Comaprison of effectivness of quarternary amonium salts on different microbes.

We tried to compare antimicrobial effectiveness of Benzalkonium bromide with various lengths of alkyl chains fixed in effective cationic section of the molecule in this dissertation. We worked with homologues that had the alkyl chain length 12, 14 and 16 carbon atoms and also with the mixture of homologues mixed in ratio 1 : 1 : 1. We were comparing the antimicrobial effectiveness against the members of G+, G- bacteria, torula, fungi and spores. We used agents from the Czech collection of microorganisms of the Natural science faculty of the Masaryk University Brno. Namely they are: Staphylococcus aureus, Escherichia coli, Proteus vulgaris, Serratia rubidaea, Pseudomonas aeruginosa, Candida albicans, Aspergillus niger and Bacillus subtilis. We determined MIC, MBC, MBC/N and MBC-B by the help of suspense micromethod for all strains except the fungi and spores members. Concerning the Aspergillus niger and Bacillus subtilis we determined only the MBC. This was executed by the standard suspension method. The results of effective concentrations of particular tests are mentioned in g/100ml. The antimicrobial effectiveness in the MIC test was nearly identical for all tested preparations. Part I Diploma thesis Charles University in Prague, Faculty Of Pharmacy in Hradec Králové Study programme: Pharmacy Background: The aim of the work is to sum up findings about quorum sensing in Candida albicans communication. The quorum sensing effects were studied on farnesol on model of conversion of blastospores into germ tubes in C. albicans. Methods: Four strains of C. albicans were tested, two of them were from American Type Culture Collection and two were obtained from the pacients suffering from vulvovaginal candidosis. The method of germ tube test was standardized to evaluate the farnesol effects on germ tube induction including the possibility for testing effects of preincubation in presence or absence of farnesol. Results: There was no significant difference in the production of germ tubes between Sabouraud glucose agar and YPD medium used for the preparation of inoculum. The chemically defined medium NYP provided more consistent results than bovine serum in terms of germ tube induction if the cell density of a yeast was 106 cfu/ml. Marcela Houdková Predisposing factors of allergies XIII. Thesis Charles University in Prague, Faculty of Pharmacy in Hradec Kralové. Field of study : Pharmaceutical Background Thesis objective was to evaluate the impact of possible risk factors for the development of allergic disease. Methods Comparison of data collected from Questionnaires from years 2002 and 2005, involving patiens and healthy children born in years 1989 -1990 and 1992-1993. Results We found out statistically significant outcomes by the following risk factors: abdominal delivery; child's birth weight more than 5,5 kg; a child's first immediately after birth, after child's treatment, during the first six hours and after one day; the meat consumption 4 - 6 times per week and sometimes in two years of age, 1 - 3 times a week for the transfer of children from kindergarten to school, and the meat consumption 4 - 6 times a week and 1 - 3 times a week for the last two years. Also, we have approved statistically significant value for the period of exclusive breast- feeding and breast-feeding period with the trimming. Tereza Petlánová Consultant: RNDr. Klára Konečná, Ph.D. Title of thesis: Study of the interaction of newly synthesized compounds with bacterial agents. Background: The aim of this thesis was to test antibiotic activity of substances produced by the Department of Inorganic and Organic Chemistry, Faculty of Pharmacy of Charles University in Hradec Králové. Methods: Substances were tested by using microdilution broth method on eight strains of bacteria: Staphylococcus aureus, Staphylococcus aureus methicilin resistant, Staphylococcus epidermidis, Enterococcus sp., Escherichia coli, Klebsiella pneumoniae, Klebsiella pneumoniae ESBL positive and Pseudomonas aeruginosa. Results: Substances were divided into five groups according to associated structural features. The most effective was the group of salicilanilid derivates. In this group, the bacterial effect of all 28 tested substances were shown. Conclusion: The antibacterial effect was demonstrated in 50 from the total number of 73 tested compounds. The most susceptible strains were Staphylococcus aureus, Staphylococcus aureus methicilin resistant, Enterococcus sp. and Staphylococcus epidermidis.
