[cze][eng] Saturejka horská (Satureja montana L.) je rostlina vyskytující se ve Středozemí a oblasti Černého moře. Pro vysoký obsah esenciálních olejů má široké využití v kulinářství i v tradiční medicíně.
Saturejka se rozmnožuje generativně i vegetativně. Semena ale klíčí pomalu, a proto je vegetativní způsob množení rychlejší a zejména v in vitro podmínkách lze za krátký čas namnožit velké množství rostlinného materiálu.
V této bakalářské práci byl zkoumán vliv cytokininů na regeneraci a růst rostlin S. montana L. z nodálních segmentů v in vitro podmínkách. Nodální segmenty byly kultivovány na 6 variantách kultivačního media s přídavkem 0,5; 1 a 1,5 mg/l 6- benzylaminopurinu (BAP) a 0,5; 1 a 1,5 mg/l kinetinu (KIN). Varianta bez přídavku růstových regulátorů sloužila jako kontrolní. Podmínky kultivace byly 25/20 °C den/noc, světelná intenzita 3000 lx a časová perioda 16/8 h. Po 6 týdnech měření byly hodnoceni parametry jako: počet výhonů, výška rostliny, počet nově vytvořených nodů a počet a délka kořenů.
Z hlediska koeficientu mikropropagace (tj. počtu nově vytvořených nodů na rostlině) byla statisticky průkazně nejlepší varianta media s přídavkem 1,5 mg/l KIN, kde se vytvořilo průměrně 8,88 +/- 2,87 nodů. U výšky rostliny, která byla nejvyšší u varianty s koncentrací 1,5 mg/l KIN (2,27 +/- 1,33) a u počtu výhonů s největší průměrnou hodnotou u varianty 1 mg/l BAP (2,87 +/- 1,19) nebyly rozdíly v přídavku růstových regulátorů statisticky prokázány. Vysoké koncentrace BAP a KIN inhibovaly tvorbu kořenů a z variant převedených do ex vitro podmínek regenerovalo 46 % rostlin.
Přidání BAP a KIN do kultivačního media má vliv na regeneraci rostlin u Satureja montana L.
Winter savory (Satureja montana L.) is a plant occurring in the Mediterranean and the Black Sea region. For its high content of essential oils is widely used in culinary and traditional medicine.
Savory is propagated generative and vegetative. Seeds germinate slowly, and therefore, the vegetative propagation is faster, especially in in vitro conditions may be propagated a large amount of plant material in a short time.
The aim of this thesis was to investigate the growth effect of cytokinins on regeneration and growth plants from nodal segments of S. montana L.
Nodal segments were cultured on six variants of medium supplemented with 0.5; 1 and 1.5 mg/l 6-benzylaminopurine (BAP) and 0.5; 1 and 1.5 mg/l kinetin (KIN). Variant without growth regulators served as a control. Culture conditions were 25/20 °C day/night, light intensity 3000 lx and daily period of 16/8 hours. After 6 weeks of measurements were evaluated parameters such as: number of shoots, plant height, the number of newly created nodes and the number and length of roots.
In respect of the coefficient of micropropagation (i.e. the number of newly created nodes on the plant) was statistically significantly best variant medium supplemented with 1.5 mg/l KIN where on average create 8.88 +/- 2.87 nodes. At the height of the plant which was highest in the variant with a concentration 1.5 mg/l KIN (2.27 +/- 1.33) and the number of shoots with highest average value in a concentration 1 mg/l BAP (2.87 +/- 1.19) encore of growth regulators were not statistically proven. High concentrations of BAP and KIN inhibited the induction of roots and from variants transferred to ex vitro conditions were regenerated 46 % plants.
Adding BAP and KIN into the medium has an influence on plant regeneration of Satureja montana L.
