Cancer is one of the major causes of death in the world, especially in developed countries. There are already many antiproliferative agents used to cure patients with cancer. However, many of the currently approachable medicines are synthetic chemicals, which are invasive and very often cause alterations in healthy cells. Because of these reasons, there is a tendency to turn back to natural agents used mainly traditionally in folk medicine, because some of them might be potentially effective against tumor development. In this study, the cytotoxic effects of basil, laurel and juniper extracts on neuroblastoma cancer cells were tested by using MTT and LDH assays. The extracts at concentrations 2; 0,5; 0,1; 0,05 and 0,01 mg/ml were effecting the cells for 12 hours. Another method used for studying of extract's effects was western blotting analysis showing the level of tumor supressor p53 protein in the neuroblastoma cells after extract treatment. The results with treated cells were compared to the untreated cells serving as a control. The results of MTT assay show decrease in cell viability of cells treated with all three plant extracts at concentration 2 mg/ml. Adherence to pharmacotherapy by idiopathic inflammatory bowel diseases

Aim of the work The aim of the work was to analyze patient's adherence (compliance) to the pharmacotherapy of inflammatory bowel disease in Czech Republic, as well as to analyze quantity and character of potential drug interactions in medications of the tested kohort. Our aims were : 1. to analyze adherence to pharmacotherapy of IBD: o in the patients cohort followed up in the single IBD Unit (Gastroenterological Centre, the 4th Medical Department, Prague) o in the patients cohort followed up in a few gastroenterological ambulancies in the Czech Republic 2. to analyze potential drug interactions in medications of IBD patients Results 1. Adherence to pharmacotherapy of IBD in the patients cohort followed up in the single IBD Unit (Gastroenterological Centre of the 4th Medical department, Prague) 177 patients were enrolled in the study (84 male, 93 female, 117 CD, 60 UC). The patients evaluated their adherence to the pharmacological treatment in an interview. From 47 patients, treated by mesalazine, the urine sample for HPLC analysis of mesalazine was taken. Observations of dispensing of several drugs with dependence potential in Pharmacies

Monitoring of the dispension of drugs with abuse potential in pharmacy Kristýna Zábranská Prof. MUDr. Zdeněk Fendrych Medication over the frame of standard treatment is a serious as well as underestimating problem. In the conseqeuncies it leads to hard medical and economic damage. Analysis of the dispension of prescription drugs with abuse potential (analgesics, benzodiazepines, nonbenzodiazepine hypnotics) in the pharmacy in 2006 confirmed that the consumption of this drugs is high, especially in the elderly (51 - 60 years). Group of the patients in this age was dispensed 72% of all dispensed drugs, that were monitoring. Monitored drugs were more frequently prescribed to women, in particular benzodiazepines. The three most prescribed substances were diazepam, alprazolam and bromazepam. In this sequence they were dispensed to women too. To the group of men there was the most often prescribed substance diazepam, zolpidem (Hypnogen) and tramadol (Tramal).

Göteborg University, Sweden; The Sahlgrenska Academy; Institute for Neuroscience and Physiology; Department of Pharmacology Nowadays, the number of patients with lower urinary tract symptoms increases. The goal of this project was to explore the effect of 4-DAMP (4-diphenylacetoxy-N- methylpiperidine) on the regulation of the overactive bladder syndrome (OAB). Furthermore, the effect of induced cystitis by cyclophosphamide on the urothelium changes, was observed. It was found that there are some unspecified mechanisms that can compensate for the present cystitis. But only poor effect on incontinence and different effects in different doses lead to conclusion that 4-DAMP is not the best for the treatment, but additional tests are needed.
Characterisation of Rat Alveolar Cell Line R3/1

The rat cell line R3/1 displays several phenotypical features of alveolar epithelial type I cells. In order to characterise the cell line as an in vitro model for drug disposition studies, R3/1 cells were cultured on Transwell filters and the transepithelial electrical resistance (TEER) was measured as a parameter for the integrity of cell layers. Presence of cell junctional proteins (i.e., E-cadherin, occludin, ZO-1 and ZO-2) was studied on mRNA (by reverse transcriptase-polymerase chain reaction, RT-PCR) and antigen level (by immunofluorescence microscopy, IFM). Moreover, the expression pattern of catabolic peptidases (carboxypeptidase M, aminopeptidases A, B, N and P, γ-glutamyltransferase, dipeptidylpeptidase IV, angiotensin-converting enzyme, and endopeptidases 24.11 and 24.15) was analysed in R3/1 cells and compared to rat alveolar epithelial I-like cells in primary culture. TEER values were peaking at ~99±17 Ω cm2 after 5 days in culture. Addition of 0.1 µM dexamethasone together with foetal bovine serum at 20% increased TEER by 65%. However, none of culture conditions used in our study yielded monolayers with TEER values comparable to those of primary cultures of rat pneumocytes. Molecular biological evidence of Pneumocystis jirovecii (carinii) with application of the real-time PCR

Pneumocystis carinii is an opportunistic pathogen, which often causes fatal pneumonia in patients under immunosuppressed or immune deficient conditions due to AIDS, cancer chemotherapy, or immunosuppressive therapy for organ transplantation. Different techniqes of microscopy and a nested polymerase chain reaction (PCR) are widely used for detection of this opportunistic fungus. But these methods are less sensitive and time-consuming. We focused our attention on the level of specific DNA by a quantitative PCR technique. This procedure has the advantage of greater precision and more objectivity. In this report we describe a real-time PCR assay suitable for use with the LightCycler system. We were successful in implementation and optimalization of quantitative real-time PCR for Pneumocystis carinii. We have reached sensitivity 1000 copies of DNA Pneumocystis carinii /ml. We have set a limit of detection to be 50 copies of DNA Pneumocystis carinii /ml. Real-time PCR procedure was optimized and validated in laboratory of molecular biology of Department of clinicall biochemistry and diagnostics of Fakultni Nemocnice in Hradec Kralové. A standard routine was established to be utilized in general practice
