Using of cannabis extracts for the acne preparation of cosmetic products

Hemp belongs among important plants with therapeutic effects thanks to its high content of substances. This bachelor thesis brings information about the use of hemp in acne preparations. The theoretical part of this thesis focuses on the characterization of hemp and its active substances. The next chapters deal with acne and Propionibacterium acnes, which is involved in the course. For the experimental part, the extracts of two hemp varieties (Ferimon and Carmagnola) were characterized in terms of antioxidant and cannabinoid presence. The cosmetic preparations with hemp extract content were prepared and commercial products were purchased too. In both the extracts and the cosmetic preparations, their inhibitory effect against Propionibacterium acnes was observed. Micrococcus luteus, Candida glabrata and Escherichia coli were also tested. The results show that dried hemp flower extracts contain the largest amount of active substances. All hemp extracts had effect against gram-positive bacteria, in cosmetic preparations it was minimal effect or no effect.
Probiotics and prebiotics in food and other products

Theoretical part of this thesis focuses on present state and research of probiotics and prebiotics, their use for microflora modulation of host and their beneficial effects on the health of individuals. Furthermore thesis deals with efficiency of probiotics strains, which depends on the food matrix and other various factors. The experimental part focuses on the identification of chosen bacterial strain, which is contained in probiotics product. It is realized due the isolation of bacterial DNA by phenol extraction and by use of magnetic particles and subsequent analysis of obtained DNA by polymerase chain reaction (PCR).
Comparison of biotechnological procedures for pure proteins preparation

The production of recombinant proteins is a biotechnological process during which proteins are produced in foreign organisms by gen manipulation. To form a recombinant plasmid the gene encoding the desired protein is isolated and inserted into an expression vector. The plasmid is then transformed using physical or chemical method into a suitable host, where the recombinant gene is translated into amino acid sequence in the newly synthetized protein. The theoretical part of this bachelor thesis includes characteristics of proteins, methods of recombinant protein preparation and compares individual expression systems. Three isoforms of the p53 protein, which were synthesized in the E. coli microorganism, were selected for processing the experimental part. The transformed recombinant plasmid contained two tags for purification, HIS-tag and GST-tag, making it possible to compare the efficacy of the two purification methods. HIS-tag purification was found to work for all three isoforms better, with concentrations of recombinant proteins were several times higher than those of the GST-tag. The p53 proteins are about 50 kDa long, what was confirmed by sodium dodecyl sulfate polyacrylamide gel electrophoresis.
Determination of selected red fruit species in plant-based food using multiplex PCR and instrumental methods

In many cases food is becoming the subject of adulteration, including fruit products that contain raspberries, strawberries and blueberries. This bachelor thesis is focused on the detection of strawberries, rapsberries and blueberries in model and commercial baby food products using multiplex PCR and HLPC. The theoretical part of this thesis is focused on composition of red fruits, their importance in human nutrition and especially about isolation of DNA from plant material. The aim of the experimental part of work was the analysis of selected commercial and model mixtures fruit purees containg raspeberries, strawberries and blueberries using instrumental and molecular biological methods. The results of these metods are compared. Commercial purees were bought in retail grocery shop. Model mixtures of these purees were prepared in our laboratory. DNA was isolated from fruit purees after and it’s amplifiability was comfirmed, it was successfully used in multiplex PCR to confirm the presence of raspberries, strawberries and blueberries in fruit purees. In the instrumental part, certain phenolic substences which are specific to red fruits were detected by HPLC in model and commercial mixtures.
Detection of selected fruit species in plant-based foodstuffs through instrumental analysis and methods of molecular biology

Food is often subject to adulteration, including fruit products containing strawberries. This work deals with the detection of strawberries in model and real fruit products, so that possible falsification can be detected. The aim of the work was the preparation of model and real fruit products, their analysis using selected instrumental and molecular-biological methods and mutual comparison of the results of these methods. Model mixtures of fruit products were prepared and commercial ones were purchased. The content of polyphenols and flavonoids was determined by UV-VIS spectrophotometry, which were characterized in more detail by HPLC. DNA was isolated from the fruit purees, suitable primers confirmed the amplifiability and formation of a specific product for strawberry, and this product was verified by electrophoretic control. In the instrumental part, certain substances were detected by extraction from model and commercial mixtures and the HPLC method, and the amount of selected substances was determined. The lowest amount of strawberry DNA that could be detected by selected primers in the PCR method was 0.7 ng /µl, in commercial mixtures this DNA could not be detected. In the HPLC method, the strawberry was detected in all samples by focusing on a particular fingerprint and the amount of pelargonidin-3-glucoside in the samples. The instrumental method can be considered more suitable in this case.
Identification of probiotic bacteria in cosmetic creme using PCR technique

