Utilization of PHA producing bacteria in bioremediation technologies

The aim of this work is study of utilization of PHA producing bacteria in bioremediation technologies. For this study were used bacteria Pseudomonas putida KT2440 and two isolates from soil contaminated by petroleum - Pseudomonas gessardii (D2) a Pseudomonas fulva (D3). The experimental part describes especially study of feather biodegradation using selected microbial strains. All the tested bacterial strains were capable of feather degradation and utilization as the sole carbon source. During biodegradation experiment, we monitored weight loss of feather, protease and keratinase activity, concentration of bacterial biomass and PHA content as well as pH. The highest biodegradation ability and keratinase activity was observed in Pseudomonas putida. None of tested bacteria accumulated detectable amount of PHA during growth on waste feather, nevertheless, bacterial biomass grown during feather degradation can be used as an inoculum for PHA production on waste frying oil and octanoic acid. Using this experimental setup, high PHA content (54% of cell dry weight) was achiaved in Pseudomonas putida. Another part of the thesis deals with biodegradation of petroleum oil. The highest capability of growth on this carbon source were determined in Pseudomonas fulva.
Production of mcl-PHA by selected bacteria

The aim of this work is to study the possibility of microbial production of polyhydroxyalkanoates with medium chain length (mcl-PHA). For production two bacterial strains were used: Pseudomonas putida and Pseudomonas aeruginosa. Theoretical part of bachelor thesis deals with the biosynthesis of polyhydroxyalkanoates, their material properties and microbial producers. In experimental part selected bacteria were cultivated on different substrates to evaluate their ability to utilize various carbon sources and accumulate mcl-PHA. Content and composition of mcl-PHA accumulated in biomass were analyzed by gas chromatography with FID. Pseudomonas putida produced more mcl-PHA in medium containing canola oil as sole carbon source when compared with Pseudomonas aeruginosa. P. putida produced 2,11% mcl-PHA in medium containing glycerol as carbon source.
Microcalorimetric study of bacterial degradation of oil and its products

This bachelor thesis deals with a study of the oil degradation as one of the possible products of petroleum using bacterial stems Pseudomonas Putida a Pseudomonas Chlororaphis. The work is motivated by the research of usability of a method which has not been used for those purposes so far – calorimetric analysis. Using IČ spektrometry (ATR), calorimetry and UV-VIS spectrometry was studied a degree and a character of interactions. Calorimetric analysis showed that the Pseudomonas Putida has bigger abbility of the degradability than Pseudomonas Chlororaphis. There was also detected that the optimal concentration for more effective degradation of oil fuel is 5 g/l and 20 g/l. By measurement of the absorbance deppending on time was verified that the bigger is the concentration of the sample, the higher is the resulted absorbance. Using UV-VIS analysis was determined a quantity of biomass. Using IČ spectometry in the ATR mode (reduced reflectance) was detected structure of the sample and also verified that the bacterial cultures Pseudomonas Putida and Chlororaphis degradated entire amount of petroleum.
