Ball screw testing
Kukliš, Richard ; Jaroš, Aleš (referee) ; Fiala, Zdeněk (advisor)
The main goal was to design a test rig and create a methodology for evaluating the measured data from the life tests of linear motion systems with ball screws. The first part is the research of basic knowledge about ball screws, which are important for understanding the behavior of the entire ball screw motion system and methods created from previous experiments on similar issues. The next part of the thesis is directed at the design and selection of individual parts and sensors for the testing rig. The methodology is based on the evaluation of measured parameters describing the condition of the tested ball screw and it includes verification of the functionality of the test rig from the first measured data.

This master’s thesis focuses on the design of a combined tool for machining holes of a universal chuck. The thesis analyses the component and technologies used for the production of precision holes for pinions in the chuck body. Furthermore, the current production of precision holes is described, including the used machine, tools, cutting conditions and technological process with machine times. In the next part of the thesis, a proposal of more efficient production of precision holes is made accompanied by a design of a combined tool. The design includes its structural treatment, used cutting tool inserts and drill, the method of clamping, used material, the heat treatment, the determination of the cutting conditions and the development of a new technological procedure. The thesis concludes with a technological and economic evaluation, where the existing and the newly proposed technologies for the production of precision holes for pinions in the chuck body are compared.
Design of production technology for a component of a rotary shape
Dvořák, Dominik ; Fiala, Zdeněk (referee) ; Kalivoda, Milan (advisor)
For the design of the interlocking shaft production technology, this component was first described and after defining the limiting conditions of production, three variants of its implementation were selected. The technologies used were characterized and subsequently the most economically efficient variant was chosen, which also met the specified criteria. A production process with a tool sheet was compiled for this variant. Furthermore, the NC code was created in the control system of the production machine, followed by its verification during the simulations. Two prototypes were made. The first of them was used to optimize the design of production technology, the second was produced after simulations to verify the correctness of the proposed solution. This was followed by a technical and economic evaluation, used to predict the production costs of the entire production volume and compare them with the estimate.

