HPLC in quality control of food supplements based on goldenrod and containing rutin and chlorogenic acid

RNDr. Dalibor Šatínský, Ph.D. Title of the diploma thesis: HPLC in quality control of food supplements based on goldenrod and containing rutin and chlorogenic acid In this diploma thesis, a high-performance liquid chromatography (HPLC) method was developed and validated for the separation and determination of chlorogenic acid, 3,5- dicaffeoylquinic acid, rutin, quercetin, and kaempferol. This method was applied to the analysis of food supplements based on goldenrod: drops Celík zlatobýl (Valdemar Grešík), Celík obecný BIO (Bylinné kapky), Celík obecný (Bylinné kapky), Zlatobýl (Green Idea), Zlatobýl - tinktura z pupenů (Serafin), Zlatobýl - tinktura z bylin (Serafin), Zlatobýlová směs (Dědek kořenář), Zlatobýl (Inca Botanica), and capsules Celík (Ex Herbis). Furthermore, two food supplements containing rutin (Solgar and Walmark®) and a food supplement Zelená káva (Botanic) containing chlorogenic acid were analyzed. The specifics of nursing care of a patient with an inserted PICC - awareness of nurses

This bachelor's thesis discusses the treatment of PICC catheters. The theoretical part of the thesis provides comprehensive information about the PICC catheter and about the specifics of care for patients with an inserted PICC. The thesis mainly focuses on the treatment of this access under aseptic conditions in order to prevent catheter infection, where the correct procedure of the nurse is important. At the same time, the goal is for the provided information to agree with the current recommendations of national and international companies involved in creating educational resources to treat long-term vascular accesses. The aim of the empirical part of this bachelor's thesis is to obtain information about nurses' knowledge on treating PICC catheters through quantitative research. Data collection was conducted in a selected hospital using self-designed questionnaires. The questionnaire consists of 25 questions, of which 14 questions are used to verify knowledge of the given topic. 150 questionnaires were distributed and 109 questionnaires were returned for analysis. Based on the results of the questionnaire, a simple manual with pictograms was created and is available at the end of the thesis. Distribution of aerosol particles from ship diesel engines

3 Abstract Measurements of aerosol particle size distributions in the range 5 nm - 4 µm at 1.5 m above sea level were carried out on board a boat in German waters of the Baltic Sea. The measurements were aimed at characterizing aerosol particles from marine aerosol and from ship diesel engines and their dynamics, i.e. the aging of the exhaust plume in the marine boundary layer. The marine aerosol of PNC5-340 1335-1939 cm-3 is characterized by Aitken, accumulation and coarse modes of CMD 20 and 41-55 nm and 123-159 nm and 1.6 µm, respectively. The ratio of the accumulation to the Aitken mode increases with increasing shipping density. Aerosol particles from diesel engines in exhaust plumes with an age of 2 - 60 s and at a distance of 150 - 600 m from the surveyed ships are characterized by PNC5-340 1-4 -105 cm-3 and CMD 20 nm. The modal size distribution of supermicron particles of Da 1 - 4 µm does not change in the exhaust plume, the total mass of supermicron particles increases only by 1 - 6% compared to the background. The dynamics of the size distributions of aerosol particle numbers is mainly due to the formation, i.e. expansion and dilution, of the plume. Modeling tasks, consisting in the transformation of a mental image into the formal representation, very often occur in the study of computer science, mathematics, but also in other technical and non-technical fields. From the didactic point of view, it is interesting in modeling tasks (among others), which methods/procedures solvers (students) use to manage more demanding complex tasks. It is often possible to encounter students, who have significant problems solving complex tasks (or they are not able to solve them satisfactorily at all), even though they did not have any problems with similar simpler tasks before during the course. A possible explanation can be seen in the suboptimal use of modeling strategies (e.g. divide&conquer, iterative approach, etc.) or their total absence. This research is focused on the analysis of the role of strategies in modeling tasks, the possibilities of their support/development and teaching methods. The study is conducted, which aimed to analyze the possible impact of strategies in the procedures of university students in solving modeling tasks in the areas of 3D modeling and databases. The goal of the research was also to analyze the possibility of generalization of modeling strategies and their transferability between domains with different type of model.
Thrombin generation in pathophysiological conditions

Introduction: Thrombophilic conditions may be accompanied by increased thrombin generation (TG), especially during pregnancy when the increase in thrombin generation results from a series of changes in the haemostatic system. During administration of anticoagulant treatment, it is desirable to reduce the increase in thrombin generation. Aims of the study: The first aim of our study was to compare TG in individuals with a proven factor V Leiden mutation and individuals with elevated levels of factor VIII. Another aim was to compare patients with VTE and also with or without proven FV Leiden mutation that are taking different anticoagulation treatments. The final aim was to assess thrombin generation in pregnant women and their response to LMWH administration during severe thrombophilic conditions. Group of patients and methods: There were included 170 healthy blood donors in the control group, the congenital and acquired thrombophilia group included 44 individuals with FV Leiden mutation and 38 individuals with factor VIII level >150 % without other proven thrombophilia. 347 subjects with VTE taking direct coagulation inhibitors were included in the anticoagulation group. Production of a part of the TV bracket

