Animal Models of Intrauterine Infections

In mammals, embryonic and fetal development takes place in the mother's uterus under sterile conditions. The embryo, and later the fetus, is connected to the placenta by the umbilical cord and floats in amniotic fluid in the amniotic cavity lined by the amniotic membrane. Preterm birth is associated with decreased weight of newborns and increased morbidity and mortality. Intrauterine infections are one of the leading causes of preterm delivery. Various etiological agents, such as viruses, bacteria, yeasts and protozoa, can cause these infections. These infections induce expression of inflammatory mediators, which participate in triggering of preterm birth. One of these mediators are cytokines. The bachelor thesis is based on a literature review dealing with animal models of prenatal infections, which are exploited in the study of regulatory mechanisms of preterm births or eventually in the study of experimental manipulation of these mechanisms. Various infectious agents and their parts, such as LPS, and the routes of administration are compared. In addition, the advantages and disadvantages of different animal models are discussed with respect to the type of placentation, duration of gestation, animal weight and litter size. It focuses primarily on celiac sprue, describes the disease, its clinical manifestations, symptoms, complications, methods of investigation, diagnosis and treatment. Part of the thesis deals with relationship of gluten to other diseases such as gluten sensitivity or autism.
Genetic mapping in Xenopus

The diploid amphibian Xenopus tropicalis represents a significant model organism for studies of early development, genes function and evolution. Such techniques as gynogenesis, injection of morpholino antisense oligonucleotide into fertilized eggs or transgenesis were established. In the recent ten years, many efforts have been made to complete the sequence information. X. tropicalis genome has been sequenced but the completion of its assembly only on the basis of sequence data has been impossible. Therefore, our first work was focused on one of approaches for a genome completing- genetic mapping. First of all, the genetic map of Xenopus tropicalis was established pursuant linkage and physical positions of markers. Since the map contained gaps, we developed a new method for genetic mapping based on the next generation sequencing of laser microdissected arm. Using Illumina next generation sequencing of fifteen copies of a short arm of chromosome 7, we obtained new insights into its genome by localizing previously unmapped genes and scaffolds as well as recognizing mislocalized portions of the genome assembly. This was the first time laser microdissection and sequencing of specific chromosomal regions has been used for the purpose of genome mapping. These data were also used in the evolution study of...
Cell death-regulating micro RNAs and their role in the development and pathological processes.

MicroRNAs are small protein non-coding, ~ 22 nucleotides long dsRNAs. Their main task is suppression of gene expression via removal/destabilization of mRNA or its targeting to degradation. These small molecules play an important role in the regulation of many cellular processes and have been found to affect expression of more than 30% of human genes. Among the processes affected or regulated by miRNAa belongs also programmed cell death. Although this work is mainly focused on the analysis and characterization a role of distinct miRNAs in the regulation of apoptotic cell death, miRNAs can also participate in the regulation of autophagic cell death or programmed necrosis. MiRNA can enhance cellular sensitivity to apoptosis by suppressing the expression of death receptor genes, but can also drive cells to apoptosis by regulating expression of anti-apoptotic protein Bcl-2. In many different organisms were already discovered and described thausends of micro RNAs anddozens of them participate in the regulation of cell death.
