Endophytic fungi in above-ground tissues of plants of the family Ericaceae

Many studies focus on endophytic fungi. However, above-ground tissues of plants of the family Ericaceae are not largely examined. Most of the literature concerns the fungi forming ericoid mykorhiza. This bachelor thesis sums up the diversity studies of the endophytic fungi in the family Ericaceae, compares the frequency and the overlap of fungi in different host plants and geographical regions and also the occurence of fungi in living and dead plant tissues. The methods of fungal isolation and identification are accented.
Characterization of fungal community in spruce (Picea abies) litter using cultivation and T-RFLP

Fungi have a key role in the decomposition of coniferous litter and affect nutrient cycling in forest ecosystems. Therefore, great emphasis is placed on exploring the diversity of these organisms. The aim of this thesis was to describe fungal diversity in spruce litter and revealed temporal development of this community in a forest regenerating after bark beetle outbreak. Another objective was to compare sites with different length of bark beetle damage. The study area was located in the Bohemian Forest mountain range. Litter bags with spruce needles were placed on the forest soil and several samplings were performed in the course of three years. Diversity of fungi and changes in the fungal community were assessed by two methods: cultivation of needles on 2ř MEA and fingerprinting method T-RFLP. In total 71 fungal species were obtained from needles during a three-year succession by cultivation approach. Using T-RFLP 122 different fragments were generally recorded. The dominant species were Scleroconidioma sphagnicola, Thysanophora penicillioides, Hormonema dematiodes, Ceuthospora pinastri, species of genus Chalara, Trichoderma polysporum, Mycena galopus and unknown species Helotiales sp. 1. The effect of oribatid mites on the composition of fungal community in pine litter

4 Abstract The aim of this study was to determine whether oribatid mites affect the variety of fungi in litter of Scots pine (Pinus sylvestris), and if the dispersibility of fungi particles is related to oribatid mite species. To this end, I have prepared several semi-natural systems, in which I observed dispersion of fungi separately and in the presence of two oribatid mite species: Phthiracarus sp. and Adoristes ovatus. I assessed the experiment in two ways, by a cultivation of washed needles on agar medium and direct observation of needles under a binocular magnifier. In the next part of this study I isolated particles of fungi present on the surface and in the faeces of oribatid mites Adoristes ovatus, Porobelba spinosa and Spatiodamaeus verticillipes and compared it with the respective oribatid mite species. The most frequent needles fungi spread by oribatid mites were Oidiodendron maius var. maius, Oidiodendron muniellense, Oidiodendron setiferum, Umbelopsis ramanniana, Mucor hiemalis f. hiemalis and Mortierella hyalina. Interactions of microscopic fungi and testate amoebae in Norway spruce litter

Both testate amoebae and fungi are common inhabitants of coniferous litter. Their interactions in this environment were rarely studied, although they reach high biodiversity and can play a significant role in nutrient cycling in this environment. In this study, a cultivation of litter needles in the damp chambers was used to investigate interactions between fungi and testate amoebae. Observation of spruce litter needles in environmental scanning electron microscope was used to better characterize testate amoebae communities directly on the needles. Additionally, two experiments changing the biotic conditions in the microcosm were used to follow a principle of the interactions. Three species of testate amoebae from litter needles were able to colonize the filter paper on the bottom of the damp chambers. Occurrence of Phryganella acropodia and Assulina muscorum on the filter paper was significantly fuelled by the presence of mycelium. Assulina muscorum was associated with the fungal spores and Arcella discoides was attracted by sporulating colonies of Cladosporium spp. in the damp chambers. In situ ruminal degradation of organic matter of Trifolium pratense L.

Seven clover sampes (Trifolium pratense L.) were collected at three different miters I (n = 3), II (n = 3) and III (n = 1) during the growing season from 10th of May to 17th of August. The samples were analyzed for chemical composition, gross energy (BE) content, in vivo sheep digestibility of organic matter (KS OH) and gross energy (KS BE) and in situ rumen degradability of organic matter and neutral detergent fibre (NDF). The contents of ash, crude protein (NL), crude fibre (CF), NDF, acid detergent fibre (ADF), acid detergent lignin (ADL) and BE were significantly (P < 0,05) affected by the date of cutting time. The averaged values were for ash 119,2 g/kg of dry matter, NL 197,7 g/kg of dry matter, CF 214,1 g/kg of dry matter, NDF 400,7 g/kg of dry matter, ADF 296,2 g/kg of dry matter, ADL 73,8 g/kg of dry matter and BE 18,2 MJ/kg of dry matter. KS OH and KS BE generally decreased with higher dates of cutting time. On average KS OH and KS BE amounted 72,4 % and 70,2 %, respectively. The effective ruminal degradability of organic matter (EDOH) was in average 81,7 % for miter I, 79,7 % for miter II and 75,2 % for miter III. In situ organic matter degradability characteristics were in average 41,9 % for the immediately degradable (soluble) fraction (parameter a), 47,1 % for the potential degradable fraction (parameter b) and 0,105 h-1 for the fractional rate of degradation (parameter c). The effective ruminal degradability of NDF (EDNDF) also generally decreased (P < 0,05) with increasing date of cutting time with values of 63,0 % for miter I, 59,7 % for miter II and 51,6 % for miter III. Strong correlation (P < 0,05) was observed between the EDOH and EDNDF (r = 0,979).

KS OH and KS BE generally decreased with higher dates of cutting time. On average KS OH and KS BE amounted 72,4 % and 70,2 %, respectively. The effective ruminal degradability (ED) of NDF generally decreased (P {>} 0,05) with increasing date of cutting time with values of 66,1 % (May 10), 63,6 % (May 18), 59,2 % (May 25), 64,8 % (June 29), 57,4 % (July 7), 56,9 % (July 13) a 51,6 % ( August 3). In situ characteristics were in average 77,1 % for the potential degradable NDF fraction (parameter b), 0,0703 h-1 for the fractional rate of degradation (parameter c) and 77,8 % for the digestible NDF (parameter DNDF).
