Chiral HPLC separation of newly synthesized amino acids

The aim of this work was to develop and to optimize HPLC method for enantioseparation of newly synthesized derivatives of amino acids. The set of these analytes contained four N- blocked derivatives of D,L-Phenylalanine, three N-unblocked derivatives of D,L- Phenylalanine, , one methylated derivative of D,L-Tyrosine and D,L-Tyrosine. Two separation modes i.e. reversed phase and polar-organic modes and two columns i.e. Chirobiotic® T and Chirobiotic® T2 were used. Chiral stationary phases of these columns were composed of macrocyclic antibiotic teicoplanin coated on silica gel support. As mobile phases in revesed phase mode, methanol and acetate buffer were used with Chirobiotic® T column. The most suitable concentration were 20 mM and the most suitable pH value were 4.00. Under the above mentioned conditions eight analytes of ten were separated and the optimal conditions were found. Polar-organic mode and Chirobiotic® T column were suitable only for enantioseparation of three N-blocked derivatives of D,L- Phenylalanine. Mobile phases were composed of methanol with small additions of triethylamine and acetic acid. Chirobiotic® T2 column in reversed phase mode was not suitable for tested set of analytes. Chromatograhic characterization of chiral and achiral separation systems

Dissertation thesis is a 5-publications' collection concerning characterization and application potential of cyclodextrins, polysaccharides and macrocyclic antibiotics based chiral stationary phases. The effects of stationary phase and mobile phase are studied. This approach ensures the complex insight into separation systems studied. Systems with different nature of chiral selector were studied by HPLC. Namely, macrocyclic antibiotics and derivatized polysaccharides were used for experiments. Former ones provided better results for enantioseparation of non-coded amino acids than latter ones. Dynamic coating procedure was used for preparation of a new chiral stationary phase. Characterization of new cationic cyclodextrin based chiral stationary phase was performed. Linear free energy relationship method was used for characterization of two different separation systems, i.e. newly prepared stationary phase and commercially available stationary phase. Based on results obtained, newly prepared stationary phase showed better results for separation of different achiral groups of analysts. New stationary phase prepared by dynamic coating was compared with chromatographic system, in which the chiral selector was used as a mobile phase additive. Title: The strategy and tendency ofTurkey's economic accesion to EU Turkey has status of "applicant country'' and its negotiation about membership in EU proceeding the longest time among all candidates and member countries, for all time of existence ofEU and that since 1963. This work is concemed at appreciation ofTurkey's Republic position in relations with EU. Thesis monitors and follow its progress of tendency of membership in EU - especially in economic aspects. The main aim is to finde, which economic indexs are comparable worth with EU and its acquis communitarie and whether is Turkey acceptable applicant state for EU. Trough the sport sphere measure hypothesis, whether specific Turkish temper has place in Europe. According to determined macroeconomic indexs, which are in many cases comparable with some member countries of EU, Republic of Turkey is from the economic aspect acceptable candidate country to EU and their specific temper don't mean any problem in acces10n proces. Optimization of HPLC method for separation of tetracyclines

The aim of this work is to develop and to optimize HPLC method for separation of a set of four tetracycline antibiotics - tetracycline, chlortetracycline, oxytetracycline, and doxycycline. Four different reversed octadecyl-silica stationary phases in various mobile phase compositions were examined in isocratic elution. The baseline resolution of all the analytes was obtained by using two columns - Astec C18 and Atlantis C18 I. The optimized separation system consisted of Atlantis C18 I. column, ACN/0.1% formic acid 15/85 (v/v) as the mobile phase and flow rate of 2 ml/min. The baseline resolution was achieved and the analysis time did not exceed 27.5 minutes.
