Application of liquid chromatography in the analysis of biological material for clinical research

RNDr. Lenka Kujovská Krčmová, Ph.D. Title of doctoral thesis: Application of liquid chromatography in the analysis of biological material for clinical research The dissertation focuses on the use of liquid chromatography for clinical research. It is divided into two main sections, Theoretical part and Commentary on selected published articles. The theoretical part describes the application of high-performance liquid chromatography in practice. Key parameters for the development of LC methods and trends in current bioanalytical practice are discussed. Furthermore, the sample collection and preparation of different biological matrices is explained, including all the obstacles and pitfalls that need to be taken into account. Modern trends are also briefly mentioned. The chapter on sample preparation also includes a section on derivatization and its use in the analysis of platinum derivatives, which is related to the commentary on the published work. The main part of the thesis deals with commentary on four published articles. The first presents a review article dealing with the use of microextraction in pipette tips in clinical and forensic toxicology....

A Comparison of the Determination 25(OH)D in the Human Serum by Methods UHPLC-MS/MS and CMIA Abbott Architect

In this rigorous work UHPLC-MS/MS and Abbott Architect CMIA methods for determination of 25(OH)D (calcidiol) in human serum were compared. Human serum samples were firstly analysed by the principle of CMIA and subsequently identical samples were processed by UHPLC-MS/MS method that was developed and validated in Research Laboratory of III. Internal Gerontometabolic Clinic of University Hospital in Hradec Králové for 25(OH)D3 and 25(OH)D2.clinical monitoring. Comparison of these methods was performed using the statistical software NCSS 2007 (Number Cruncher Statitistical Software, Kaysville, Utah, USA). Descriptive statistical methods described basic properties of both result sets (mean, median, standard deviation, etc.). The evaluation was tested using pair T-tests to assess the diversity or consensus between the two datasets. Correlation of the two sets of values was investigated by regression analysis. Based on statistical analysis of the results we can conclude that the values obtained by UHPLC-MS/MS and CMIA Abbott Architect methods are statistically significantly different (p = 0.0000), but the correlation is very good (r = 0,8832). Therefore the both methods are well comparable and the results can be converted to each other.

Title of Doctoral Thesis Analysis of biologicaly active substances using liquid chromatography Presented dissertation thesis is dedicated to the development of methods using liquid chromatography and involves thin-layer chromatography, high performance liquid chromatography and up-to date modification - ultra-high performance liquid chromatography. The paper deals with isolation techniques of tested substances from biological matrix or pharmaceutical preparation. All developed methods are confirmed by means of validation procedures which include primarily precision, accuracy, selectivity linearity and sensitivity. Thesis is divided into two thematic parts. In the first one derivatives of amphetamine are evaluated in biological material. The second part deals with evaluation of pharmaceutical preparations. Firstly the Theoretical part is devoted to theoretical principles of chromatography, it is focused to liquid chromatography, especially HPLC, TLC, UHPLC.
