Optimization of miRNA analysis in fine-needle biopsy samples of pancreatic cancer tissue.

Pancreatic cancer (PC) is extremely severe malignant disease with a five-year survival of less than 5%. Currently there is no reliable tool for the diagnosis of PC in its early stages. At the time of clinical symptoms most patients are in an advanced stage of the disease and the treatment does not usually have a significant effect. For these reasons emphasis is gradually shifting to the search for the suitable molecular markers for improvement of the diagnosis and assessment of the survival prognosis with respect to a possibility of surgical treatment. MiRNA represent one of the most promising markers, although, their examination in pancreatic tissue is a complicated process. One of the reasons is the very small amount of the source material coming from a fine needle biopsy. A second cause of problems is the subtle character of the pancreatic tissue resulting in significantly lower yields of molecular genetic analysis when compared to other epithelial tissues. An additional negative factor is heterogeneity of the tissue resulting in disproportionate representation of tumor cells within the sample. A suitable choice of procedures for isolation of nucleic acids (NA) and subsequent analysis including quantification of tumor cells is critical for accurate evaluation of the miRNA levels. The main focus of this thesis has been to study of domestication traits. Identification of genes of domestication traits has a huge potencial for plant breeding. The experimental part has been devoted to the study of pod dehiscence as a domestication trait in pea (Pisum sativum L.). The goal of this work has been to evaluate the expression of candidate genes of pod dehiscence in pea (Pisum sativum L.). For this experiment was used JI64 line pea (Pisum sativum ssp. elatius L.) and JI92 (Afghan cultural type of pea Pisum sativum ssp. sativum L.) and recombinant inbred lines (RILs) arising from reciprocal crossing. Using the method of qRT-PCR (Quantitative Reverse Transcription PCR) have been analyzed candidate genes IDEH, SHAT, bZIP, SPT, NAC and compared with the expression of the reporter gene tubulin.
Molecular and biochemical characterization of serine protease SmSP1 in \kur{Schistosoma mansoni}

SmSP1 is a chimerical serine protease consisted of three domains (cub, LDLa and trypsin-like) and found in Schistosoma mansoni. Its characterization was performed by molecular techniques such as PCR screen, qRT-PCR and RNA interference (RNAi) to gain information about expression profile, level expression and susceptibility to RNAi. Further, protein expression was carried out to gain an antigen for immunization and recombinant for biochemical studies. Results of PCR screen and qRT-PCR suggested possible function of SmSP1 in egg and adult stages but SmSP1 gene was not found susceptible to RNAi in NTS. Recombinant from E. coli was successfully used for immunization. Active recombinant was likely expressed in Pichia pastoris but expression conditions are unstable and expression optimization is necessary.
