Application of Private Label Food Market in Selected Company

The topic of this diploma thesis is use of private foodstuff labels in a chosen Czech market marketplace between 2009 and 2014. First part of the diploma thesis deals with private labels portion with respect to overall goods volume as well as with progress of overall product volume in particular product categories. Second part of the diploma thesis deals with progress of private lablel product price and price of some competitive products subsequently the topic of this diploma thesis is an intensity of private labels price relevance on venality. Third part of the diploma thesis deals with depreciation of ratio indicators on the the particular marketsale.
Micropropagation of Satureja Montana L.

Winter savory (Satureja montana L.) is a plant occurring in the Mediterranean and the Black Sea region. For its high content of essential oils is widely used in culinary and traditional medicine. Savory is propagated generative and vegetative. Seeds germinate slowly, and therefore, the vegetative propagation is faster, especially in in vitro conditions may be propagated a large amount of plant material in a short time. The aim of this thesis was to investigate the growth effect of cytokinins on regeneration and growth plants from nodal segments of S. montana L. Nodal segments were cultured on six variants of medium supplemented with 0.5; 1 and 1.5 mg/l 6-benzylaminopurine (BAP) and 0.5; 1 and 1.5 mg/l kinetin (KIN). Variant without growth regulators served as a control. Culture conditions were 25/20 °C day/night, light intensity 3000 lx and daily period of 16/8 hours. After 6 weeks of measurements were evaluated parameters such as: number of shoots, plant height, the number of newly created nodes and the number and length of roots. In respect of the coefficient of micropropagation (i.e. the number of newly created nodes on the plant) was statistically significantly best variant medium supplemented with 1.5 mg/l KIN where on average create 8.88 +/- 2.87 nodes. At the height of the plant which was highest in the variant with a concentration 1.5 mg/l KIN (2.27 +/- 1.33) and the number of shoots with highest average value in a concentration 1 mg/l BAP (2.87 +/- 1.19) encore of growth regulators were not statistically proven. High concentrations of BAP and KIN inhibited the induction of roots and from variants transferred to ex vitro conditions were regenerated 46 % plants. Adding BAP and KIN into the medium has an influence on plant regeneration of Satureja montana L.
Micropropagation of Lippia dulcis Trev.

Lippia dulcis Trev. (Verbenacea) is a perennial herb with a creeping stem from Central America. It is known for its healing properties for problems with the respiratory or digestive system, but also its sweet taste, which causes terpene hernandulcin because of which the leaves of Lippia dulcis Trev. are 1,000 times sweeter than sugar. Generatively, the plant reproduces by seeds and vegetatively by cuttings. The main objective of this study was to investigate the influence of cytokinins kinetin (KIN) and 6-benzylaminopurine (BAP) with the addition of auxin indole-3-acetic acid (IAA) on the growth and regeneration of plants from nodal segments Lippia dulcis Trev. in vitro conditions. Nodal segments were cultured on 6-variants of MS medium supplemented with growth regulators at concentrations of 0.5; 0.75 and 1.0 mg / l BAP and KIN with combination 0.5 mg / l IAA and one variant of pure MS medium as a control. Nodal explants were cultured for 35 days (the time of subculturing) and measured every 7 days. The following parameters were evaluated: regeneration, number of shoots, length of shoots, number of nodes, number and length of roots. The measurement results show that a combination of growth regulators auxin-cytokinin have no positive influence on the growth and regeneration Lippia dulcis Trev. in vitro conditions. The best regeneration (100%), growth (on an average length of the shoots 3.05 +- 1.42) and the highest number of newly created points (5.79 +- 2.4) occurred at the explants cultured on a clean MS medium without addition of growth regulators. Among the variant with pure MS and media variants with the addition of growth regulators are statistically significat defferences, but without a favorable influence on the development of nodal explants.
Evaluation of managers functions

The subject of this thesis is to evaluate various managerial positions in selected executives working in different management levels (a line, middle and senior manager) at Stock Božkov Ltd. The theoretical part focuses on terms of management, the manager, styles and roles of managerial work. The following is a key part of the literature review and characterization of sequential and parallel managerial functions. The practical part focuses on the actual survey, which was conducted by observing the daily work of selected managers and a personal interview with each of them. In the end, the work compares ten defined theoretical assumptions from literature the identified practical results. It was found that five theoretical assumptions (no. 1, 4, 7, 8 and 10) was confirmed in practice, and five (no. 2, 3, 5, 6 and 9) was not confirmed. Furthermore, the results indicate that the proportion of managerial functions and activities performed within these functions depends on the level of management. Finally, the proposed recommendations executives selected one that would contribute to improving utilization managerial functions designed for different types of executives. Line leaders were advised to delegate more tasks to the department shift supervisor, head of middle management was advised to focus on department performance and its completing and work more with subordinates and senior managers was raised in support of his decision to find qualified and competent people to key positions of line managers, which are suppressed due to its functions and duties.
The effect of oviposition on egg quality at the beginning and end of the laying cycle

