Design of special asphalt mixtures SAL for the repair of cement concrete pavements
Kalfeřt, Martin ; Špaček, Petr (referee) ; Varaus, Michal (advisor)
In this thesis layer with increased resistance to crack propagation (SAL) are designed. The theoretical part describes the SAL layer, further the input materials, their production and design of various mixtures. The following is a description of the tests, which include tests of permanent deformation, crack propagation, low-temperature characteristics, test, flexural strength and relaxation. After the test results and conclusion are stated.
Desing and evaluation of Bituminous Stress Absorbing Pavement Layer
Coufalík, Pavel ; Špaček, Petr (referee) ; Kudrna, Jan (advisor)
This master's thesis deals with the possibility of special asphalt-rubber mixture using - Stress Absorbing Layer (SAL). In the theoretical part the SAL and test methods are described. In the practical part, several different mixtures are designed and selected mixtures were tested. Low-temperature characteristics, permanent deformations, bending tensile relaxation, stiffness modulus and fatigue properties were determined. Based on the test results the mixtures were compared and their benefit is evaluated according to Performance Pavement Design Method, the pavement structure was evaluated by using of computer programme LayEps.
Assessment of surveying techniques for analyzing the condition of embankment dams: a case study of R3 Hájecká dam.
Pechar, Jiří ; Jerman, Jan (advisor) ; Špaček, Pavel (referee)
The diploma thesis deals with the evaluation of methods for surveying the construction of dams and for identifying faults in their body. In the beginning of the work, the issue of bulk dams is described with an emphasis on the material composition and structural elements in their bodies. The work also describes the types of dam failures, their characteristics, causes and statistics of their frequency of occurrence. Finally, the text of the thesis describes the monitoring of bulk dams. The practical part is dedicated to the evaluation of the applied engineering geological, hydrogeological and geophysical methods and their suitability for determining the condition of bulk dams. Based on the results, the benefits and limitations of individual methods and the possibility of their combined use to achieve more accurate results are described. For the evaluation of the above methods, the R3 Hájecká earth fill dam was selected, on which water began to seep through the left bank side of the dam body in the course of 2020. Of the safety ones, the water level in the reservoirs was lowered before the field work, which reflects the choice of survey methods carried out.
Návrh internetové marketingové komunikace pro firmu Včelařství Špaček
Špaček, Petr
Špaček, P. Proposal of internet marketing communication for the company Včelařství Špaček. Bachelor thesis. Brno. Mendel University in Brno, 2022 Bachelor thesis deals with internet marketing communication. It is divided into a theoretical part and the actual work. The theoretical part describes concepts such as marketing communication, characterizes tools for communication and presents analyses for internal and external environments. In the second part, the methodology of the thesis is described, describing the procedure for the actual thesis and the research itself. The research consists of a questionnaire survey. Based on the results of the questionnaires and analyses, a new internet marketing communication for the company Včelařství Špaček was designed.
Detection of epithelial-mesenchymal transition markers (EMT) in cells in vitro
Špaček, Petr ; Szotáková, Barbora (advisor) ; Trejtnar, František (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Petr Špaček Supervisor: Doc. Ing. Barbora Szotáková, Ph.D. Consultant: RNDr. Věra Králová, Ph.D. Title of diploma thesis: Detection of epithelial-mesenchymal transition (EMT) markers in cells in vitro Epithelial-mesenchymal transition (EMT) is a process during which motile mesenchymal-like cells develop from non-motile parent epithelial cells. Physiologically, EMT plays important roles during embryonic development and wound healing. Loss of control over this mechanism can lead to fibrosis and cancer progression. Motile mesenchymal-like cells can pass through the basal lamina, get into the blood vessels and spread to distant tissues. Transition is regulated by EMT biomarkers. The biomarkers comprise wide spectrum of proteins, including cell surface proteins (E-cadherin, N-cadherin), cytoskeletal proteins (vimentin), microRNA (miR 200) and transcription factors (Snail, Twist). In this study, expression of EMT biomarkers was evaluated using RT-PCR and Western blotting. The ability to migrate was assessed using real-time analysis with the x-CELLigence system. Two known triggers of EMT, the StemXVivo™ EMT Inducing Media Supplement (IS) and TGF-β, were compared in human oral cancer cell lines DOK and...

Frost susceptibility of improved soils to pavement subgrade
Lorenc, Daniel ; Špaček, Petr (referee) ; Stehlík, Dušan (advisor)
This diploma thesis deals with the issue of the effect of freezing on soils in subgrades of road structures. The theoretical part compares methods of determining the frost susceptibility of soils used in selected countries of the European Union and the Czech Republic. It also explains the principle of frost heave in subgrade layers. The aim of the practical part is to compare the direct and indirect method of determining frost susceptibility in the Czech Republic and the California bearing ration CBR or the immediate bearing ratio IBI.
