Denisa Janů Supervisor: PharmDr. Ondřej Janďourek, Ph.D. Title of diploma thesis: Current possibillities of laboratory diagnostics of EBV Background: The aim of this diploma thesis was to get acquainted with the issue of Epstein-Barr virus infection and its laboratory diagnostics. Other aim was to divide the patients into groups according to the serological results, then evaluate the prevalence of EBV and evaluate the incidence of primary EBV infection in the tested group of patients for the years 2019-2022. Methods: Chemiluminescence immunoassay and indirect immunofluorescence were used to determine specific antibodies against EBV. In some cases, the examination was completed by determining the avidity of anti-VCA IgG antibodies and the microblot array method. Real-time PCR was used to detect EBV DNA. Microsoft Excel 2016 was used to process data and to create tables and graphs. Incidence of hyperlipoproteinemia and atherosclerotic changes in long-term survivors of Hodgkin lymphoma in childhood or adolescence

The survival of children and adolescents with Hodgkin lymphoma has improved dramatically over the last decades, but even young adult long-term survivors are at increased risk of developing multiple cardiovascular late sequelae of previous treatment. We evaluated the presence of modifiable cardiovascular risk factors (hypertension, hyperlipoproteinemia, hyperinsulinemia, obesity) in young adult Hodgkin lymphoma survivors, who were disease- free for a minimum of 10 years after the end of their treatment, and in age- and gender- matched healthy volunteers. We assumed higher prevalence of modifiable cardiovascular risk factors together with earlier development of atherosclerotic changes in this specific population. All survivors and volunteers uderwent physical examination, duplex ultrasonography of both common carotid arteries, all participants completed questionnaire on their eating habits and physical activities We performed biochemical analyses, including lipid profile, fasting glucose and insulin levels, markers of inflammation, endothelial dysfunction and oxidative stress. Nursing care of the patient with lymphoma

(v AJ) I chose the topic for my bachelor thesis: Nursing care of a patient with lymphoma. I consider this topic as a very actual because the occurence of lymphoma is increasing (In: Linkos, 2019). Lymphomas refer to tumors which originate from lymphocytes. The theoretical part deals with the issue of lymphomas. I focus mainly on their diagnostics and treatment. And I deal with nursing care and the Lymphoma Help patient organization. Before I started to work on my bachelor thesis I had made a research in the National Medical Library. Furthermore, I used information from the databases: BMC, PubMed, EBSCOhost and Medline. I also used therapeutic recommendations - Cooperative lymphoma groups. Methology: I chose a case study of a patient with B-cell lymphoma from large cells, diffuse for the preparation of this thesis. I used Marjory Gordon's functional health model. Nursing standard procedures, potential risks of nursing care and their prevention are mentioned and described in a specific case. Objective:. (v AJ) I chose the education of a patient with lymphoma as the topic for my bachelor thesis. I consider this topic very actual because the occurence of lymphoma is increasing (In: Linkos,2019). Lymphomas are referred as lymphocyte-derived tumors. I deal with the problem of lymphomas in the theoretical part. I mainly focus at diagnostic and treatment. I had started to prepare this bachelor thesis after the background research in the National Medical Library. I also drew the information from databases: BMČ, PubMed, EBSCOhost a Medline. I also used treatment recommendations - Cooperative Lymphoma Groups. Methology: I chose a case study of a patient with B-cell lymphoma from large cells, diffuse. Structural study of the DCL-1 receptor using mass spectrometry

DCL-1 (CD302) is a type I transmembrane C-type lectin receptor, which is expressed on monocytes, macrophages, granulocytes and dendritic cells. However, its extracellular domain lacks the amino acids motives essential for carbohydrate binding in the presence of calcium ions, suggesting that it does not have the classic binding capacity found in other C-type lectin receptors such as the mannose receptor. No exogenous or endogenous ligands have been identified yet, though. Due to internal colocalization with F-actin we can assume, that this unconventional lectin receptor plays a role not only in endocytosis and phagocytosis but also in the cell adhesion and migration. The receptor DCL-1 was first identified as a genetic fusion partner of human DEC-205 multilectin receptor in Hodgkin's lymphoma cell lines. The experimental part of this thesis deals with the characterization of disulfide bonds and data acquisition for validation of DCL-1 crystal structure. First the production and refolding conditions were optimized to obtain the highest amount of DCL-1 protein, precisely its extracellular domain. These optimal conditions were used to prepare the protein for in-gel digestion using specific endopeptidases in the presence of cystamine followed by LC-MS analysis.
