Cell Viability Measurement

This work discusses the viability of cells. It describes the principle of fluorescence, fluorescent methods, fluorescent microskopy and the method of dyeing of the cells with a special kit that contains calcein AM and ethidium homodimer dyes. Further it describes the program for analysis and evaluation of viability on two different cellular cultures.

Tato diplomová práce se zabývá tkáňovým inženýrstvím, zejména tvorbou homogenní, izotropní a planární vrstvy buněk srdečního svalu pomocí dvou technologii:”scaffold-based” a ”scaffold-free”. Nejprve popsaný histologie srdeční stěny, buňky srdečního svalu a buněčné kultury. Následuje popis tkáňového inženýrství, který zahrnuje technologii “cell sheet” a tkáňové inženýrství na bázi scaffoldů. Na konci teoretické části je popsána aplikace tkáňového inženýrství a buněčná vizualizace. Praktická část věnovaná tvorbě planární buněčné vrstvy z kardiomyocytů a fibroblastů s využitím informací z teoretické části.
Cell layer cultivation in the microfluidic system

The theoretical part of this thesis describes the principles of cell culture in vitro and modern in vitro models for endothelial cell layer cultivation. It also describes the principles of confocal and fluorescence microscopy. The practical part is dedicated to working with cell cultures and realization of experiment with cell cultivation in microfluidic system. Cell layer was photographed using laser confocal scanning microscope Leica TCS SP8 X during the entire experiment. For processing and analysis of the obtained images, an algorithm in the software MATLAB was proposed.
Measurement of cultivated cells' viability with fluorescence

This work is aimed to explain to possibilities of measuring viability of cultivated animal cells by staining cells with fluorescent probes. Flourescent probes described in this work are calcein AM and ethidium homodimer-1. There are also mentioned conditions and possibilities of cell cultivation. Additionaly there were created programs for cell´s viabillity detection in Matlab.
