Výskyt a vyhodnocení patogenů hrachu rolního a hrachu setého na lokalitě Moravské Budějovice

Pea (Pisum sativum L.) is the most cultivated legume in the Czech Republic. The quantity and quality of harvested seeds are influenced by determining, limiting and reducing production factors. The focus of the thesis was on the reducing factor, namely the occurrence of pathogens in the crop of sown pea (Pisum sativum subsp. sativum) and field pea (Pisum sativum subsp. arvense). The pathogens evaluated were Botrytis cinerea, Fusarium solani f.sp. pisi, Fusarium oxysporum and Erysiphe pisi var. pisi. Part of the experiment took place in locations Moravské Budějovice, Jaroměřice nad Rokytnou, and Zálesí in 2022; the other part was conducted in the laboratory of the Crop Science, Breeding and Plant Medicine at Mendel University. In the field trials were the varieties Arkta and Salamanca. And in the laboratory trials were the varieties Astronaute, Arkta, Dora, Impuls, Salamanca and Turnia. In the laboratory experiments with the pathogens F. solani f.sp. pisi and F. oxysporum, the variety Salamanca had the lowest average degree of infestation and Impuls the highest. Sown pea had a lower index of infection by the pathogen B. cinerea compared to field pea. Cantus had no significant effect on the disease incidence. For the pathogen E. pisi, the infestation of field pea was significantly higher.

This bachelor thesis describes the protection of the possibility of protection against gray grape varieties in organic and integrated production. In the first chapter, the bachelor thesis deals with the characteristics of Botrytis cinerea, we find the grape biology of the grape, it describes the mushroom originator, the symptoms of the attack and the noble rot. The next chapter deals with other rotations that may appear in the vine during vegetation. The third chapter describes the impact of decay on grape quality. In the last chapter the thesis deals with protection systems in organic and integrated production, against Botrytis cinerea.

Resume The aim of the bachelor thesis was to study literary sources of gray mold (graygrape rot). The thesis describes the general characteristics of fungi, taxonomic classification, biology and gray mold symptoms. Furthermore, the options of protection against the gray mold, summarization o fthe defense reactions of the plant after the attack and how the gray mold effects the plant and grapes at all stages. Part of the work payattention to changes in the composition of the wine, which is produced from infected grapes both in the stage of the disease and in the stage of noble Botrytis cinerea. Last but not least, the thesis deals with territories with suitable climatic conditions for the development of Botrytis cinerea and the unique wines from it.

Bachelor thesis deals with the possibilities of protection against gray grape rot in conditions of ecological viticulture. In the first chapter we introduced the gray mold (Botrytis cinerea). Description of sponge fungi, mushroom biology and signs of Botrytis cinerea mold fungal attack. The life cycle of fungi and the conditions in which it occurs is described in detail. Another chapter deals with other rot, which during the vegetation attack the vines. For these fungi, we also partly described their biology and the possibilities of direct and indirect protection. In the third chapter we focused on the effects of decay on grape quality. It has been explained how grapes affected by rot can affect the aroma and the resulting quality of the wine produced, how they influence the enzymatic activity in the grapes and finally how the rot influences the acids in the grapes. The fourth chapter deals with the possibilities of direct and indirect protection in organic viticulture against gray rot. In indirect protection, all possible ways to protect vines from attack were dismantled. In direct protection, we introduced environmental protection. We have described the products that increase the resistance of the grapevine, as well as the preparations that act directly against the gray mold. In the last chapter, appropriate measures have been proposed to combat gray mold.
Vyhodnocení výskytu patogenů kdouloně, dřínu a maliniku na lokalitě Žabčice

This thesis is focused on the occurrence of pathogens on the school farm Žabčice. Pathogens have been evaluated with quince varieties 'Champion', 'Ironda' and 'Hruškovitá'. This mainly involved evaluation of the bacteria Erwinia amylovora and Monilia ssp. Based on the results was done by comparison with the previous thesis and it was concluded. For raspberry incidence is assessed fungus Botrytis cinerea, where due to weather conditions prevent the massive expansion of the next fruit. We found there was probably also a raspberry ringspot virosis, which was intended only as a symptom based on these symptoms was determined by Atlas. At the last review, dogwood is due to weather conditions and high temperatures in 2015 appeared curling sheets. At the end of the vegetation was reflected leaf spot, which however didn´t affect the coming harvest.
Methodology of detection Penicillium expansum, Monilia frutigena, Botrytis cinerea and Neofabraea alba using multiplex PCR

Multiplex PCR for rapid and reliable detection of species causing rotting apples: Penicillium expansum, Monilia fructigena, Botrytis cinerea and Neofabraea alba is described here. Reaction is optimized for using in a common molecular laboratory. Described method allows both detectino of the pathogens from pure mycological cultrures, and their detection directly from the rot apple tissues. Cultivation of the micromycethes on cellophane surface is also described. References for mycological determination literature are listed, and can be helpful for determination in case of negative results of PCR.