Master’s thesis is focused on the design of the production of a part of the TV bracket. Part is made of steel sheet with thickness of 2,5 mm. The structural steel S 235 JRG1 was chosen as the most suitable material with following powder coating. From the possible variants, the production by progressive forming tool was chosen with respect to production requirements and a series size of 25 000 pieces by year. Part will be made in seven steps and it will require total force of 2 500 kN. Production will be realized on a mechanical eccentric press KSTU 3150-25-F3-P-SE from ANDRITZ Metals with maximum force of 3 150 kN.
Development of ceramic filter equipment for electro-insulating liquids

Cílem této diplomové práce byla příprava makroporézních keramických filtrů na bázi mullitu a oxidu křemičitého, které by byly vhodné pro filtraci elektroizolačních kapalin v distribučních transformátorech. První část této práce tvoří literární rešerše, která se zabývá tématem elektroizolačních kapalin a jejich degradace, metodami přípravy makroporézní keramiky a jejího využití pro filtraci, včetně přehledu používaných materiálů. Experimentální část se zabývá přípravou porézních filtrů pomocí dvou vybraných metod, replikační metody a přímého pěnění. U filtrů připravených pomocí optimalizovaných parametrů bylo vyhodnoceno fázové složení, mikrostruktura a pevnost v tlaku. Filtry připravené replikační metodou byly tvořeny otevřenými propojenými póry o velikosti 150 až 350 µm, dosahovaly porozity kolem 80 % a pevnosti v tlaku až 2,4 MPa. Keramické pěny získané přímým pěněním dosahovaly porozity 68-73 % a otevřených propojených pórů menších než 100 µm. Filtry bylo nutné zvětšit na rozměry dané filtračním zařízením. Přímé pěnění se ukázalo jako nevhodné pro přípravu filtrů ve větším měřítku, nicméně filtry připravené replikační metodou byly úspěšně zvětšeny a jsou připraveny pro testování jejich filtrační schopnosti. Tyto filtry s otevřenými propojenými póry, unimodálním rozložením velikosti pórů a dostatečnou mechanickou pevností jsou potenciálními kandidáty pro použití v prvním kroku filtrace elektroizolačních kapalin a mohou tak představovat udržitelnější alternativu k v současnosti používaným papírovým filtrům.
The production of a component of a lever lock

The master‘s thesis deals with the suggestion of the production of a component of a lever closure, which is used as a quick-clamping mechanism. The part is made of low-carbon steel DC01 in the form of a sheet with a thickness of 1.5 mm and an annual production of 200,000 pcs. The individual production methods were chosen using a progressive combined tool that combines cutting and bending. After evaluating the technology, determining the developed shape of the part and choosing its location on the line was chosen a semi-finished product in the form of metal coils sheet. After determining the production procedure, which consists of four steps, technological, construction and control calculations were carried out on the basis of which the tool was constructed. An eccentric press LEN 63 C was chosen as a suitable machine for production. A feed line marked ARPL RAL 150 was also chosen for production.
MODETA JIHLAVA

The topic of my Master‘s thesis is an architectural study of the reconstruction and revitalization of the former MODETA factory building, located near the Jihlava Zoo. The building is designed to have multifunctional use for the zoo. The construction program defines the building as a complementary facility to the functional complex of the Jihlava Zoo. The location is near the historical center of the city of Jihlava, along the Jihlávka river. The project was preceded by a pre-diploma project, in which I focused on the design of mobile containers for the Jihlava Zoo. In the building, there will be an animal exhibition connected to the zoo. The new section will be fully functional even in the winter season. You will find here the Water World with an exhibit of a small crocodile - the Dwarf Caiman. There will also be an Orangutan exhibit and a Night World, where mammals will have an opposite regime (night during the day). The exhibit of small monkeys is located on the 2nd floor, where there is also a 2nd observation point for orangutans. You can also observe them from a bridge that connects the building to the zoo. For zoo staff, there is a facility with offices, a workshop, and sanitary facilities. A self-service restaurant with a capacity of up to 150 people on the 1st floor serves both the public and employees, whose facilities are connected to the restaurant. The public will also have access to a congress and exhibition hall, where it is possible to create two separate events or combine spaces by sliding mobile partitions to increase capacity. The halls are connected by elevators and stairs to accommodation on the 3rd floor and the restaurant for refreshments on the 1st floor. Accommodation can be used by the public as well as educational institutions. The IQ Park is located on the 2nd floor, accessible by a separate entrance or directly from the Night World. There will be interactive equipment with animal themes and a screening room with a capacity of 60 people.
MODETA JIHLAVA

The work focuses on the revitalization of the former Modeta textile factory complex in Jihlava. It addresses its integration into the structure of the adjacent zoo area. The aim of the project is to create a multifunctional facility that will provide new spaces for zoo activities. The building houses exhibitions of Indian rhinoceroses, small mammals, and reptiles. Additionally, the building includes a restaurant with a capacity of up to 150 people and accommodation options. Like the congress hall, these spaces are accessible from the entrance foyer. Above the hall, there is an IQ park and coworking spaces. New changing rooms for zoo employees are designed in connection with the technical yard.
