Application of microextraction techniques for analysis of biological samples

This bachelor thesis deals with the analysis of basic drugs in biological samples by in line coupling of supported liquid membrane (SLM) to capillary electrophoresis (CE). The tITP method was included as an on-line preconcentration step to achieve better detection limits with ammonium acetate as the tITP stacker and acetic acid as the CE background electrolyte. The practical part of the thesis focuses on the optimization of the tITP and the SLM extraction conditions in the analysis of basic drugs in complex biological samples (human urine). The best analytical results were achieved with hydrodynamic injection of ammonium acetate for 15 s and basic drugs for 15 s. The donor was alkalised with 10 mM sodium hydroxide and acceptor was acidified with 10 mM hydrochloric acid. From the results, the in-line coupling of SLM extraction to CE and stacking by tITP is an efficient method allowing direct analysis of complex samples with improved sensitivity of the analytical method.

The diploma thesis deals with design of the polyfunctional center and the whole complex for the Brno University of Technology, which would serve for its representation. The land is in Brno on Kraví hora and its location has huge potential. However, the area is not used by the university, so the existing buildings are in poor condition. Therefore, the aim of the proposal is to find a suitable use of the land that would meet the university's requirements together with the city's intentions.
Kraví hora - BUT Family silver

The subject of the diploma thesis is to design a project study and a building program that would become a compromise between the requirements of the city of Brno and BUT in the area of Kraví hora in the Brno city center. The city of Brno would like to transform the area into a public park. But BUT considers it as its "family silver" because they realize its extraordinary value. Options of a constructions are really limited because of many area regulations. My proposal counts on the demolition of all existing buildings in the area, respects property area relations , protection zone, continuity with existing traffic roads, height regulation, imaginary raster of surrounding buildings, slope of terrain and orientation to the cardinal points. My suggest represents a study of five groups of objects which creating a rectangular raster. As a compromise solution, I chose the presentation and promotional areas of the individual faculties of BUT for the content of the buildings. Faculties have spaces for hobby groups for the public comprise all age groups included. I also designed the BUT Museum of History, a multimedia hall, a cafeteria, studios, a student leisure center and an observation tower with a climbing wall.

The subject of this diploma thesis is to continue on the previous atelier work, which was focused on elaboration of a strategic urban-architectural study of the Kraví hora area and the BUT holding located there. A multifunctional facility, which would include activities that BUT still lacks, should be designed there. In any case, it should not be an extension of the teaching, research or accommodation capacities, since there have been built enough of these by BUT recently. At the same time, the proposed activities should not be controversial with the interests of the city. The BUT area at Kraví hora is one of the most valuable areas of the city of Brno. The city, which has tried to buy it from BUT several times, has an eminent interest in this area. However BUT considers it as its "family silver", because they realize its extraordinary value. Nevertheless, due to the binding regulatory conditions, construction possibilities are very limited. The aim of this study is to find such a use of this area, which would be a compromise corresponding to both the city's expectations and the needs of BUT. The intention of the previous project, which has been continued by this diploma thesis, was the removal of unsatisfactory buildings and the reduction of the garden colonies; despite to a study created by the City of Brno but partially kept, cultivated and reorganized; and rebuilding of the BUT area, which will be opened to the public and communicate with park. The public will obtain an access to nowadays inaccessible and impermeable areas by removing inappropriate buildings and fences, as well as by the modification of the public space and creating one major spine communication realized as an extension of the existing road and which will serve only for the supply, hikers and cyclists. The aim of the diploma thesis is a detailed study of the VUT area at Kraví Hora to a scale of 1: 200. The aim is to combine the interests of VUT and Brno in this extraordinary territory for the benefit of both subjects. For the needs of the city, I created a publicly accessible park in which I placed the VUT museum buildings and the spaces for the promotion of individual VUT faculties that the Technical University lacks. I have also placed the object of the creative business center here. These are rented spaces especially for VUT graduates and VUT employees who want to do creative work. Both buildings with a green walkable roof are embedded in the terrain so as not to disturb the overall impression of the landscape and create a natural transition between the buildings and the city park.
Rapid fabrication of microextraction devices for direct determination of basic drugs in dried blood spots