Probiotic bacteria are found in a wide range of food products and their internal use brings many positive effects for the human body. Thanks to these benefits, probiotic bacteria began to be used in the cosmetic industry with the assumption of similar positive effects as with internal administration. The aim of this bachelor's thesis was the theoretical part, which was devoted to various genera of probiotics, which are most often used in cosmetic products, in which skin diseases, surface probiotics are most often used and what effect they bring in treatment. Subsequently, the most commonly used molecular-biological methods for the identification of probiotic bacteria were described. In the experimental part, the DNA of probiotic bacteria declared by the manufacturer was isolated from two cosmetic creams and subsequently their presence was verified using the polymerase chain reaction method.
Identification of DNA from plant foods using molecular techniques

The aim of this bachelor thesis is the identification of plant DNA in a complex matrix using molecular techniques. The work first focuses on a search of literature sources concerned with molecular techniques of DNA identification. Next, it deals theoretically with methods of isolation and characterization of plant DNA. Specifically, we shall work with blueberry DNA. In the experimental part, DNA from four commercial products was isolated and subsequently subjected to PCR and HRM analysis.
Identification of probiotic bacteria with the use of PCR from plant frementation diary products

Probiotics are encountered daily, either by targeted application of dietary supplements containing live cultures of lactic acid bacteria or by consuming traditional fermented products including dairy or non-dairy products. These cultures can significantly affect intestinal microbiota, particularly during digestive discomfort or problems associated with diarrheal diseases or infectious gastritis. This work focuses on the isolation of probiotics from fermented plant-based products that are also suitable for vegetarians and vegans, whose numbers are currently increasing. In addition to traditional lacto-fermented foodstuffs, there is another alternative in the form of Kombucha. Probiotic bacteria isolated from real samples were subjected to polymerase chain reaction (PCR) to identify them. Presence of the Bacteria domain was detected, and with more specific amplification, the presence of the genus Lactobacillus was detected too, using the specific primers. The amplification resulted in individual fragments were visualized by agarose gel electrophoresis.
Uses of active compounds from honey in cosmetic products

The bachelor thesis was focused on the preparation and characterization of extracts and liposomes with active substances from chosen types of honey with the aim to apply these extracts to the cosmetic products. The extraction of active substances from chosen material was optimized during the work. As the most suitable organic solvent for extraction was chosen diethyl ether. All the extracts were tested on the content of total polyphenols, sugars and antioxidation activity. The final samples were also tested on the content of flavonoids. Extracts were encapsulated to the liposomes, which were tested especially for their antioxidation activity, size and stability. Then, extracts and liposome particles were applied to the cosmetic products. Cosmetic creams and gels were prepared and tested for antioxidation activity and antimicrobial activity against the strains Serattia marcescens and Micrococcus luteus. Antimicrobial activity of gels was proved especially against the strain of grampositive bacterias – Micrococcus luteus. From this point of view another one testing strain was chosen – Propionibacterium acnes. Finally, the chosen liposomes were tested for their contact with human cells, immortalized keratinocytes HaCat. No cytotoxic effect was observed and tested materials can be applied to cosmetics against acne.
Proteomic approach for the study of cancer cell line profiles.

Triple-negativní karcinom prsu (TNBC), velice agresivní podtyp rakoviny prsu, je známý svou nepříznivou prognózou a omezenými možnostmi léčby. V tuto chvíli je chemoterapie považována za hlavní způsob léčby. Za účelem vyvinutí nových účinných léčiv je snaha pochopit molekulární základ této nemoci. V důsledku toho bylo již několik potenciálně aktivních látek pro tento konkrétní typ rakoviny prsu podrobeno výzkumu. V poslední době se mnoho studií zabývající touto záležitostí provádí za použití proteomiky jakožto prostředku ke studiu proteomů rakovinných buněk. Rakovinové buňky obsahují klíčové rozdíly v proteinech, které regulují mechanismy buňky. Mapování těchto mechanismů může nakonec umožnit diagnostikovat stav organismu. Tato práce se zaměřuje na proteomické studium buněk TNBC a porovnává neošetřené buňky s buňkami, které byly podrobeny léčbě retinoidy. Separace proteinů a peptidů byla úspěšně provedena elektroforézou na 1D a 2D gelu. Kromě toho byly vzorky podrobeny enzymatickému štěpení vybraných proteinů, které byly poté identifikovány pomocí hmotnostní spektrometrie MALDI-TOF (MS). Proteiny, které se podílejí na procesu epiteliálně-mezenchymálního přechodu (EMT), byly poté kvantifikovány a porovnány mezi vzorky.
