Multivalent carbohydrate ligands of galectins

Galectins are proteins, wich belong to a group of lectins that are able to bind to saccharide units and they specifically recognize glycans exposed to the surface of the cells. Galectins participate in vivo, for example, in carcinogenesis, angiogenesis or fibrosis. Their occurrence increases significantly in connection with a number of pathogenic processes, therefore they can be used as markers for some types of cancer or cardiopathology and also for the targeted binding of therapeutics and/ or imaging agents in diagnosis and therapy. Galectin-3 has a specific structure known as chimeric and it is capable of forming multivalent oligomers. The natural ligands of galectins are glycans containing terminal β-galactosides, especially N-acetyllactosamine, but the binding of monovalent glycans is very weak. Glycoconjugates with high affinity to galectin receptors are optimally multivalent, biocompatible and stable in vivo. These criteria accomplish carbohydrate ligands conjugated to soluble and structurally flexible N-(2-hydroxypropyl)methacrylamide (HPMA) copolymers. The understanding of digital education on elementary school and its cceptance amongs the pupils

5 ABSTRACT The diploma thesis deals with the topic of digital education at primary school and its acceptance among pupils. The project is an inspiring attempt to link formal and informal education in the learning process. This study has become an inspiration for making digital teaching more effective and developing digital literacy for primary school pupils. The theoretical part is divided into three main chapters, which deal with the area of cognitive development of early school age children, digital education and digital technologies used in teaching. The knowledge from the professional literature in the theoretical part is the basis for the practical part, which through the creation of two educational geolocation games researches the development of individual digital competencies on a sample of two classes. The aim of this work is to find out how digital literacy and its individual competences can be developed in the primary school with respect to the age and cognitive development of pupils. Engineering of microbial glycosidases for modifying synthetic potential

Glycosidases (EC 3.2.1.) alias glycoside hydrolases are enzymes that catalyze the cleavage of a glycosidic bond between two carbohydrates or between a carbohydrate and an aglycone. Under suitable conditions (especially reduction of water activity in the reaction mixture), these enzymes are also able to synthesize a glycosidic bond. By targeted mutagenesis of the catalytic centre of the enzymes, it is possible to suppress or completely abolish their hydrolytic activity. Enzyme synthesis using glycosidases makes it possible to prepare bioactive galactosides, for example galectin ligands. The present work deals mainly with β-galactosidase from Bacillus circulans, its recombinant expression and mutagenesis. In the first part of the work, the commercially prepared plasmid of -galactosidase from B. circulans isoform A that I designed was used for recombinant expression in E. coli. It was necessary to optimize the conditions of the enzyme production. As it is a large protein (189 kDa), the expression vector pCOLD II and cold production at 15 ř C were used. The enzyme is specific for the formation of the β-1,4 glycosidic bond and has been used to synthesize complex tri- and tetrasaccharide ligands that cannot be prepared with a crude commercial preparation containing undesirable enzyme activities....

The Reflection of Great Depression in 1929-1933 in Czech literature of economy until 1945 - fundamental overview

The fundamental aim of this bachelor thesis is the essential overview of reflection of Great Depression in czech economic literature untill 1945. The focus is given on works of the main economists and their most important works concentrated on economical crisis. For this goal the political and economical development since the foundation of Czechoslovakia related with development of the economical crisis is analyzed in theoretical part of work. The explanation of crisis of some political parties is included in this theoretical part of work. Then follows short biography ? introduction of selected economists and their prefered economical view and directions. This bachelor work could be used as a study material in the course Macroeconomics and History of economic thinking aimed on Czechoslovakia during 1929 ? 1933.
Current problems of the school integration from the point of view of school and family

The diploma thesis deals with integration of children with special education needs into the common primary schools. The theoretical part is focused on phenomenon ``integration{\crqq}, its types, evolution of integrated education in the Czech Republic and abroad, and integration legislature. Further the conditions necessary for realisation of school integration and other factors influencing its success are discussed. The practical part involves a performed research. The aim was to find out the actual problems of integration from point of view of the following people: directors of primary school, infant school teachers and parents of integrated children. To obtain these data the questionnaire research was used, which was done on 17 primary schools. The results of this research are presented and finally discussed in this thesis.
