The importance of different sources and sequencing protocols in increasing the accuracy of NGS analysis in diagnostic applications

1 ABSTRACT Continued advances in next-generation sequencing (NGS) technology, such as capacity, speed, and reduced cost per sequenced base, revolutionize personalized medicine and bring genomics into routine clinical practice. Nevertheless, NGS is still under rapid development, and the variability of sequencing protocols and validation procedures is one of its current bottlenecks. This thesis aimed to study the influence of different sample sources and NGS protocols (NGS library construction-sequencing-data analysis) on the accuracy of NGS analysis in diagnostic applications. In the first study, performed during the COVID-19 pandemic outbreak, we developed NGS protocols suitable for a whole genome analysis of the SARS-CoV-2 virus. Subsequently, in the second study, we examined the suitability of human saliva-derived gDNA for genomic/genetic analysis of selected variant types compared to traditional blood-derived gDNAusing validated NGS protocol and statistical comparison of the obtained data. Whole genome analysis of the SARS-CoV-2 genome was performed using two captured- based approaches and one amplicon-based approach to study the quality and effectivity of the respective NGS protocols. Lifelong learning for intensive care nurses

The aim of the thesis was to describe the interest and approach to the lifelong education of nurses in the field of intensive care. We focused on the areas of education and on the forms and methods that are most often used for lifelong learning. Further education requires orientation of nurses in the area defined by legislation, professional education and motivation. Deepening of knowledge and skills leads to the improvement of the quality of care for critically ill patients who are in intensive care units. The theoretical part of the thesis defines the foundations for understanding the issue of lifelong education of nurses in the field of intensive care. The research part was implemented using the method of quantitative research, through a non-standardized questionnaire survey. The participants were general nurses, of different ages, levels of education, from different fields of intensive care, types of hospitals and regions of the Czech Republic. The research group consisted of 216 nurses. Several conclusions emerged from the research investigation. Nurses are interested in deepening their knowledge in technical skills, but are limited by time, family and finances. That is why they choose e-learning as a form of further education organized by the employer. We discovered that nurses do not feel sufficiently supported by the employer for further professional education. Employers cover the costs associated only with the specialized study, the so-called ARIP. It doesn´t cover the costs associated with the education in the form of a subsequent master's study. Specialized education ensures the nurse's professional competence. Lifelong education contributes to maintaining competencies. In our investigation, a statistical relationship was observed between orientation in this area and the highest level of education achieved. As the level of education increases, the orientation of nurses in this area increases. We did not demonstrate a relationship between specialized education, the so-called ARIP, length of experience and orientation in the field of lifelong learning. According to the knowledge gained in the theoretical part and analyses made in the practical part, the aim is to suggest the best possible method of price formation with regard to the company strategy. The practical part contains calculations of selected methods and structure of costs and revenues with regard to the expected volume of revenues. Finally, this thesis deals with the evaluation od the economic impacts on the restaurant.
Improvement of the sales strategy using the e-commerce tools

This Bachelor Thesis deals with the improvement of the sales strategy using e-commerce tools for the company Lipoelastic a.s. For this reason, the first part of this work is dedicated to currently available e-commerce tools, outlining their ob-jectives and subsequently monitoring their results. In the second part of the work, Lipoelastic is analysed in details, along with the e-commerce tools that are currently used by the company. After that, suggestions regarding the better usage of those tools, and new alternatives that can improve the company's situation are proposed. The newly suggested tools consist primarily of the creation of an e-shop fully supported by online advertisement. The level of investment for selected tools and expected costs for a period of two years is included as well.
Value for money jako nástroj hodnocení PPP projektů : mezinárodní srovnání a návrh úpravy metodiky pro hodnocení projektů v České republice /

