Focusing system for Ti:Sa laser in the LIBS+LIFS setup
Vémola, Tomáš ; Křupka, Ivan (referee) ; Kaiser, Jozef (advisor)
In this work, the problem of focusing Ti:Sapphire laser beam into LIBS+LIFS interaction chamber is resolved. The task is to design an optical setup with demanded properties. Short theoretical description of the basic principles of atomic spectroscopy, mostly for LIF experiments in LIBS plasma, is also discussed.
Preparation of Xenopus tropicalis transgenic testicular stem cell culture.
Vegrichtová, Markéta ; Tlapáková, Tereza (advisor) ; Drobná Krejčí, Eliška (referee)
Testicular stem cells (TSCs) are relatively accessible potential source of pluripotent cells, which are particularly important for their application in regenerative medicine. Xenopus tropicalis is a useful model organism to study the migration and differentiation potential of stem cells. This amphibian is characteristic by outer fecundation and embryonic development of a great amount of embryos after fertilization. Oocytes and embryos are large enough (about 1 mm) to be suitable for micromanipulation micromanipulations. Laboratory of Developmental Biology, Faculty of Science, Charles University in Prague succeeded in the establishment of a mixed cell culture of TSCs growing on feeder layer of pre- Sertoli cells. This culture was derived from the testes of juvenile Xenopus tropicalis male. In the study of their differentiation potential it was found, that leukemia inhibitory factor (LIF) is the decisive factor allowing rapid proliferation of stem cells and their forming into characteristic colonies. This protein is produced by both types of cells which are present in the culture. The mouse LIF has the same positive effect on the proliferative potential of stem cells, which points at the evolutionary conservation of metabolic pathways associated with the maintenance of the stemness. RT-PCR analysis...
Velocity measurements on continuous micro water jet using fluorescent PMMA seeding particles
Zeleňák, Michal ; Ščučka, Jiří ; Souček, Kamil
The paper deals with application of optical diagnostic techniques for velocity\nmeasurements of continuous micro water jet. The combination of Particle image\nvelocimetry, Particle tracking velocimetry, Laser induced fluorescence and\nShadowgraphy methods have been applied together with the use of fluorescent\nPMMA seeding particles. An experimental closed water circuit was designed with\na low-pressure water pump, and a series of experiments was conducted to\nmeasure the velocity of seeding particles in the water jet flow below the diamond\nmicro-nozzle outlet. The basic principles of the methods used in experiments are\nalso described.

PIV and LIF study of flow and thermal fields of twine plumes in water
Broučková, Zuzana ; Trávníček, Zdeněk
Flow and thermal fields of a pair of plane plumes in water are investigated using PIV and LIF experiments. The plumes are generated from thermal line sources (electrically heated cylinders, diameter D = 1.21 mm). Either continuous or pulsating heating were used with the same heating input power. For a continuous heating, low frequency oscillations were identified and the natural frequency was evaluated as 0.5 Hz. Pulsating heating was used at frequency of 0.25 Hz. The maximum time-mean velocity magnitude at the continuous and pulsating heating were commensurable, approximately 0.007 m/s. Temperature fields reasonably agree with these findings.

Measurement and analysis of abrasive particles velocities in AWJ
Zeleňák, Michal ; Foldyna, Josef ; Martinec, Petr ; Cárach, J. ; Gurková, Lucie ; Nováková, Daria
Abrasive water jet (AWJ) is a progressive technology commonly used in many industrial applications. Potential of this technology is high and research in the field of optimal geometry design of an injection system has not been completed yet. New designs of the injection cutting head based on numerical simulation of the flow provide important information about processes occurring during the formation of AWJ. Detailed knowledge of velocities of abrasive particles during the abrasive water jet generation is vital for the verification of the numerical model. The aim of the experimental work was to test the applicability of the LIF (Laser Inducted Fluorescence) technique combined with the PTV (Particle Tracking Velocity) processing algorithms to analyze particle velocities at the output of the focusing tube. The combination of commonly used water pressures with abrasive mass flow rate was tested and the obtained results of abrasive particles velocities are presented in this paper.

MARTÍNEK, Petr
The study analyse presence and distribution of mutation in the LIF and IL-11 genes. The alterations were studied in peripheral blood samples by temperature gradient gel electrophoresis and sequencing.
Interakce UV záření s povrchovým nábojem v N2 povrchovém DBD
De Benedictis, S. ; Ambrico, P. F. ; Dilecce, G. ; Šimek, Milan
In this paper we report on space and time resolved spectroscopic measurements on Masuda type surface DBD fed by N2. In our configuration the discharge presents some defects that lead to the formation of some bright spot inside a regular discharge region formed by streamers. We have carried out an optical investigation of both the regular discharge region and the bright spot one with the aim to stress the aspects of photo emission contribution in the dynamics of microdischarge. We have carried out also spectroscopic space and time resolved investigation of discharge emission when UV photons by a pulsed laser interact with the dielectric surface of DBD. There is a similarity between the time evolution of emission from bright spots and that produced by laser photons.
