Determination of nitrates in vegetables by capillary izotachophoresis

This bachelor thesis deals with determination of nitrates in vegetables employing the capillary isotachophoresis technique. The literature review is devoted to occurrence of nitrates in vegetables, their health risks, the influence of food processing onto nitrate content, commonly used methods of determination as well as the capillary isotachophoresis and its practical application. Experimental part focuses on the application of capillary isotachophoresis for determination of nitrate content in vegetables obtained from conventional agriculture and organic farming.
Capillary isotachophoresis and zone electrophoresis applied to determination of effective charge of polyelectrolytes

Capillary electromigration methods, isotachophoresis (CITP) and zone electrophoresis (CZE) were applied to determination of effective charge of poly-alpha-L-glutamic acid and poly-alpha-L-lysine using both CITP-dedicated device and common CZE apparatus. Combination of CITP and CZE methods in both these types of devices proved to be a suitable platform for determination of effective charge of a wide range of polyelectrolytes – weak and strong, polycations and polyanions.

Theoretical principles of capillary isotachophoresis in moving boundary systems

This contribution provides fundamental theoretical background of ITP in moving-boundary systems. It is based on the approximation of capillary ITP as a 1D process and the general condition of ITP migration, i.e., the existence of a self-sharpening boundary capable to form a stack of zones moving with equal velocities.
Capillary isotachophoresis of anions with electrospray-ionization mass-spectrometric detection

We present ITP-ESI-MS as a powerful tool for selective and sensitive analyses of anionic analytes. Our extended concept of ITP includes besides regular and freeacid ITP also self-maintained ITP in moving boundary systems. Easy theory describes the system properties including their stacking-window diagrams. Simple ESIcompatible electrolyte systems allow flexible tuning with very satisfactory results as demonstrated on the example of analysis of pharmaceuticals in waters.

Water extracts of the dust samples collected in different periods in air-filtration devices in Prague car-traffic tunnels and in Parisian metro station were analyzed both in cationic and anionic capillary isotachophoresis modes.