This dissertation deals with the economic evaluation of Public Private Partnerships (PPP) projects. Generally, PPP projects are evaluated using the evaluation methodology, which is called Value for Money (VFM). The aim of this dissertation is to propose some changes in the Value for Money methodology, which is used in the Czech Republic. All proposed changes should lead in to more frequent PPP realization in the Czech republic, whereas there should be no reduced requirements regarding the economic efficiency of this kind of projects. The Value for Money demonstrates the difference between two ways of project realization. The first one is conventional public procurement project, which is based on the separation of financing and providing of design, construction, operational and maintenance phase of the project. The second way is the Public Private Partnership project, where one private subject, in principle the Special Purpose Vehicle (SPV), provides all the project phases mentioned above. That means the PPP projects are usually very large, expensive and long-running. Value for Money methodologies are used in many countries, where the PPP projects are prepared and realized. Of course, the methodologies differ based on the legislative, institutional and economic environment. In the Czech republic the VFM methodology was created and published in the 2008. But the utilization of the methodology is very marginal. That means there are very limited experiences, which allows changing the methodology without causing enormous costs for submitters or advisors. All the proposed changes are formulated based on provided research. The research consisted in the analysis and comparison of Value for Money methodologies in selected countries and in the determination of key Value for Money drivers in PPP projects in the Czech republic, which allows identifying the specifics and specialties of Czech PPP market. Using these outputs some changes were formulated. The most important and fundamental changes are the change of the structure of the evaluation methodology, the change in the qualitative evaluation questions formulation, the change in the quantitative PSC methodology and the changes in the legislative environment.

This diploma thesis deals with the topic of personnel management at an airline. The thesis discusses personnel management generally. The end of the literature is focused on the differences in the personnel management of the flight department at an airline. Quanti-tative research among employees of a selected airline on the topic of recruitment methods was conducted and subsequently documented. Further, a new innovative procedure was created for the recruitment of new employees to the flight department. This proposal was additionally economically quantified by comparison with the costs of a traditional tender.
Vliv různého zpracování půdy na výnosy a ekonomiku pěstování jarního ječmene

The aim of this thesis was to elaborate analysis of cultivation of cereal crops and spring barley in the Czech Republic, EU and worldwide, and to evaluate the impact of different methods of tillage on yields and economics of cultivation of spring barley. The effect of different soil tillage intensity on yields and economics of cultivation of spring barley was observed in the long -- term experiment. This experiment was conducted on loamy chernozem soil in sugar-beet producing region in Ivanovice na Hané. Yields obtained between years 1990 -- 2014 were evaluated. Spring barley was grown in three crop rotations. The forecrop of the spring barley was always sugar beet grown after maize for silage, winter wheat and spring barley. Four variants of tillage were evaluated: ploughing to the depth of 0,22 m, ploughing to the depth of 0,15 m, straight sowing into non-tilled soil and shallow loosening soil to the depth of 0,10 m. In all three crop rotations, the highest average yield was obtained in variant - ploughing to the depth of 0,15 m and the lowest average yield was obtained in variant - ploughing to the depth of 0,22 m. For the economic evaluation of the impact of different soil tillage intensities were determined variable costs, revenues and gross margins. In all three crop rotations the lowest gross margins were achieved in variant with ploughing to the depth of 0,22 m. In the crop rotation of maize for silage -- sugar beet -- spring barley and in the crop rotation of spring barley -- sugar beet -- spring barley were the highest gross margins determined in variant with direct sowing into non-tilled soil. In the crop station of winter wheat -- sugar beet -- spring barley were the highest gross margins determined in variant shallow loosening soil to the depth of 0,10 m. The results indicate that in these soil and climatic conditions, the reduction of intensity of tillage for spring barley grown after sugar beet is sustainable alternative to the traditional method of tillage.

Soukupová, A., Proposal strategies for Legito. Bachelor thesis. Brno: Mendel University, 2016. The aim of the bachelor thesis is to define the suggested strategies for start-up Legito. The work is divided into theoretical review and result part. The theoretical review is created on the base of studying literature. In the result part, there are analysed internal and external environment, which show the factors that are needed for creation a SWOT analysis. Based on the SWOT analysis there are de-fined recommended strategies. The company's management chose the user expe-rience testing as the main of them. This strategy was analysed in detail and it in-cludes a calculation of the costs.
Efektivita vybraných nástrojů content marketingu

Following diploma thesis is focused on content marketing and efficiency of its selected tools. Different types of templates suitable for making content in a form of online published articles were selected. Efficiency was analysed from different point of views - regardless of gender, with respect to gender and also in terms of cost effectiveness. Eye tracking research methods and in-depth interview were used. Final recommendations were devised to adjust marketing strategies based on evaluation of each article.

The thesis includes several tables that describe the technological equipment, calculation of the costs and a comprehensive analysis of these costs.. moreover, based on the analysis, there are specified the costs for producing one litre of wine.