The egg quality is affected by many factors, among which various interactions may occur. The aim of the diploma thesis was to assess changes in the quality of eggs laid in the morning, midday, and in the evening at the beginning and the end of the laying cycle in six laying hybrids: Bovans Brown, Bovans Sperwer, Isa Sussex, Moravia Barred, Moravia BSL and Dekalb White, each individually housed in cages. The experiment was realized at the beginning of lay, between 19 and 26 weeks of age and at the end of lay betwen 64 and 70 weeks of age. During the experiment, were conected eggs at 6 a.m., 9 a.m., 12 p.m. and 5 p.m. and were recorded daily. In total, 300 eggs from each genotype and of each period were evaluated. At the beginning of the laying cycle, the interactions of oviposition time and genotype (P 0.03), were observed in the yolk weight (P 0.004) and the color of the yolk. The highest weight yolk had the eggs laid by white-egg hybrid Dekalb White early in the morning (13.79 g at 6 a.m.) and during the day the weight of yolk decreased. However, the yolk weight of brown-egg Bovan Brown hens increased until 9 a.m. and then decreased again. The darkest yolk was laid by genotype Bovans Brown at 12 p.m. (8.0 g), while the lightest yolk was laid by Dekalb White at 12 p. m. (3.00). At the end of the laying cycle, interactions of oviposition time and genotype (P 0.028) were observed in the albumen percentage. The highest values had hybrid Moravia BSL with eggs laid in 12 a.m., 5 p.m. 64.23 %, 63.33 %), and the lowest percentage had hens of Bovans Sperwer at 9 a.m. (58.19 %). Oviposition time significantly (P 0.003) affected only the strength of the shell. At the beginning of the laying cycle, we recorded higher strength at 9.00 a.m. (54.89 g.cm-2) compared to the eggs collected at 6 a.m. and later (41.39 a 24.58 g.cm-2). The genotype (P 0.005) affected most of the parameters of quality.
Effect of breed and storage on goat milk quality

The aim of this diploma thesis was in theoretical part to summarise current knowledge about composition, physico-chemical, technological and sensory properties and cryostability of goat milk including a comparison of this characteristics for dairy goat breeds which are used for industrial and farm dairy production. Goat milk has high nutritional and therapeutic value. It is suitable for dairy industry processing as well as cow milk. In the Czech Republic, where is traditional cow milk consumption, the interest of consumers in goat milk products increased recently. The main Czech dairy goat breeds are White and Brown-Shorthaired Goats. In the practical part of diploma thesis were compared the 2 main Czech dairy goat breeds, White and Brown-Shorthaired Goats, with a rare breed Girgentana Goat. Cryostability of goat milk was observed for 16 weeks (0, 1, 2, 3, 4, 8, 12 and 16 weeks) for White and Brown-Shorthaired breed. Characterized parameters were main components (total solid, non-fat solid, fat, lactose, crude proteins and casein), minor components (citric acid, urea, free fatty acids and fatty acid profile), physico-chemical properties (density, freezing point and titratable acidity), technological properties (cheeseability, fermentability and thermostability) and for fresh milk also sensory identity and acceptability. It was found that milk from Girgentana Goats contains the significantly (p < 0.05) highest amount of total solid, fat and lactose. The highest amount of crude proteins (p < 0.05) was on the contrary observed in Brown-Shorthaired Goat milk H1. These two samples had also the best cheeseability (p < 0.05) but the worst sensory acceptability because of their strong goat flavour and taste. During the storage test of White and Brown-Shorthaired Goat milk samples was found significantly (p < 0.05) negative influence of storage in frozen state only on cheeseability. This could be explain by the negative changes in the conformatin of proteins, especially casein fraction.
Caseinomacropeptide biological properties

The aim of the bacherol thesis was to summarize the current knowledge about biological properties of caseinomacropeptide. Caseinomacropeptide is hydrophilic, mostly glycosylated peptide which commes from kappa-casein during its chymosine proteolysis. Polypeptide has a lot of interesting functional and biological properties thanks which has a wide potential for food industry and development of new products with added value. Its valuable physico-chemical properties include surface activity (emulsifying and foaming properties), gel-forming and adsorption properties, thermo- and acidobasic stability. From the biological effects may be mentioned antibacterial, anticariogenic, antiviral, dietary, effects on the gastrointestinal tract, hypoallergenic and immunomodulatory properties and prebiotic. In many studies was observed that the higher biological activity of caseinomacropeptide (native or hydrolysed forms) is connected with its glycosylation. One of the most important parameters is also sialic acid content. Good bioactive properties also demonstrated polypeptide conjugates with galactooligosaccharides. Antibacterial effect of caseinomacropeptide was recorded for example against pathogenic strains of Escherichia coli ETEC K88, Salmonella enterica subsp. enterica serotyp typhimurium CECT 443 a Listeria monocytogenes CECT 935. Polypeptide acted as prebioticum for Bifidobacterium animalis subsp. lactis BB12, Bifidobacterium bifidum CCDM 94, Bifidobacterium thermophilum RB 267, Lactobacillus bulgaricus ATCC 7517, Lactococcus lactis subsp. Lacti and Lactobacillus rhamnosus RW-959-M. Anticariogenic properties of caseinomacropeptide were successfully used against selected strains of Streptococcus mutans, sanguis and sorbinus and model hydroxyapatite system. Glycosylated forms of polypeptide also stimulated gastric and pancreatic enzymes and cholecystokinin secretion but any positive influence of caseinomacropeptide on appetite and hunger suppressing and food and energy intake decreasing has not been observed.
The Development of Local Government in the Municipality of Mníšek pod Brdy

The main aim of the thesis was determined to identify factors influencing political decisions of municipal government in Mníšek pod Brdy city. The Diploma Thesis is divided into three parts, the first of which is dedicated to local authorities, including their composition and powers, as well as issues of municipal elections, local political partisanship, and the participation of citizens in the conflict lines in local political space. Next part of this thesis was focused on indicators of proportionality elections and theories dealing with different views of the local coalition. The second part deals with the general characteristics of the city and its socio-economic structure. The last, main part of the Diploma Thesis is devoted to the presentation of each electoral period from 2002 till 2014. The principle of development and function of coalition within decision-making of the local government is described on the basis of analysis. In the end of the thesis factors, that influence coalition relationships and their reasons, are presented.
