on the analysis. Selected determination of nitrogen ions in natural water samples were performed from June 2014 to April 2015. Consequently the values were graphically processed and evaluated. A conservation reagent affects the resulting values only for the determination of ammonium ions. Stability of measured ion concentration of nitrogen in water samples is at least two hours, even with ammonium nitrate nitrogen it can be extended by conservation cold of samples.
Infrastucture and time to trade as barriers

Bachelor thesis is oriented on analysing of the impact of time, infrastructure and logistics on international trade. Time and the quality of infrastructure do not influence only trade volumes, but what is even more important, the long procedures for export and import and disorganized infrastructure contribute to the reduction of the probability that companies or countries will enter time-sensitive products markets, where for instance consumer electronics and clothing belong to. In the practical framework, the application of the gravity equation for bilateral trade was used in order to find out whether it is significant on given sample of European Union countries in the year 2012. By means of using statistical and econometric tests such as Fisher´s test, RESET test or Test of a Goodness of Fit, the overall significance of the model and individual variables was proved.

There were analyzed the amount of caffeine, amounts of polyphenols, color, sensory properties and the spectra were measured using near infrared spectroscopy in three kinds of coffee. Analyses were made after roasting coffees. The coffee was then packaged into three different types of packaging - paper, package with one-way valve and into a vakuum package, each series of samples were stored in three different conditions - normal conditions (20 °C, 40% RH), a fridge (4-6 °C, 80% RH) and variable conditions (15-30 °C, 40-60% RH). After three, six and nine months was then carried out analyses for all samples. Subsequently the differences between the baseline sample and the results in different time were evaluated and the results were compared. It was found out that coffee quality sharply decreased after three months, caffeine over time decreased slightly. Sum of polyphenols sharply decreasesd after 3 months and their value was at a similar level. The color of the coffee changed after 3 and 6 months rapidly, but then the colour approached the original surface.
Nurse communication with the bereaved in practice.

This bachelor´s thesis describes the issue of communication, its division and communication barriers. It also explores communication with the bereaved, caregiving and their reactions to the painful loss of their loved one. The empirical part of the bachelor's thesis is processed in the form of qualitative research, the method of questioning and the technique of structured interviews with prepared questions. The research population consists of probands who are selected purposefully namely, general nurses, who work at the station of the internal and surgical departments. The research investigation is at the station of the internal and surgical department in the chosen medical facility of the South Bohemian Region. For the research part of the bachelor's thesis, three objectives are set, which are: to find out whether general nurses have experience in communicating with the bereaved. To map whether the general nurses provide enough time for possible questions when communicating with the bereaved and to map whether the general nurses provide intimacy for the bereaved. The results of the research show that general nurses in the internal department communicate with the bereaved more often than general nurses in the surgical department. Differences are also in the length of the conversation and in the description of the environment in which communication with the bereaved takes place. The research investigation points to the fact that the general nurses feel uncomfortable when they communicate with the bereaved. Furthermore, the research shows that the time provided for the questions is not limited, space for questions is provided especially at the end of the interview and the bereaved are encouraged to ask additional questions. Further, general nurses know and have the same procedures for the meeting and describe the same content of the interview with the bereaved. The general nurses also describe identical arrangements for the environment in which communication with the bereaved takes place. Based on the results of the research investigations, a manual is created that contains ten recommendations for general nurses who encounter communication with the bereaved. The manual can serve as a guide to make communication more effective. I believe that the obtained results will make communication with the bereaved in healthcare facilities more effective.
Generation and analysis of double deficient transgenic mice for kallikrein-related peptidase 5 and kallikrein-related peptidase 14

Kallikrein-related peptidases (KLKs) constitute a highly conserved serine protease family. Based on in vitro experiments, KLKs are predicted to play an important role in a number of physiolog- ical and pathophysiological processes. However, their role in vivo remains not fully understood, partially due to a lack of suitable animal models. In this work, we aim to prepare a KLK5 and KLK14 double-deficient mouse model. Both KLK5 and KLK14 were proposed to be involved in epidermal proteolytic networks critical for maintaining skin homeostasis. However, both KLK5 and KLK14 single-deficient mouse models show minimal or no phenotype, likely due to similar substrate specificity resulting in functional compensation. Double-deficient mice cannot be easily obtained by crossing due to localization of the Klk5 and Klk14 genes within the same locus on chromosome 7. We report that KLK5 and KLK14 double-deficient mice were success- fully generated, mediated by transcription activator-like effector nucleases (TALENs) targeting Klk14 by microinjection of TALEN mRNA into KLK5-deficient zygotes. Furthermore, we show that KLK5 and KLK14 double-deficient mice are viable and fertile. We believe that these novel mouse models may serve as a useful experimental tool to study KLK5 and KLK14 in vivo.
Rhythms of restlessness

In my bachelor thesis Rhythms of restlessness I adress, through personal associations and memories, the themes of work ethics and the capitalist-oriented culture of today's society. In an installation made up of a series of objects and drawings embedded within them, I materialize my intimate attitudes or feelings by combining moments from memories with moments from everyday routine activities that are an overlooked part of our everyday "autopilot" functioning. The office chair here acts as a memory, metaphor or symbol of working, and its various manipulations across the installation carry references to my associations associated with this phenomenon. Through this work, I explore the relationships of my immediate surroundings to what it means to function in today's success-oriented rather than happiness-oriented time, and it also serves as a way for me personally to become aware of my own place in the context of my artistic practice. The work builds on my previous projects, in which I have often explored themes of my own intimacy and psychology in the context of functioning in today's world. It is closely related to my most recent work entitled Bedroom Routines, which metaphorically worked through domestic routines and moments, following a stylized character who is my "avatar" in the work.
