Comparison of different ways of breeding, nutrition and controlled reproduction of Lota lota

Based on the results reached after 216 days of experimentation with various breeding types, it can be said that the type of feed is not of paramount importance. This fact is based on the previous adaptation of submitted food, this is especially important for artificial feed. From our point of view, the more important factor is temperature, intensity and method of breeding. Prey selections choices involving 11 species of fish were performed on age groups of different burbot. All groups of burbot preferred the following species: grass carp, pikeperch, tench and stone moroko. The non-preferred species was ruffe. And different preferences were set for common carp, silver carp, burbot, sunfish and also for roach. The study evaluated reproductive parameters in burbot (Lota lota ) and focused on incubation time in two localities. Burbot spawning was observed from January to early February at an average temperature of 1.55 +- 0.9 °C in Borova Lada and 4.78 +- 0.11 °C in Mydlovary. For burbot at the age of 2+, at an average 1166 +- 100 were determined number of swollen eggs per gram. Relative fecundity was at 257260 +- 70678 eggs per kilogram of female. The absolute fecundity was at 24232 +- 6753 eggs.The last parameter evaluated was the incubation period. The average incubation period in the years 2020-2022 was 202 +- 24.55 °D in Mydlovary and 75.8 +- 16.67 °D in Borova Lada. In connection with artificial spawning, in this study, we further point out the possibility of peeling artificially spawn eggs with NaClO, in which we did not observe a negative effect.
Unruptured intracranial aneurysms : Does the risk of endovascular treatment outweigh natural history?

Summary: Background: The management of unruptured intracranial aneurysms (UIAs) remains controversial. The international study of UIAs (ISUIA) found low rates of rupture and appreciable treatment risks, which can be interpreted that many UIAs should not be treated. Results of endovascular coiling of 131 patients with UIAs are presented along with a risk-benefit analysis based on patient and aneurysm characteristics. Patients and methods: From December 1996 to September 2005, 131 patients (93 women and 38 men, mean age 51 years) with 151 UIAs were treated using detachable coil embolisation. Data on procedural complications, patient and aneurysm characteristics, clinical and radiological follow-up were entered into a prospectively collected database. A risk-benefit analysis was performed. Results: Endovascular treatment was successful in 145 (96%) aneurysms. One aneurysm ruptured intraoperatively with a fatal outcome. Ten thromboembolic events occurred, leaving one patient moderately disabled. The combined morbidity and mortality rate at 6 months per patient is 1,5% (95% confidence interval: 0,07 - 5,7%). Risk-benefit analysis comparing these data with published natural history suggests that treatment with low complication rates can be offered to most patients with UIAs. Expression and purification of protein photo-initiated nanoprobe: tool to study clinically relevant protein-protein interactions

Cytochrome b5 is a key protein in the function and regulation of the mixed function monooxygenase (MFO) system in mammalian endoplasmic reticulum and is, therefore, a clinically relevant target for biochemical studies. To study its interactions within the MFO system using photo-initiated crosslinking, we have developed cytochrome b5 mutants with methionine in several key amino acid positions within the primary amino acid sequence, such as serine 23 and leucine 41. Also, naturally presented Met in positions 96, 126 and 131 were mutated to Leu with no effect to cytochrome b5 activity. Our protein was expressed in E. coli B834 auxotrophic type with L-2-amino-5,5-azi-hexanoic acid (photo-Met) present in the cultivation medium. This methionine analogue with photolabile diazirine ring is readily incorporated in Met positions into the primary sequence of proteins by aminoacyl-tRNA synthetases. The whole expression protocol was optimized to achieve maximal percentage of photo-Met incorporation into the expressed protein sequence. Up to 93.4% incorporation of photo-Met was achieved. Awareness of pediatric patients and their parents before anesthesia

The primary aim of the thesis was to describe current awareness system of pediatric patients and their parents before anaesthesia and whether and to what extent the two groups are informed about anaesthesia. Furthermore, the thesis presents actual opinions of both responding groups on the current state of clinical practice and suggestions for its improvement. In accordance with available sources, so far no similar research has been carried out in the Czech Republic. A total number of 215 respondents at two biggest teaching hospitals in the Czech Republic have been addressed (Teaching Hospital Motol, Prague and Teaching Hospital Brno), the survey was finished by 131 people (60.93 %). Consequently, the questionnaire output was statistacally processed. The findings demonstrate a need of an improved anaesthesia awareness of pediatric patients and parents before the surgery. Obviously, both parents and children require printed information materials, although parents still tend to prefer rather the oral form - the anaesthesia interview.
