Interaction of selected anthocyanidins with farnesoid X receptor
Jeřábková, Jana ; Pávek, Petr (advisor) ; Martin, Jan (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmacology & Toxicology Student: Jana Jeřábková Supervisor: Doc. PharmDr. Petr Pávek, Ph.D. Title of diploma thesis: Interaction of selected anthocyanidins with farnesoid X receptor Human farnesoid X receptor (FXR) is a member of nuclear receptor superfamily that act as ligand-activated transcription factors. FXR binds to specific regulatory DNA regions and induces expression of many target genes. These regulated genes are involved in bile acid metabolism and transport, maintaining blood lipids, liporoteins and glucose homeostasis and also contribute to maintain intestinal bacterial balance, hepatoprotection and liver regeneration. The interest of recent studies is to test the range of FXR ligands for treatment and prevention of many diseases such as cholestais, cholesterol gallstone disease, steato-hepatitis, dyslipidemia, atherosclerosis, type 2 diabetes mellitus, metabolic syndrome, liver cancer and other forms of cancer such as breast cancer. In this experimental diploma thesis we are focused on testing of potencial ligands of human farnesoid X receptor from the group of natural plant pigments anthocyanidins (cyanidin, delphinidin, malvidin, pelargonidin, peonidin and petunidin) using the human hepatoma cell line...
Study of Novel FXR Ligands
Jeřábková, Jana ; Pávek, Petr (advisor) ; Schreiberová, Jolana (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmacology & Toxicology Candidate: Mgr. Jana Jeřábková Consultant: prof. PharmDr. Petr Pávek, Ph.D. Title of Thesis: Study of Novel FXR Ligands Human farnesoid X receptor (FXR) is ligand-activated transcription factor that belongs to the nuclear receptor superfamily. FXR binds to its response element (FXRE) as monomer or heterodimer with retinoid X receptor (RXR) resulting to activation of transcription of target genes. FXR positively or negatively regulates a wide range of target genes involved in maintaining the homeostasis of bile acids, cholesterol, lipid and glucose metabolism. Furthermore, FXR transcriptionally regulates genes involved in regulation of the immune response, in the development of inflammation and maintaining the intestinal barrier function and genes controlling coagulation and remodeling of the vessel wall. The interest of current scientific studies is to find the new ligands of FXR, which would be suitable for therapeutic use. Besides the natural bile acids, the first medicinal product containing semi- synthetic FXR ligand, obeticholic acid (6-ECDCA) as active substance was registered last year for the treatment of primary biliary cholangitis. In this experimental rigorosum thesis we focused on...

Harmonization and Coordination of the Income Taxes in the European Union
Jeřábková, Jana ; Láchová, Lenka (advisor) ; Matějka, Václav (referee)
This thesis deals with the development of various types of cooperation in tax matters and consequently historical development efforts of the harmonization and coordination. First of all, the thesis is focused on the income tax and current developments within the European Union and the impact of the economic crisis on the region. Income taxes are one of the most important issues, as they affect almost everyone. Development in this area has a significant relationship with development of economic conditions and with increasing mobility of the subject of tax.
