Optimization of Hericium erinaceus Cultivation and Subsequent Fermentation with Probiotics

The focus of this diploma thesis is to optimize cultivation of the hericium erinaceus mushroom on cereal substrates with subsequent fermentation with probiotic bacteria. Theoretical part of this thesis focuses on description of the mushroom hericium erinaceus and its nutritional value, biological properties, other uses and cultivation. Furthermore, an overview of the nutritional value of the used cereal substrates is provided, along with the characterization of probiotics, their biological effects, and metabolism. Finally, a chapter dedicated to the description of the traditional koji fermentation process was included. In the experimental part, the cultivation of hericium erinaceus was optimized on seven cereal substrates – barley groats, cornmeal, wheat bran, buckwheat, amaranth, white and red quinoa. Using spectrophotometric methods, the content of water soluble proteins, total carbohydrates, total phenolic compounds, flavonoids, and antioxidant activity was determined in the cereal substrates themselves and substrates enriched with mycelium. Furthermore, the content of fatty acids was determined in the samples using the gass chromatography method and lipophilic compounds were analyzed using high performance liquid chromatography. Cereal substrates enriched with mycelium showed increased antioxidant activity and concentrations of water soluble proteins, phenolic compounds, flavonoids, and lipids. Moreover, elevated concentrations of tocopherols, ubiquinone, and ergosterol were detected through high performance liquid chromatography analysis. Substrates with abundant mycelial growth, such as barley groats, cornmeal, red quinoa, and amaranth, were selected for fermentation with probiotic bacteria Lactobacillus rhamnosus. After the fermentation process, spectrophotometric methods were repeated, and the concentration of lactic acid was determined using high performance liquid chromatography. Samples with probiotics showed a slight increase in their antioxidant activity, concentration of phenolic compounds, flavonoids, and water soluble proteins. The concentration of lactic acid in control samples and samples with probiotics was almost unchanged. The obtained results from this study suggest that cereals enriched with mycelium and the fermentation product of probiotics could be utilized in the development of dietary supplements or functional foods.
Structural analysis of polysaccharides from mushroom hericium Erinaceus mftccb 134

The aim of this research was the structural characterisation of polysaccharides isolated from fruiting bodies of the cultivating medicinal mushroom Hericium erinaceus MFTCCB 134. Cold and hot water extracts (CWE, HWE) were obtained and purified, and followed by alkali extraction from the reminder solids. The composition and structure of the fractions were analysed by FTIR spectroscopy, and neutral sugars/linkage analyses were determined by GC/FID and GC/MS, respectively. Galactose, fucose and glucose were found in purified CWE,\nwhile glucose predominated in purified HWE.
Active preparatives based on koji fermentation and medical mushrooms extracts

This bachelor's thesis deals with the optimization of the growth of the rare species fungus of the hedgehog mushroom on cereal substrates for the purpose of the subsequent to cultivation of lactic acid bacteria. The theoretical part is focused on the description of the rare fungus species and the use of the cereals as substrate and the characterization of their active substances. It also deals with lactic fermentation and its use. In the experimental part, the preparation of the cereal substrate for the cultivation of the hedgehog mushroom was optimized. The method of inoculation of the cereals substrate with liquid mycelium in combination with plate mycelium was chosen as the most applicable. In this combination, the substrate does not dry out and grows evenly from both sides of the substrate. The composition of the cereal substrate with grown mycelium was characterized (antioxidant activity, carbohydrates, proteins). Finally, the substrate was inoculated with lactic bacteria. The lactic bacteria added nutritional value to the product. Fermentation products (antioxidant activity, carbohydrates, proteins) were characterized, and bacterial viability was analysed.
Substrate processing for cultivation of Hericium erinaceus with different supplements and heat treatments

The aim of the thesis was to determine if the substrate treatment with temperature lower than 100°C could replace the sterilization at 120°C. Another task was to compare different types of substrates and supplements. For the substrate treatment were selected temperatures of 90°C for 48 hours, 90°C for 20 hours, 121°C for 2 hours, 80°C for 20 hours and 70°C for 24 hours. The treatment with temperature lower than 100°C could replace the sterilization at 120°C. In the experiments were used substrates compound of beech sawdust or straw pellets supplemented by wheat bran or wheat groats. The research was also focused to influence of substrate fermentation on growth characteristics of Hericium erinaceus. The most suitable substrate for mycelium growth and subsequent fructification of Hericium erinaceus were beech sawdust. Faster mycelial growth and higher yield of fruiting body were achieved in the case of supplemented substrate of sawdust beech by wheat bran or wheat groats. There were observed no effects of fermentation substrates on mycelium growth of Hericium erinaceus. Another part of the research was dedicated to mycelium growth and fructification of Hericium coralloides. The best substrate for Hericium coralloides is beech sawdust without supplements.
