Signalization of adenylate cyclase toxin of Bordetella pertussis in macrophages.

Adenylate cyclase toxin (CyaA) is a key virulence factor of Bordetella pertussis, the causative agent of whooping cough. The toxin targets primarily myeloid phagocytes expressing CD11b/CD18 (αMβ2, CR3, Mac-1) and by elevation of cytosolic cAMP levels it paralyses their macropinocytic and opsono-phagocytic functions. Here, we dissected the cAMP-regulated pathway responsible for the block of macrophage macropinocytosis and characterized the capacity of CyaA-treated macrophages to shut- down Akt (protein kinase B, PKB) signaling; that controls nitric oxide (NO) production by macrophages. By using specific activators of protein kinase A (PKA) and for the exchange protein activated by cAMP (Epac), we show that activation of the cAMP effector Epac inhibits macropinocytosis in macrophages. Moreover, upon transfection of macrophages by the constitutively active and dominant negative variants of a downstream effector of Epac, the small GTPase Rap1, inhibition or upregulation of macrophage macropinocytosis was observed, respectively. It was reported previously that the Epac/Rap1 pathway regulates activity of tyrosin phosphatase SHP-1 as well as of protein phosphatase 2 A (PP2A). Camps and courses motivated by art activities

The theoretical part deals with the meaning of the following concepts: camp, courses, animations, painting camps, one of art courses at DDM, which take place every month. The practical part will be based on my own experience with organizing activities, summer painting, courses, suburban camps and art courses. All of these are motivated with their art activities and their analysis. Together with the kid´s animations, we are going to get closer to the Western town called Stříbro and its surroundings from a historical perspective. I was inspired by book about Stříbro and I also combined it with my art aktivity.

Summer Camp Focusing on Floorball

Bachelor thesis deals with the processing of a summer camp project focusing on floorball. It is focused on analysis of the conditions how to organize the camp. The analysis result is used for a proposal how to realize the camp and its program. The main task is to create the best solution for a floorball summer camp on Centre of Sports Activities conditions and implement it to its real form.

Technical Analysis

The main goal of the thesis "Technical analysis" is to analyze the issue of technical analysis itself, more specifically the CAMP model, which deals with optimizing the portfolio, then design a program to calculate it (generate the required results). This analysis corresponds to the structure of the thesis, while in the beginning of the work are mentioned an important theoretical basis. There is described the financial system, financial and capital markets in connection to the Burza cenných papírů Praha, a.s. and other major capital markets. In addition, I discuss the methodology of technical analysis, CAMP model and the source code and the description of Visual Basic. Then I use this methodology in practice and focus on the goal of the work. Finally I analyze the options, benefits as well as weaknesses of the CAMP model and my program in the Czech capital market conditions.

Innovation of a product portfolio using an example of Vysočina region

The main aim of my bachelor thesis is to evaluate the condition of camps in Vysočina region. The next part of thesis is a situation analysis, as STEP and Porter's five forces model analysis focused on one particular camp in the region. The survey was conducted through questionnaires, which were processed after data collection. For clearer overviews of the camp's owners or operators opinions, there were seven guided interviews with them. The result of the collected information was to create an innovative tourist product. The project was designed to involve visitors in a new trend that is currently gaining worldwide popularity. This is called glamping.

The valuation of the company Poděbradka, a.s.

The aim of this thesis is to estimate the market value of czech soft drink producing company Poděbradka, a.s. to the date of 31rd December 2013. The thesis is divided to two logical units -- theoretical and practical. The teoretical unit describes principles, theories and generally recognized ways of corporate valuation in the Czech Republic and in the world. Practical unit continues and expands the theory in its application. Due to the extent of the thesis, just two main yield methods of valuation will be used -- method of discounted cash flow, which is used mainly in Anglo-American valuation praxis and method of capitalized netto earnings, which is mainly used in German-speaking countries. Master thesis ends with terminal valuation statement, which defines the market value of the valuated company.

The Representation of Men with Homosexual Orientation in Czech and Slovak Contemporary Drama

The present thesis focuses on the representations of gays in Czech and Slovak contemporary theater plays. Its aim is to analyze the queer and campy aspects of the representations of gays in the mainstream plays and uncover the principles of creation of the gay character in the structure of dramatic Arts. The results of analysis explain the integrative mode of representation of gays in Czech and Slovak contemporary plays.

Aesthetics of Camp

This thesis deals with the phenomenon of camp and its manifestations in modern western culture. The aim of this thesis is to indentify the signifiant features typical for camp as an aesthetic stance and for camp as an object. Attention is paid to the relationship of the camp and kitsch, and their differences and similarities. The relationship of camp to the high and low art or dandyism is also examined. The main text used for this thesis is an essay "Notes on Camp" by Susan Sontag.

Analýza výkonnosti zahraničních fondů kolektivního investování v ČR

Práce popisuje ukazatele a metody hodnocení fondů kolektivního investování a tyto metody pak aplikuje na 6 vybraných zahraničních akciových podílových fondů nabízených v ČR. Zahrnuty jsou ukazatele a metody zkoumající výnosnost (např. výnosová míra, relativní výnosnost), ukazatele zkoumající riziko (např. směrodatná odchylka), poměrové ukazatele vyplývající z modelu oceňování kapitálových aktiv (CAMP) a některé doplňkové ukazatele (např. procento kladných měsíců, resp. měsíců s kladným výnosem, maximální ztráta za periodu). V práci je také porovnáváno riziko a výnos jednotlivých fondů vzhledem k benchmarku.