Public Participation in Environmental Decision-making
Balážová, Julie ; Sobotka, Michal (advisor) ; Fiala, Zdeněk (referee)
Public Participation in Environmental Decision-making Abstract This thesis is dealing with participation of public in environmental administrative proceedings. The topic is widely discussed in connection with efforts to quicken planning and construction proceedings. It is, however, far from limited to them - the subject matter is scattered in a multitude of legal acts. Therefore, the aim of this work is to present various acts and analyse the requirements for and the extent of participation of persons in proceedings conducted under them in a compact manner. Firstly, the readers are familiarised with forms of "participation" and various definitions of "public". The legal sources of this topic are introduced next, and a few, that will be addressed in detail in the following parts of this thesis, are selected. Afterwards, the author explains how the code of administrative procedure defines parties to proceedings, how is that important for talking about other acts and what problems can arise from applying these provisions in practice. The next part introduces the readers to an international document that deals with public participation in general - the Aarhus convention and explains its importance in European and Czech law. Furthermore, the author focuses on characterization of legal acts on integrated...
Kinetics of selected substances in dermal exposure (Polycyclic aromatic hydrocarbons)
Kotingová, Lenka ; Fiala, Zdeněk (advisor) ; Bencko, Vladimír (referee) ; Čegan, Alexander (referee)
Kinetics of selected substances in dermal exposure (polycyclic aromatic hydrocarbons) Skin is one of the largest human body organs and its proper functioning is vital for human body. Dermal exposure represents one of the most important pathways of exogenous substances into the body. Data on kinetics of transdermal absorption of polycyclic aromatic hydrocarbons (PAH) have been yet so far insufficient. Present estimates of health risks level connected to dermal exposure to PAHs, are loaded with great deal of uncertainty. The aim of the dissertation thesis was employment and validation of appropriate testing method of transdermal absorption of substances in vitro and the use of this method to obtain new findings on the influence of concentration, dissolvent and form of application, on the measure of dermal absorption of selected PAH representatives. For the testing of transdermal absorption of substances in vitro, the method using static Franz vertical diffusion cell was chosen. As an absorption membrane, full leather of rear surface of the auricle of a domestic pig was used. Content of analytes in samples of receptor fluid was analyzed by gas chromatography with mass spectrometry (GC-MS) and by high performance liquid chromatography with fluorescence detection (HPLC), for which a new methods was developed and...
Development of New Methods for Monitoring of Occurence of Pharmaceuticals in the Environment
Aufartová, Jana ; Solich, Petr (advisor) ; Fiala, Zdeněk (referee) ; Garcia-Campaña, Ana M. (referee)
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Analytical Chemistry Candidate Mgr. Jana Aufartová Supervisor Prof. RNDr. Petr Solich, CSc. Title of Doctoral Thesis DEVELOPMENT OF NEW METHODS FOR DETERMINATION OF SELECTED PHARMACEUTICALS IN ENVIRONMENTAL SAMPLES During last decade, the consumption of pharmaceuticals is increasing substantially. At the same time their occurrence in environment is increasing as well. Pharmaceuticals are released into environment by excretion, as conjugated or unchanged active compounds, as unused pharmaceuticals, which are not disposed according to the recommendations (e.g. thrown away into the rubbish or flushed down the toilet). Other possible sources of pharmaceuticals are in the agriculture, livestock and aquaculture. The current water treatment technologies do not remove all traces of pharmaceuticals in wastewaters. Therefore the monitoring of their occurrence in surface and wastewaters become more important. Antibiotics and steroid hormones are a group of drugs used in human and veterinary medicine. The main problem with these groups of substances is the emergence of bacterial resistance, in the case of antibiotics, and the effect on the endocrine system, including the reproductive cycle, in the case of steroid hormones. The...
The Possible Assessments of Unprofessional Exposures of Low Frequency Noise
Potužníková, Dana ; Fiala, Zdeněk (advisor) ; Bencko, Vladimír (referee) ; Argalášová, Ĺubica (referee)
v anglickém jazyce The possible assessments of unprofessional exposures of low frequency noise Introduction: Along with technical development, the number of sources of low frequency noise increases in the human environment and the number of complaints from people exposed to noise from these sources increases as well. The statutory limit for non-professional exposure to low frequency noise has not yet been established (due to the limited amount of data on low frequency noise exposure risks) in the EU countries (USA) and no WHO recommendations have been issued in this area. For these reasons, the hygienic limit for municipal/non-professional exposure to low frequency noise was not specified in the Czech Republic. Objecticve: The present dissertation thesis was to investigate whether the short-term experimental exposure to low frequency noise with the tonic component induces significant changes in the circadian rhythm of cortisol and alpha-amylase in saliva in the experimental set of volunteers. Saliva is a biological material that allows low-invasiveness collection. The work was supposed to assess whether possible changes could be used as objective indicators of the presence of the stress response to the noise exposure. Material and metods: An experimental study was conducted on a set of 100...

Vytlačilová, Jitka ; Jahodář, Luděk (advisor) ; Kočí, Vladimír (referee) ; Fiala, Zdeněk (referee)
The aim of the presented doctoral thesis was to use the sludge worm Tubifex tubifex as a model for biological activity testing. In the presented study, the Tubifex worm was used to evaluate photosensibilisation, i.e. sensibilisation of an organism towards UVA radiation (365 nm; radiant flux density of 350 μW/cm2) after contact with certain substances. The tested substances included proven photosensibilizing agents - rose bengal, khellin and xanthotoxin; isolated natural substances - thiophene polyacetylene umbelliferon, scopoline, and scopoletine; synthetic compounds - salicylanilides and thiosalicylanilides; and penicillin drugs Augmentin and Ospamox and their active substance standard amoxicillin. The final evaluation of photosensibilizing properties of the substances was carried out according to the Commission Directive 2000/33/EC PHOTOTOXICITY - IN VITRO 3T3 NRU PHOTOTOXICITY TEST. The test was performed in 24-well plates where one set of plates with the photosensibilizing agent was irradiated and the second set was placed in an incubator (20±2řC) as a dark control without any light presence for the whole period of the experiment. The plates were evaluated immediately after the given period of irradiation (15, 30, 45, 60, 120, and 240 minutes). Mortality and percentual number of damaged individuals were...
