Title of diploma thesis: Production of secreted aspartic proteinases by pathogenic yeasts The objective of this thesis was to identify the effect of farnesol as a quorum sensing molecule, which facilitates intracellular communication of Candida albicans, on the production of secreted aspartic proteinases (Sap). The theoretical section summarizes the morphology of Candida, the virulence factors, the illnessess cause by Candida and its treatment. A separate chapter focuses on secreted aspartic proteinases and on farnesol. The strains tested came from clinical isolates belonging to the Clinical Microbiology Institute of FN Hradec Kralove; two further strains were collected as well (10231 and 25188). The ATCC 10231 strain, which did not show farnesol production but the production of farnesol acid, was included in the test. Special media contained a source of bovine hemoglobin as the only source of nitrogen and had differing pHs. Quantitation of agents in exhaled breath condensate.

Measurement of parameters in exhaled breath condensate constitute (EBC) a rapidly progressing field of research. The collection procedure is simply and comletely noninvasive. EBC contains large number of biomarkers including hydrogen peroxide, nitrite, nitrate, 3-nitrotyrosine, nitrosothiols, adenosine, prostaglandines, thromboxanes, leukotrienes, isoprostanes, cytokines. Use of EBC is based on hypothesis that aerosol particules exhaled in human breath reflect the composition of the bronchoalveolar extracellular lining fluid. The term EBS stricly relates to exhaled samples collected by cooling the exhaled breath. Measurement very low concentrations of some biomarkers requires very senzitive analytical methods. The evaluation of the available studies revealed that asthma, COPD and ARDS compered with healthy control groups lead to a significant increase in hydrogen peroxide concentration. The result of the work indicate that the content of hydrogen peroxide in the breath condensate provides new information for diagnosis and therapy of inflammatory diseases of the respiratory tract when the collection of specimen and the measurement procedure are standardized.
Change monitoring of the coagulation during physiological pregnancy.

This project monitored changes of coagulation during physiological pregnancy by metering of choice coagulation parameters. Includes also the summary of coagulation changes near gravid women and complications that are may their owing to come. For change monitoring was measured value of Protrombin test (PT), Activated parcial tromboplastine test (APTT) and level of fibrinogen (FBG) and D-dimer (DDI) near healthy of gravid women in time up to 20., 30. and 40. week of pregnancy. Metering was effected on Dade Behrings analyzer in haematology laboratory CHL and TC General faculty hospital Prague. Outcomes were statistically processed. Was found, that the value of coagulation tests PT (P=0,0793) and APTT (P=0,1232) not show during pregnancy any relevant changes, happens only to their mild fall in terms of physiological divide. On the contrary, level FBG (P<0,0001) and DDI (P=0,0311) during pregnancy increased. Values of all parameters depend on a week pregnancy. Pregnancy is hypercoagulation state, which is effected above all decrease fibrinolytic activities and products of placenta. Imbalance among for- and anticoagulative action is able to make many complications, threating mother also fetus. Hypercoagulation conditions bear to also found increased levels of FBG and DDI.

Pardubice Company (AMAc-1, AMAc-2, AMAc-3, AMAc-4, AMAc-5); Composition of paint unknown (we only known that paint include photocatalytic additives); Pathogen bacterial strains: E. coli, S. aureus, P. aeruginosa. Methods: Contact of microbes with antimicrobial photocatalytic paint: pre-activated with UVA-light, activated with UVA-light at the same time, non-activated with UVA-light. Main findings: Graphs show how number of CFU microbes is changing in time; Definition of efficacy (-k): negative value of tangent of line linear regression. Conclusions: Confirmation of hypothesis: grow of efficacy of antimicrobial photocatalytic paint after exposition UVA-light; Influence on fall of number microbial CFU of strain E. coli; Significant after UVA irradiation (activation); S. aureus: fluctuating of results, all the same definite fall of bacterial CFU after activation UVA-light; Most resistant to efficacy of antimicrobial paint was P. aeruginosa; Influence of...