A 3D-printed cutter was designed for rapid fabrication of supported liquid membrane (SLM) extraction devices compatible with injection system of commercial capillary electrophoresis (CE) instrument. Low cost and single use microextraction devices were fabricated from polypropylene micropipette tips with high precision (dimension variances ≤ 3% RSD). Practical applicability of the microextraction devices was examined by in-line coupling of SLM extractions to CE for direct analysis of four model basic drugs in dried blood spot extracts. Excellent repeatability of the hyphenated technique was achieved (RSD ≤ 7.6%), transfer of analytes across the SLM ranged from 23 to 73% and limits of detection were as low as 0.07 μg/mL.
Hollow fibre-liquid phase microextraction of basic drugs from dried dried blood spots

Hollow fibre-liquid phase microextraction (HF-LPME) was applied for rapid and efficient extraction of basic drugs from dried blood spots (DBS). Basic drugs from DBS were first extracted into 10 mM NaOH solution, which acted as the HF-LPME donor solution, and simultaneously the neutralized drugs were transferred across the HF and preconcentrated in 10 mM HCl acceptor solution inside the HF lumen. Mixture (1:1, v:v) of 1-ethyl-2-nitrobenzene and dihexyl ether was used for impregnation of the porous HF membrane. Analytes in the acceptor solution were injected directly from the HF into capillary electrophoresis instrument for further separation, detection and quantification. Repeatability of the hyphenated analytical system ranged from 9.8 to 13.8 % (RSD) and enrichment factors of 52 – 78 were obtained for model basic drugs spiked to DBS extracts at a concentration level of 0.1 mg/L.
Study of the authenticity of chocolate with different content of cocoa solids

This master's thesis deals with authenticity of chocolate with different content of cocoa solids. The theoretical part describes the production of chocolate products and the issue of food authenticity. Experimental part of this thesis is focused on the determination of total cocoa solids in the selected samples. Different samples of chocolate containing 50–90 % of cocoa were grated to fine particles, that were subjected to extraction without previous chemical treatment. The methylxanthines theobromine and caffeine were detected by HPLC to determine the content of fat–free cocoa solids. Fat–free cocoa solids content were found in the range 19,7–43,1 g/100 g of dry matter and the content of total fat was determined by the gravimetric method and was found in the range 30,9–55,4 g/100 g. The results of this thesis prove that, the producers meet the legislative requirements and the declared values are corectly declared on the wrappers of the product.
Application of polymer membranes in microextraction techniques for analysis of biological samples

The thesis presents experimental study on application of various polypropylene membranes (with different thicknesses, porosity and pore size) for direct coupling of membrane microextractions with capillary electrophoresis. No comprehensive study, which describes effect of these membrane parameters on extraction recoveries, was published in the past. Previous scientific works prefer application of 100 µm polypropylene membranes for their easy handling and satisfactory extraction efficiencies. Experimental part includes examination of selected polypropylene membranes as supported liquid membranes in-line coupled to capillary electrophoresis for analysis of basic drugs from complex samples. Membranes with three thicknesses (25, 100 and 170 µm) were tested. The highest extraction recoveries were achieved for the 25µm thick polypropylene membrane. Various pH conditions of donor and acceptor operational solutions were examined for extractions from real complex matrices (urine, plasma). The optimal extraction conditions were 10 mM NaOH as donor phase and 10 mM HCl as acceptor phase. 25µm membranes offer higher extraction recoveries, reduced consumption of organic solvents for membrane impregnation, similar mechanical stability and similar clean-up ability compared to thicker polypropylene membranes.
Temporary dwelling of migrants-reception and residential center

The theme of diploma thesis is design of reception center and residential center in Brno city. These asylum facilities are first two steps to proceeding for granting international protection. Both of these facilities provide accomodation and other services, such as provide of meals, social and psychological services, legal help or clients can take part in wide range of free-time activites. Designed complex is located at Červený kopec in Brno-Štýřice and it consists of 6 building divided into three parts - reception center, residential center and central structures used by the both facilities, such as administration, kitchen and maintenance services.
