The role of parvalbumin interneurons in seizure initiation, propagation and termination in a mouse model of focal cortical dysplasia of type II

Focal cortical dysplasia is a structural and functional malformation of cortical development caused by mutations in mTOR signalling cascade that can result in pharmacoresistant epilepsy. Parvalbumin interneurons are probably the most important inhibition force in the brain that can help to stop epileptic seizures and it is possible that there are changes in function of these interneurons that are dependent on focal cortical dysplasia. This thesis had the aim to create a murine model of focal cortical dysplasia type II with labeled parvalbumin interneurons. This model was created by injecting plasmid with mutated mTOR gene during in utero electroporation. Cranial window implantation was then performed over created lesion together with calcium indicator injection for measuring neuronal activity under two-photon microscope. Our results confirm the same morphological characteristics as other studies has shown and also occurence of spontaneous seizures. We have shown statistically significant differences in size of pyramidal neurons depending on lesional or non-lesional position. For parvalbumin interneurons there was no such difference. Causes of epilepsy due to disorders of cortical development of the brain

Malformation of cortical development are an important cause of childhood epilepsy. It is estimated that up to 40% of these cases are medication-resistant. Pathophysiological mechanisms involved in the development of epilepsy are unknown. Identification of new type of genes associated with MCD leads to development of new experimental models recapilute human clinical symptoms. Aim of this study is to summarize information on current progress in this area obtained from MCD experimental models. These informations will help us better understand epileptogenesis. The use of stem cells in the treatment of Amyotrophic lateral sclerosis

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease caused by degradation of motoneurons (MN). No effective treatment is currently available. Cellular therapy is considered to be a promising experimental treatment that could target the pathology of complex disease through many potential mechanisms. We compared the effect of three types of repeated applications of human mesenchymal stem cells (hMSC): intrathecal administrations, intramuscular administrations and the combination of these applications. Best results were obtained after combined repeated hMSC administrations. We observed the rescue of MN, neuromuscular junctions and decreasing levels of proteins involved in the signaling of necroptosis (Rip1, cl-casp 8), apoptosis (cl-casp 9) and autophagy (beclin 1), decreasing astrogliosis and the level of astrocytic connexin 43. Neural precursors derived from induced pluripotent stem cells (NP-iPS) are considered as other promising candidates for ALS therapy. Intraspinal administration of NP-iPS increased mRNA expression of BDNF and IGF-1, on the other hand decreased expression of proapoptotic casp 3. Using Rating in Regulation Practice

This master thesis deals with the issue of using rating in the context of determining capital requirements. The focus is on using the Standardised Approach and the Internal Rating-Based Approach to calculate credit risk capital requirements. Different approaches to managing the risk and evaluating the capital adequacy. Criticism of rating agencies as well as the complexity of Internal Rating-Based Approach is taken into account. Furthermore, the thesis discusses the issue of responsibility of external rating agencies for their evaluations, and together with the issue of lacking competition. Various alternatives to external rating are discussed. The internal rating process is discussed in more detail along with the factors which affect rating evaluations of exposures to various entities. Subsequently, the thesis focuses on determining the rate of provision for impairment expected losses on receivables, and a possible impact of accepting IFRS 9 on capital adequacy.
